Maternal exposure to high temperature in fish induces intergenerational emotional and cognitive disorders, associated with dysregulation of neurodevelopment genes in eggs and progeny

Project description:Female fish are known to be sensitive to temperature during reproduction, but the long-term consequences on offspring adaptive behaviour and their underlying intergenerational mechanisms remain unknown. We studied the intergenerational consequences of female rainbow trout (Oncorhynchus mykiss) exposure to high (17°C) or normal temperature (12°C) on offspring behavioural phenotypes. We also analysed genome-wide gene expression in eggs and embryos to elucidate the mechanisms by which thermal maternal exposure impacts offspring behaviour. Here we show that a thermal maternal stress induces emotional and cognitive disorders in offspring. Fear responses to a novel environment were inhibited in 17°C offspring indicating global emotional blunting. Thermal stress in mothers also decreased spatial learning abilities in progeny. Behavioural phenotypes were associated with the dysregulation of several genes known to play major roles in neurodevelopment. This is especially true for auts2, a key gene for neurodevelopment in fish and mammals, more specifically neuronal migration and neurite extension, and critical for the acquisition of neurocognitive function in fish and mammals. In addition to auts2, our analysis revealed the dysregulation of another neurodevelopment gene (dpysl5) as well as genes associated with human cognitive disorders (arv1, plp2). Our study also revealed major differences in maternal mRNA abundance in the eggs following maternal exposure to high temperature indicating that some of the observed intergenerational effects are mediated by maternally-inherited mRNAs accumulated in the egg. Together, our observations shed new light on the intergenerational determinism of fish behaviour and associated underlying mechanisms. They also stress the importance of maternal history on fish adaptive capacities in a context of global climate changes.

Music compensates for altered gene expression in age-related cognitive disorders

Project description:Extensive literature has explored the beneficial effects of music in age-related cognitive disorders (ACD), but limited knowledge exists regarding its impact on gene expression. We analyzed transcriptomes of ACD patients and healthy controls, pre-post a music session (n=60), and main genes/pathways were compared to those dysregulated in mild cognitive impairment (MCI) and Alzheimer’s disease (AD) as revealed by a multi-cohort study (n=1269 MCI/AD and controls). Music was associated with 2.3 times more whole-genome gene expression, particularly on neurodegeneration-related genes, in ACD than controls. Co-expressed gene-modules and pathways analysis demonstrated that music impacted autophagy, vesicle and endosome organization, biological processes commonly dysregulated in MCI/AD. Notably, the data indicated a strong negative correlation between musically-modified genes/pathways in ACD and those dysregulated in MCI/AD. These findings highlight the compensatory effect of music on genes/biological processes affected in MCI/AD, providing insights into the molecular mechanisms underlying the benefits of music on these disorders.

Transcriptional dysregulation of synaptic genes in a human iPSC model of major mental disorders

Project description:Schizophrenia and other psychiatric disorders are postulated to be developmental disorders resulting from synapse dysfunction. How susceptibility genes for major mental disorders could lead to synaptic deficits in humans is not well-understood. Here we generated induced pluripotent stem cells (iPSCs) from four members of a family in which a frame-shift mutation of Disrupted-in-schizophrenia-1 (DISC1) co-segregated with psychiatric disorders and further produced different isogenic iPSC lines via genetic editing. We showed that mutant DISC1 causes synaptic vesicle release deficits in iPSC-derived forebrain neurons. Mechanistically, mutant DISC1 dysregulates the expression of many genes related to synapses and psychiatric disorders and depletes wild-type DISC1 and the NCoR1 transcription co-repressor complex. Furthermore, mechanism-guided pharmacological inhibition of phosphodiesterases rescues synaptic defects in mutant neurons. Our study uncovers a novel gain-of-function mechanism through which the psychiatric disorder-relevant mutation affects synaptic functions via transcriptional dysregulation and provides insight into the molecular and synaptic etiopathology of psychiatric disorders.
