Project description:The red imported fire ant Solenopsis invicta is an invasive pest in the USA, eastern Asia and Australia that causes billions worth of damage where it has been introduced. In the insect colony, workers perform tasks based on their age as well as the subcaste, where the younger workers as well as the members of the smaller subcastes of workers tend to remain in the nest and tend to the brood while the older ants as well as the members of the larger subcastes of workers leave the nest to perform activities such as foraging. In eusocial insects, juvenile hormone has been identified to be a catalyst for behavioral changes among the worker caste, but the involvement of this hormone in S. invicta task allocation has not been investigated. Here, we conducted RNAseq analysis to identify genes associated with worker division of labor by comparing the expression profiles of foragers and nurses from the medium subcaste. Additionally, we evaluated the changes in the transcriptome of nurses in response to the application of the juvenile hormone analog, S-hydroprene. We found 816 differentially expressed genes between foragers and nurses and 100 differentially expressed genes between nurses treated with acetone and nurses treated with S-hydroprene. Here, we focused on the differentially expressed genes between foragers and nurses that were associated with different molecular pathways such as energy metabolism, glycolysis, juvenile hormone synthesis and metabolism, and immunity. Our results showed that the juvenile hormone analog induced changes in worker gene expression, some of which were as expected if the juvenile hormone is involved in regulating the shift from nursing to foraging in S. invicta workers. This analysis and data from this experiment can provide a slew of potential target genes to be used for control of this invasive pest.

Project description:Purpose: We aimed to provide a basis for the adaptive mechanism and a rich resource for the discovery and identification of novel genes involved in the cold and heat stress response in Solenopsis invicta. retinal transcriptome profiling (RNA-seq) to microarray and quantitative reverse transcription polymerase chain reaction (qRT–PCR) methods and to evaluate protocols for optimal high-throughput data analysis Methods: Solenopsis invicta was reared at lab condition, and incubated at 4 different temperature for 24h (10, 20, 30 and 40℃). RNA was extracted using Trizol reagent and Illumina sequencing was performed at Macrogen. Illumina short reads were quality-filtered and Illumina-based de novo transcriptome assembly was performed. Differential Gene Expression Analysis was studied for different tempearture conditions. Results: Totally 99,085 unigenes was obtained which at least 19,154 were annotated with gene descriptions, gene ontology terms, and metabolic pathways. 86 GO functional sub-groups and 23 EggNOG terms resulted. DEGs with log2FC≥10 were screened and were compared at different temperatures. We found 203, 48 and 66 specific DEGs co-regulated at 10, 20 and 40℃, respectively compare with 30℃. Comparing transcriptome profiles for differential gene expression resulted various DE proteins and genes, including cytochrome P450, NADH dehydrogenase subunit 1, cuticle protein and HSP which have previously been reported to be involved in cold and high temperature resistance. GO analysis revealed that antioxidant activity up-regulated under high temperature stress. Conclusions: we compared the transcriptomes of S. invicta under high- and low-temperature stresses using RNA-Seq technology based on the high-throughput sequencing. Comparative transcriptome analysis identified many genes and a large number of changes were discovered in metabolic pathway through the GO and KEGG enrichment analysis. Our data will facilitate further molecular investigations and genomic research. Many high- and low-temperature significantly up-regulated genes were first identified in this study. These newly found genes may be important and necessary to RIFA overwintering and its behavior for adaptation in new environment as well as quarantine area.

Project description:Among social insects, task allocation within its group members remains as one of the paramount pillars of social functionality. Division of labor in many eusocial insects is maintained by behavioral flexibility that can shift according to the needs of the colony they reside in. Workers typically, over time as they age, shift from intranidal nurses to extranidal foragers. If the needs of the colony change, either from the needs of the adults or the brood therein, workers shift their behavior in order to compensate for the need of a particular task to be done. This shift, either accelerating towards a behavior associated with an older worker, or regressing back into the nest, is not clearly understood in social insects outside of honeybees. In this study, evaluated how brood type affected the red imported fire ant, Solenopsis invicta, worker task reversion and acceleration. Through observation of worker behaviors performed over multiple time-points per day, we discovered that worker task reversion and acceleration does occur within this ant species. Furthermore, the type of brood influenced the rate at which this occurred, with larvae having the strongest effect of all types. Finally, there was a propensity for workers to maintain their new behavior throughout the experiment. This study shows that the needs of brood within a social insect colony can influence the behavior workers perform, reversing the age polyethism that is common among social insect species.

Project description:Vitellogenin has been proposed to regulate division of labor and social organization in social insects. The red imported fire ant (Solenopsis invicta) harbors four distinct, adjacent vitellogenin genes (Vg1, Vg2, Vg3, and Vg4). Contrary to honey bees that have a single Vg ortholog as well as potentially fertile nurses, and to other ant species that lay trophic eggs, S. invicta workers completely lack ovaries or the ability to lay eggs. This provides a unique model to investigate whether Vg duplication in S. invicta was followed by subfunctionalization to acquire non-reproductive functions and whether Vg was co-opted to regulate behavior within the worker caste. To investigate these questions, we compared the expression patterns of S. invicta Vg genes among workers from different morphological subcastes or performing different tasks. RT-qPCRs revealed higher relative expression of Vg1 in major workers compared to both medium and minor workers, and of Vg2 in major workers when compared to minor workers. Relative expression of Vg1 was also higher in carbohydrate foragers when compared to nurses and protein foragers. By contrast, the level of expression of Vg2, Vg3, and Vg4 were not significantly different among the workers performing the specific tasks. Additionally, we analyzed the relationship between the expression of the Vg genes and S-hydroprene, a juvenile hormone analog. No changes in Vg expression were recorded in workers 12 h after application of the analog. Our results suggest that in S. invicta the Vg gene underwent subfunctionalization after duplication to new functions based on the expression bias observed in these data. This may suggest an alternative and still unknown function for Vg in the workers that needs to be investigated further.

