Project description:In order to explore the molecular mechanism of SJMHE1 intervention in allergic rhinitis, an OVA-induced allergic rhinitis model was established and intervened by subcutaneous injection of PBS and SJMHE1. Project description:Allergen exposure was thought to play a critical role in the etiology of AR. And allergen avoidance, the practice of avoiding exposure to allergens, has been generally advised as the management of AR. However, the effect is uncertain and the underlying mechanism is far from known. We used gene expression microarrays to identify genes differentially regulated by allergen avoidance in allergic rhinitis mouse model. Affymetrix Mouse Gene 1.0 ST arrays were used to identify the expression profiling of nasal mucosa in three groups of mice: (1) mice sensitized and challenged with saline (control group); (2) mice sensitized and challenged with ovalbumin (OVA) and sacrificed 2 hours after the last challenge (OVA group); (3) mice sensitized and challenged with OVA and sacrificed 4 weeks after the last challenge (4w-after group).

The HDAC Inhibitor Sodium Butyrate Prevents Allergic Rhinitis and Alters lncRNA and mRNA Expression Profiles in the Nasal Mucosa of Mice

Project description:Our previous research confirmed that histone deacetylase inhibitor sodium butyrate (NaB), delivered intranasally, has exhibits therapeutic effects on a mouse model of allergic rhinitis (AR). However, whether it is effective on AR when administered orally and prophylactically, as well as its potential effects on gene expression, remained unknown. Methods: In this study, we investigated the preventive effect of NaB on AR when added to the diet of newly weaned mice and evaluated changes in lncRNA and mRNA expression profiles in the nasal mucosa. Mice were randomly divided into three groups as follows: C group (control group, no treatment), AR group (treated with ovalbumin [OVA]), and NaB+AR group (treated with both OVA and NaB). The NaB+AR group was given NaB in their feed (30 g/kg), whereas the other two groups were fed normal feed from 3 weeks to 6 weeks of age. At 7 weeks of age, OVA administration was initiated to induce AR in the AR and NaB+AR groups. Then, behavioral assessments, western blotting, and gene analysis were performed.

Effects of anticholinergic agent on miRNAs profile and transcriptome in murine model of allergic rhinitis

Project description:Purpose: Ipratropium bromide (IB) is able to ameliorate symptoms of allergic rhinitis (AR) by neuroimmunologic mechanism. We intended to explorer whether IB induced effect through modulating specific alteration of miRNA profiling or not. Methods: Nasal sRNAs (miRNAs) profiles of control mice and OVA-induced AR mice that were pre-treated with IB or normal saline before intranasal OVA challenge for 2 or 4 weeks were generated by high-throughput sequencing using Illumina Hiseq 2000.Raw data for sRNA (miRNA) profiles were processed to obtain corresponding clean data which were mapped to reference sequence(mm10) by TopHat(v2.0.9).The expression levels for these resulting mapped miRNAs were estimated by TPM (transcript per million),afterwards, the differentially expressed miRNAs were identified, and the candidate allergic-related miRNAs were further determined by RT-qPCR. Results: Using an optimized data analysis workflow,our data showed that in nasal mucosa of allergic mice with ipratropium bromide (IB) treatment 87 miRNAs are differentially expressed whereas in the nasal mucosa of non-allergic mice 113 miRNAs are differentially expressed, when compared with allergic mice treated with normal saline.IB treatment significantly up-regulated the levels of mmu-miR-124-3p/5p, -133b-5p, -133a-3p/5p, -384-3p, -181a-5p, -378a-5p, -3071-5p. RT-qPCR data further validated these miRNAs expression. Conclusions: Our study represents the detailed analysis of nasal mucosa miRNAs profiles of OVA-induced allergic mice treated with IB, generated by RNA-seq technology. Moreover, IB treatment orchestrated expression of allergic immune-related miRNAs of nasal mucosa in allergic mice, which may associate with ameliorated nasal allergic symptoms

This research elucidated distinct subpopulations and rare cell types of epithelial cells within the nasal turbinates, discerning alterations induced by allergic rhinitis. Microglia response to allergic airway inflammation during early postnatal period in mice

Project description:We examined how allergic inflammation in early postnatal period influences microglia using a short- and long-term airway allergy model. Male mice (C57BL/6) were immunized by intraperitoneal injection with aluminum hydroxide and ovalbumin (OVA) or phosphate-buffered saline (PBS) (control) at postnatal days (P) 3, 7, and 11, followed by intranasal challenge with OVA or PBS solution twice a week until P30 or P70. Microglia from whole brain were isolated from PBS- and OVA-treated mice by gating on CD11b+CD45int cells with flowcytometry at P30 and P70 respectively. Results suggest the dysregulation of microglial genes related to synaptic pruning and clearance, aging, and corticosteroid pathway and distinction between short- and long- term allergen challenge.
