Project description:A novel compact three-stage anaerobic digester (HM3) was developed to combine the advantages of high solids anaerobic digestion (AD) and wet AD for co-digestion of food waste and horse manure. By having three separate chambers in the three-stage anaerobic digester, three different functional zones were created for high-solids hydrolysis, acidogenesis and wet methanogenesis. The results showed that the functionalized partitioning in HM3 significantly accelerated the solubilization of solid organic matters and the formation of volatile fatty acids, resulting in an increase of 11~23% in methane yield. VS reduction in the HM3 presents the highest rate of 71% compared to the controls. Pyrosequencing analysis indicated that different microbial communities in terms of hydrolyzing bacteria, acidogenic bacteria and methanogenic archaea were selectively enriched in the three separate chambers of the HM3. Moreover, the abundance of the methanogenic archaea was increased by 0.8~1.28 times compared to controls.

Project description:BackgroundMost of the organic content of waste activated sludge (WAS) comprises microbial cells hard to degrade, which must be pre-treated for energy recovery by anaerobic digestion (AD). Electrooxidation pre-treatment (EOP) with boron-doped diamond (BDD) electrode have been considered a promising novel technology that increase hydrolysis rate, by the disintegrating cell walls from WAS. Although electrochemical oxidation could efficiently solubilize organic substances of macromolecules, limited reports are available on EOP of WAS for improving AD. In this endeavour, the mathematical optimization study and the energy analysis of the effects of initial total solids concentrations [TS] of WAS and current density (CD) during EOP on the methane production and removal of chemical oxygen demand (COD) and volatile solids (VS) were investigated. Because limited reports are available on EOP of WAS for improving biogas production, it is not well understood; however, it has started to attract interest of scientists and engineers.ResultsIn the present work, the energy recovery as biogas and WAS conversion were comprehensively affected by CD and [TS], in an integrated EOP and AD system. When working with WAS at 3% of [TS] pre-treated at current density of 24.1 mA/cm2, the highest COD and VS removal were achieved, making it possible to obtain the maximum methane (CH4) production of 305 N-L/kg VS and a positive energy balance of 1.67 kWh/kg VS. Therefore, the current densities used in BDD electrode are adequate to produce the strong oxidant (hydroxyl radical, ·OH) on the electrode surface, allow the oxidation of organic compounds that favours the solubilization of particulate matter and VS from WAS.ConclusionsThe improvement of VS removal and COD solubilization were due to the effects of pre-treatments, which help to break down the microbial cells for faster subsequent degradation; this allows a decomposition reaction that leads to biodegrade more compounds during AD. The balance was positive, suggesting that even without any optimization the energy used as electricity could be recovered from the increased methane production. It is worth noting that this kind of analysis have not been sufficiently studied so far. It is therefore important to understand how operational parameters can influence the pre-treatment and AD performances. The current study highlights that the mathematical optimization and energy analysis can make the whole process more convenient and feasible.

Project description:The potential of periodate (PI) in sludge anaerobic digestion is not tapped, although it has recently attracted great research interest in organic contaminants removal and pathogens inactivation in wastewater treatment. This is the first work to demonstrate significant improvement in methane generation from waste activated sludge (WAS) with PI pretreatment and to provide underlying mechanisms. Biochemical methane potential tests indicated that methane yield enhanced from 100.2 to 146.3 L per kg VS (VS, volatile solids) with PI dosages from 0 to 100 mg per g TS (TS, total solids). Electron spin resonance showed PI could be activated without extra activator addition, which might be attributed to the native transition metals (e.g., Fe2+) in WAS, thereby generating hydroxyl radical (•OH), superoxide radicals (•O2 -), and singlet oxygen (1O2). Further scavenging tests demonstrated all of them synergistically promoted WAS disintegration, and their contributions were in the order of •O2 - > •OH > 1O2, leading to the release of substantial biodegradable substances (i.e., proteins and polysaccharides) into the liquid phase for subsequent biotransformation. Moreover, fluorescence and ultraviolet spectroscopy analyses indicated the recalcitrant organics (especially lignocellulose and humus) could be degraded by reducing their aromaticity under oxidative stress of PI, thus readily for methanogenesis. Microbial community analysis revealed some microorganisms participating in hydrolysis, acidogenesis, and acetoclastic methanogenesis were enriched after PI pretreatment. The improved key enzyme activities and up-regulated metabolic pathways further provided direct evidence for enhanced methane production. This research was expected to broaden the application scope of PI and provide more diverse pretreatment choices for energy recovery through anaerobic digestion.

