Project description:MicroRNAs are a class of endogeneously expressed non-coding small, ~21nt RNAs involved in the negative regulation of gene expression. In plants, miRNAs are known to play a critical role in developmental and metabolic pathways, as they predominantly target transcription factors. Studies in Arabidopsis and apple have shown that few microRNAs and small interfering (si) RNAs target MYB transcription factors, which are key regulators of phenylpropanoid pathway. However, it is not well-understood how miRNAs mediate regulation of MYBs to produce secondary metabolites such as anthocyanins and flavonoids. Here we show that, a cluster of abundant miRNAs target MYB transcription factors in anthocyanin rich fruits such as grapes. Using deep small RNA-sequencing we establish that grape varieties with high anthocyanin content express abundant MYB-targeting miRNAs resulting in differential expression of MYB proteins among grape varieties, thereby regulating the phenylpropanoid pathway.

Project description:A double cropping system has been commercially adopted in subtropical regions in southern China, where there is abundant sunshine and heat resources. In this viticulture system, the first growing season normally starts as a summer cropping cycle; then, the vine is pruned and forced by hydrogen cyanamide, resulting in a second crop in January of the next year. Due to climate differences between the two growing seasons,the transcriptome varies greatly between summer- and winter- grape berries. In this study, we conducted RNA-seq for two table grape varieties (Vitis vinifera cv. Victoria and V. vinifera cv. Muscat Superior) at three developmental stages (pre-veraison, post-veraison and full maturity).The transcriptome between summer- and winter- grape berries were compared.

Project description:Grape volatiles include a great number of compounds, among which monoterpenes, alcohols,esters and carbonyls were found.Grape may be divided into aromatic and non-aromatic varieties. ‘Shine Muscat’ belongs to the aromatic cultivar. The most abundant free compounds detected in Muscat grape were linalool, geraniol, citronellol, nerol. Grapevine (Vitis vinifera L.) is an economically important and widely cultivated fruit crop. Grape quality is important for its market value and is largely decided by its taste and aroma.Gas-chromatograph mass-spectrometry (GC-MS) was performed to observe changes of the volatile compounds.

Project description:Grape berry development is a highly coordinated, intricate and complex process with many morphological, biochemical and physiological changes occurring during the ripening process. Equally, ripening is an organoleptic characteristic linked to fruit development. The fruits Seedless (FS) and Victoria (VT) grape varieties exhibit many morphological and phytochemical differences, but genetic mechanisms underlying them remain poorly explored. Herein, we comparatively analysed the phenotypic and transcriptomic patterns of Victoria (VT) and Flame Seedless (FS) grape varieties during berry development. We studied the physiological analysis and transcriptomic profiles sequencing were performed at four berry developmental stages time-points (40, 50, 60 and 80 DPA). Notably, the VT variety berry size was comparatively larger to the FS variety. At maturity, 80DPA, the FS soluble solids were 61.8% higher than VT. Further, a total of 4889 and 2802 DEG’s were identified from VT and FS 40 DPA to 80 DPA development stages, respectively. 1386 DEGs were common in the two varieties. GO analysis identified cysteine biosynthetic process, response to red light, chlorophyll binding, polysaccharide biosynthetic process and chloroplast thylakoid membrane as some of the dominant terms under the biological processes, molecular function and cellular component categories.
