Project description:The use of single cell sequencing technologies has exploded in non-model organisms, including fish and invertebrates in an aquaculture setting. One of the most expanding areas is the use sequencing nucleus from frozen tissues allowing being able to sample and sequence at different sites. Here we present a tested nucleus isolation protocol that outperforms the most reliable methods to extract nuclei (chopping in salt tween buffer) and improves its performance in non-model organisms.

Project description:The use of single cell sequencing technologies has exploded over recent years, and is now commonly used in many non-model species. Sequencing nuclei instead of whole cells has become increasingly popular, as it does not require the processing of samples immediately after collection. Here we present a highly effective nucleus isolation protocol that outperforms previously available method in challenging samples in a non-model specie. This protocol can be successfully applied to extract nuclei from a variety of tissues and species.

Assesing an improved protocol for plasma microRNA extraction

Project description:The first step in biomarkers discovery is to identify the best protocols for their purification and analysis. We have identified an optimal RNA extraction method of microRNAs from human plasma samples. We also report that the addition of low doses of carrier RNA before starting RNA extraction improves microRNA extraction and quantification. Human plasma and matched biopsies were obtained from healthy donors and patients attended at the Hospital Universitari i Politècnic La Fe (Valencia, Spain). RNA was extracted by different preanalytal conditions and reagents, testing the suitable of carrier addition at differnt doses. The best protocol was followed up by hybridation on Affymetrix microarrays.