Project description:The uncontrolled discharge of industrial wastewater leads to a significant cadmium (Cd) accumulation in waste activated sludge (WAS), posing a serious threat to the steady operation of the anaerobic digestion (AD) system in wastewater treatment plants (WWTPs). Therefore, developing a viable approach to cope with the adverse effects of high-concentration Cd on the AD system is urgently required. This study aims to investigate the potential of using anionic polyacrylamide (APAM), a commonly used agent in WWTPs, to mitigate the adverse effects of Cd in a toxic amount (i.e., 5.0 mg per g total suspended solids (TSS)) on AD of WAS. The results showed that the effectiveness of higher APAM on Cd toxicity alleviation was less than that of lower APAM at the studied level (i.e., the effectiveness order was 1.5 mg APAM per g TSS > 3.0 mg APAM per g TSS > 6.0 mg APAM per g TSS). The moderate supplement of APAM (i.e., 1.5 mg per g TSS) recovered the accumulative methane yield from 190.5 ± 3.6 to 228.9 ± 4.1 mL per g volatile solids by promoting solubilization, hydrolysis, and acidification processes related to methane production. The application of APAM also increased the abundance of key microbes in the AD system, especially Methanolinea among methanogens and Caldilineaceae among hydrolyzers. Furthermore, APAM facilitated the key enzyme activities involved in AD processes and reduced reactive oxygen species (induced by Cd) production via adsorption/enmeshment of Cd by APAM. These findings demonstrate the feasibility of using moderate APAM to mitigate Cd toxicity during AD, providing a promising solution for controlling Cd or other heavy metal toxicity in WWTPs.

Project description:Anaerobic digestion (AD), being the most effective treatment method of waste activated sludge (WAS), allows for safe disposal. The present study deals with the electro-Fenton (EF) pretreatment for enhancing the WAS biogas potential with low-cost iron electrodes. The effect of pretreatment on the physicochemical characteristics of sludge was assessed. Following EF pretreatment, the pH, conductivity, soluble chemical oxygen demand (SCOD), and volatile fatty acids (VFA) increased to 7.5, 13.72 mS/cm, 4.1 g/L, and 925 mg/L, respectively. Capillary suction time (CST) analysis highlighted the dewaterability effect of EF on WAS, as demonstrated by the decrease in CST from 429 to 180 s following 30 min of pretreatment. Batch digestion assays presented an increase in the biogas yield to 0.135 L/g volatile solids (VS) after 60 min of EF pretreatment in comparison to raw sludge (0.08 L/g VS). Production of biogas was also found to improve during semi-continuous fermentation of EF-pretreated sludge conducted in a lab-scale reactor. In comparison to raw sludge, EF-pretreated sludge produced the highest biogas yield (0.81 L biogas/g VS) with a high COD removal rate, reaching 96.6% at an organic loading rate (OLR) of 2.5 g VS/L. d. Results revealed that the EF process could be an effective WAS disintegration method with maximum recovery of bioenergy during AD.

