Project description:Previously, we identifed alveolar Type II cells as cell-of-origin for Kras induced lung adenocarcinoma. In the current study, we examined the phenotype of Type II cells after Kras activation and found evidence for proliferation of cells that co-express Type I and Type II markers. Using RNAseq, we are trying to charcterize these different alveolar epithelial cells (Type I, Type II and double positive (Type I/II+)) upon KrasG12D activation during lung adenocarcinoma.

Transcriptome in alveolar epithelial type II cells isolated from normal and COPD lungs of adult human

Project description:Alveolar epithelial type II (ATII) cells play a critical role in homeostasis and repair process of the lungs. In lung diseases such as chronic obstructive pulmonary disease (COPD), ATII cells are damaged and fall into apoptosis or senescence. Until to date, global gene expression of ATII cells in COPD lungs has not been analyzed. We isolated ATII cells from three non-COPD and three COPD patients using a FACS method. Then, we performed microarray analysis to compare gene expression profiles of ATII cells between non-COPD and COPD patients.

Gene expression in T1alpha null mutant lungs compared to wild type at dpc 18.5 and term

Project description:We compared gene expression in T1alpha (-/-) lungs vs wild type at dpc 18.5 when the phenotype is moderate and at term when the phenotype is more severe. Lungs present narrow and irregular alveolar spaces, thicker mesenchyme, reduced number of attenuated type I cells, normal numbers of type II cells and secreted surfactant. At term, but not a dpc 18.5, there is increased proliferation of distal cells. We compared gene expression of T1alpha null mutant lungs to wild type lungs at dpc 18.5. The mutant phenotype at this developmental stage is moderate but the alveolar sacs are narrower than normal. Keywords: other

Characterization of the genetic mouse model of non-small cells lung cancer p53R172HM-bM-^HM-^Fg/KrasG12D

Project description:Background: Non-small cell lung cancer (NSCLC) accounts for 81% of all cases of lung cancer and they are often fatal because 60% of the patients are diagnosed at an advanced stage. Besides the need for earlier diagnosis, there is a great need for additional effective therapies. In this work we investigated the feasibility of a lung cancer progression mouse model, mimicking features of human aggressive NSCLC cancer, as biological reservoir for potential therapeutic targets and biomarkers. Results:RNA-seq profiling was performed on total RNA extracted from lungs of 30 week-old p53R172HM-bM-^HM-^Fg/KrasG12D and wild type mice to detect fusion genes and gene/exon-level differential expression associated to the increase of tumor mass. Fusion events were not detected in p53R172HM-bM-^HM-^Fg/KrasG12D tumors. Differential expression at exon-level detected 33 genes with differential exon usage. The study provides a complete transcription overview of the p53R172HM-bM-^HM-^Fg/KrasG12D mouse NSCLC model Lung mRNA profiles of 30-week old wild type (WT) and p53R172HM-bM-^HM-^Fg/KrasG12D mice were generated by deep sequencing, in duplicate using Illumina HiSeq2000.

Comparison of KrasG12D/WT;p53Fx/Fx and KrasG12D/G12D;p53Fx/Fx MEFs

Project description:Microarray expression data generated to compare the biological impact of KrasG12D allelic duplication in p53null mouse embryonic fibroblasts (MEFs). The RAS/MAPK-signalling pathway is frequently deregulated in non-small cell lung cancer (NSCLC), often through activating mutations in KRAS. Mouse models demonstrated that activation of a single endogenous mutant Kras allele is sufficient to promote lung tumour formation, but acquisition of other genetic alterations is required for malignant progression. Using a well-established lung cancer mouse model we recently demonstrated that advanced KrasG12D-driven spontaneous tumours frequently exhibit enhanced MAPK signalling and KrasG12D allelic enrichment (KrasG12D/Kraswild-type>1), implying that mutant Kras copy gains are positively selected during lung cancer progression. To compare the oncogenic impact of a single mutant allele versus additional mutant Kras copy gain, we carried out a comprehensive analysis of mutant Kras homozygous and heterozygous MEFs and lung cancer cells and show that these genotypes are phenotypically distinct. mRNA expression pattern of KrasG12D mice with acute pancreatitis compared with wild-type mice without acute pancreatitis

Project description:Transgenic KrasG12D mice can recapitulate pancreas intra-epithelial neoplasia (PanIN). Caerulein is a cholecystokinin analogue and induces acute pancreatitis when injected intra-abdominally. Caerulein-induced acute pancreatitis will accelerate PanIN progression in KrasG12D mice. We compared mRNA profile changes between KrasG12D mice with acute caerulein-induced pancreatitis and wild-type mice without acute pancreatitis. The experiment had two groups. Experiment group: KrasG12D mice with acute caerulein-induced pancreatitis (N=6). Three mice in experiment group received 1-week caerulein injection, and the other three mice received 2-week caerulein injection. All experiment group mice started to receive caerulein injection at 1-month of age, and were sacrificed at the last day of caerulein injection. Control group: wild-type mice without acute pancreatitis (N=6). The mice were sacrificed at 1.5-month of age. Whole pancreas tissue lysate samples were subjected to mRNA array assay.
