Project description:To gain a deeper insight into roles of Arid1a in mice hippocampus development, we performed ChIP-seq and RNA-seq to analyze the genome-wide changes of histone marks and transcriptome changes before and after Arid1a deletion in hippocampus of the E16.5 and P21 mouse using Cre-lox system.
Project description:To understand the genetic regulation of gene expression and patterns of gene co-expression, we sequenced the transcriptome of the hippocampus of 258 Diversity Outbred (DO) mice of both sexes. DO mice (fourth and fifth generations of outcrossing) were sacrificed between 6-8 weeks of age and hippocampus dissected. Total hippocampal RNA was isolated using a TRIzol Plus RNA purification kit (Life Technologies) and mRNA sequencing library was prepared using a TruSeq kit (Illumina), both according to manufacturer's protocols. Paired-end 100bp reads were obtained using the Illumina HiSeq 2000.
Project description:We performed RNA sequencing for hippocampus of neonatal mice that were exposed to sevoflurane. We treated a group of neonatal mice with 2.5% sevoflurane for 2 hours on day 6, 7, 8, 9 and treated another group on day 6, 7. On day 36, we collected RNA (both total RNA and polysome-associated RNA) from the hippocampus in the aforementioned two groups and the control group (i.e., without sevoflurane treatment). We prepared the sequencing libraries with the collected RNA and sent them to Illumina sequencing platform.