Project description:Anaerobic co-digestion (Co-AD) is used to increase the effectiveness of anaerobic digestion (AD) using local "wastes", adding economic and environmental benefits. Since system stability is of existential importance for the operation of wastewater treatment plants, thorough testing of potential co-substrates and their effects on the respective community and system performance is crucial for understanding and utilizing Co-AD to its best capacity. Food waste (FW) and canola lecithin (CL) were tested in mesophilic, lab-scale, semi-continuous reactors over a duration of 120 days with stepwise increased substrate addition. Key performance indicators (biogas, total/volatile solids, fatty acids) were monitored and combined with 16S-rRNA amplicon sequencing to assess the impact of co-substrate addition on reactor performance and microbial community composition (MCC). Additionally, the latter was then compared with natural shifts occurring in the wastewater treatment plant (WWTP, source) at the same time. An almost linear increase in biogas production with both co-substrates at an approximate 1:1 ratio with the organic loading rate (OLR) was observed. The MCCs in both experiments were mostly stable, but also prone to drift over time. The FW experiment MCC more closely resembled the original WWTP community and the observed shifts indicated high levels of functional redundancy. Exclusive to the CL co-substrate, a clear selection for a few operational taxonomic units (OTUs) was observed. There was little evidence for a persistent invasion and establishment of microorganisms from typical primary substrates into the stable resident community of the reactors, which is in line with earlier findings that suggested that the inoculum and history mostly define the MCC. However, external factors may still tip the scales in favor of a few r-strategists (e.g., Prolixibacter) in an environment that otherwise favors K-strategists, which may in fact also be recruited from the primary substrate (Trichococcus). In our study, specialization and diversity loss were also observed in response to the addition of the highly specialized CL, which in turn, may have adverse effects on the system's stability and reduced resilience and recovery.

Project description:Anaerobic digestion (AD) plays an important role in waste activated sludge (WAS) treatment; however, conventional AD (CAD) process needs substantial improvements, especially for the treatment of WAS with low solids content and poor anaerobic biodegradability. Herein, we propose a submerged anaerobic dynamic membrane bioreactor (AnDMBR) for simultaneous WAS thickening and digestion without any pretreatment. During the long-term operation, the AnDMBR exhibited an enhanced sludge reduction and improved methane production over CAD process. Moreover, the biogas generated in the AnDMBR contained higher methane content than CAD process. Stable carbon isotopic signatures elucidated the occurrence of combined methanogenic pathways in the AnDMBR process, in which hydrogenotrophic methanogenic pathway made a larger contribution to the total methane production. It was also found that organic matter degradation was enhanced in the AnDMBR, thus providing more favorable substrates for microorganisms. Pyrosequencing revealed that Proteobacteria and Bacteroidetes were abundant in bacterial communities and Methanosarcina and Methanosaeta in archaeal communities, which played an important role in the AnDMBR system. This study shed light on the enhanced digestion of WAS using AnDMBR technology.

Project description:BACKGROUND:Recently, it has been indicated that anaerobic co-digestion of waste activated sludge with other waste streams at wastewater treatment plants is a promising strategy for enhancing methane production and materials recovery. The enhanced methane production can be used as a renewable source of energy in wastewater treatment plants. It can also reduce the amount of greenhouse gas emission in landfilling of the waste streams. RESULTS:According to the results obtained in this study, anaerobic co-digestion of waste activated sludge with mixed fruit waste and cheese whey improves methane production and the quality of digested sludge in comparison to the anaerobic digestion of waste activated sludge individually. It was indicated that carbon/nitrogen ratio (C/N) in the mixture of waste activated sludge, fruit waste and cheese whey improved considerably, leading to better anaerobic organisms' activity during digestion. With assessing the activity of protease and cellulase, as the main enzymes hydrolyzing organic matter in anaerobic digestion, it was indicated that co-digestion of waste activated sludge with mixed fruit waste and cheese whey enhances the activity of these enzymes by 22 and 9% respectively. At the end of digestion, the amount of cumulative methane production significantly increased by 31% in the reactor with 85% waste activated sludge and 15% mixed fruit waste and cheese whey, compared to the reactor with 100% waste activated sludge. In addition, chemical oxygen demand (COD) and volatile solid (VS) in digested sludge was improved respectively by 9 and 7% when mixed fruit waste and cheese whey was used. CONCLUSIONS:This study revealed that mixed fruit waste and cheese whey is potentially applicable to anaerobic digestion of waste activated sludge, as fruit waste and cheese whey have high C/N ratio that enhance low C/N in waste activated sludge and provide a better diet for anaerobic organisms. This is of significant importance because not only could higher amount of renewable energy be generated from the enhanced methane production in wastewater treatment plants, but also capital costs of the companies whose waste streams are being transported to wastewater treatments plants could be reduced considerably.