Project description:We used whole-genome fire ant microarrays to examine the molecular basis for social organization in Solenopsis invicta. Monogyne (single queen) fire ant colonies were collected in the field and transported into the lab were they were reared in standard conditions for two weeks. At this point, each colony was split into two sub-colonies: one sub-colony contained the functional mother queen (queenright) while the other was left queenless. Each sub-colony included a nesting chamber, containing the brood and workers performing nursing tasks, and a foraging area, separated from the nesting chamber and provided with food and water sources. For both queenright and queenless sub-colonies, foraging workers were collected in the foraging area while non-foraging workers were collected in the nesting chamber. Total RNA was isolated from pools of 10 whole workers and processed for microarrays.

Project description:Susceptible individuals who have suffered painful stings caused by red imported fire ants, Solenopsis invicta, usually experience physical health effects such as fever, dizziness, generalized urticaria, or other systemic reactions such as anaphylactic shock. Whether S. invicta stings also have negative effects on mental health is not clear. In the present study, the psychological impact of S. invicta stings was evaluated using a questionnaire that included a previously published posttraumatic stress disorder (PTSD) checklist, the Patient Health Questionnaire 9-item (PHQ-9), the Generalized Anxiety Disorder 7-item (GAD-7) scale, the Beck Anxiety Inventory (BAI), and the Pittsburgh Sleep Quality Index, 5th scale (PSQI(5)). A total of 96 valid questionnaires were obtained; 37 participants were placed in the stung group, and 59 participants were placed in the unstung group. Our results showed that symptoms of anxiety, depression and sleep disturbances were not associated with S. invicta stings (for GAD-7 scale, Pearson Chi-Square test, χ2 = 0.152, df = 1, P = 0.697; for the BAI, χ2 = 2.252, df = 1, P = 0.133; for the PHQ-9, χ2 = 0.098, df = 1, P = 0.754; for the PSQI(5), χ2 = 0.536, df = 1, P = 0.453). In total, 2 of 83 individuals stung by S. invicta met the criteria (>50) for PTSD. However, there was no significant difference on PTSD between those stung by S. invicta in the 30-day group and the over 30-day group (χ2 = 0.318, df = 1, P = 0.573). Overall, our data do not show an effect of S. invicta stings on mental health as measured using a range of indicators.

Project description:The red imported fire ant (Solenopsis invicta) is one of the deadliest invasive ant species that threatens the world by disrupting biodiversity, important functions within a natural ecosystem, and community structure. They are responsible for huge economic losses in the infested countries every year. Synthetic insecticides, especially indoxacarb, have been broadly used to control S. invicta for many years. However, the biochemical response of S. invicta to indoxacarb remains largely undiscovered. Here, we used the sublethal doses of indoxacarb on the S. invicta collected from the eight different cities of Southern China. The alteration in the transcriptome profile of S. invicta following sublethal dosages of indoxacarb was characterized using high-throughput RNA-seq technology. We created 2 libraries, with 50.93 million and 47.44 million clean reads for indoxacarb treatment and control, respectively. A total of 2018 unigenes were regulated after insecticide treatment. Results indicated that a total of 158 differentially expressed genes (DEGs) were identified in the indoxacarb-treated group, of which 100 were significantly upregulated and 58 were downregulated, mostly belonging to the detoxification enzymes, such as AChE, CarE, and GSTs. Furthermore, results showed that most of these DEGs were found in several KEGG pathways, including steroid biosynthesis, other drug metabolizing enzymes, glycerolipid metabolism, chemical carcinogenesis, drug-metabolizing cytochrome P450, glutathione metabolism, glycerophospholipid metabolism, glycolysis/gluconeogenesis, and metabolism of xenobiotics. Together, these findings indicated that indoxacarb causes significant alteration in the transcriptome profile and signaling pathways of S. invicta, providing a foundation for further molecular inquiry.

Project description:Both exogenous and endogenous compounds can induce the expression of cytochrome P450 genes. The insect cytochrome P450 genes related to insecticide resistance are likely to be expressed as the "first line of defense" when challenged with insecticides. In this study, four cytochrome P450 genes, SinvCYP6B1, SinvCYP6A1, SinvCYP4C1, and SinvCYP4G15, were firstly isolated from workers of the red imported fire ant (Solenopsis invicta) through rapid amplification of cDNA ends (RACE) and sequenced. The fipronil induction profiles of the four cytochrome P450 genes and the two previously isolated CYP4AB1 and CYP4AB2 were characterized in workers. The results revealed that the expression of SinvCYP6B1, SinvCYP6A1, CYP4AB2, and SinvCYP4G15, increased 1.4-fold and 1.3-fold more than those of acetone control, respectively, after 24 h exposure to fipronil at concentrations of 0.25 μg mL-1 (median lethal dose) and 0.56 μg mL-1 (90% lethal dose), while no significant induction of the expression of CYP4AB1 and SinvCYP4C1 was detected. Among these genes, SinvCYP6B1 was the most significantly induced, and its maximum expression was 3.6-fold higher than that in acetone control. These results might suggest that multiple cytochrome P450 genes are co-up-regulated in workers of the fire ant through induction mechanism when challenged with fipronil. These findings indicated that cytochrome P450 genes play an important role in the detoxification of insecticides and provide a theoretical basis for the mechanisms of insecticide metabolism in the fire ant.
