Project description:As part of the Taiwanese Consortium of Childhood Asthma Study (TCCAS), a new prospective sub-study was conducted to investigate the effectiveness of inhaled corticosteroids (ICS) treatment. Patients were categorized as either responders (R) or non-responders (NR), based on the improvement of their pulmonary function after ICS treatment. We analyzed the transcriptomes of asthma with ICS non-responsers and responders.
Project description:In this study, neutrophils isolated from human peripheral blood were cultured with PBS, tPA(10μg/ml) or tPA+HRG (0.5μm) for 2.5 hours at 37 ° C . Then, neutrophils were collected for proteomic analysis (n=6). Blood sample collection has been approved by Tianjin Medical University General Hospital Ethics Committee, and collection was conducted according to the Declaration of Helsinki. Informed consent was obtained from all participants, and all experiments were conducted in accordance with Chinese laws and institutional guidelines.
Project description:The transcriptional patterns of mature normal PB neutrophils (granulocytes) was examined and used as a control against which the transcriptional programs of cultured granulocytes and granulocytic cells lines can be compared. Experiment Overall Design: RNA was extracted from peripheral blood neutrophils. Three biological replicate experiments were analyzed and approximately 67% of the samples were technically replicated. Hybridizations were performed in a reference design with all samples labeled with Cy3 and a reference RNA pool labeled with Cy5.

Project description:A large, prospective, non-interventional study was designed to study gene expression changes in peripheral blood mononuclear cells (PBMCs) associated with asthma exacerbations over the course of a year. PBMC samples were collected from subjects at the time of the study visits defined as 1) Quiet: during stable disease at 3 month intervals, 2) Exacerbation: during a 14 day period of deteriorating asthma and 3) Follow-up: within 14 days after cessation of an exacerbation. Gene expression levels during stable asthma, exacerbation, and two weeks after an exacerbation were compared. This series of samples comprises of peripheral blood mononuclear cells (PBMCs) collected during the following study visits (a) quiet (N=394 samples from 118 subjects), (b) exacerbation (N=166 samples from 118 subjects) and (c) follow-up (N=125 samples from 102 subjects)
