Examining the role of ER ⍺ and β in ovarian granulosa cell transdifferentiation in aged ovaries

Project description:To examine the role of estrogen receptors (ERs) on ovarian function in aged animals and indentify genes that may contribute to ovarian transdifferentation in ovaries lacking both ER⍺ and Erβ. Gene expression was examined in single knockout and double knockout ovaries and identified genes that were different in each. Among the three knockout animals examined, only 43 genes were shared among the Ex3⍺ERKO, Ex3βERKO and Ex3⍺βERKO compared to the control. The Ex3⍺ERKO ovaries had 645 genes differentially expressed, the Ex3βERKO had 94 genes differentailly expressed and the Ex3⍺βERKO had the most differentially expressed with 2,193 genes compared to control ovaries which had one allele of each ER.

Gene expression regulated by transcription factor MiT in Drosophila

Project description:To understand the role of MiT in Drosophila, we set out to identify critical gene targets by looking at changes in the WT transcriptome induced by either gain or loss of MiT function. Mutant hindgut and malpighian tubules provided loss-of function tissue and nub-Gal4-driven expression of MiT in the wing epithelium was used for gain-of-function. In the wing disc experiment, 543 genes were upregulated by exogenous MiT, and 359 genes were downregulated (>1.4 fold; P value < 0.01). In the larval HG+MT, 897 genes were downregulated and 898 were upregulated (>1.4 fold; P value < 0.01) after MiT. Among these genes, 85 were both upregulated in wing discs and downregulated in mutant HG+MT, and are the common genes that regulated by MiT in both tissues. We set out to identify potential targets of MiT by comparing the transcriptional profiles of wing discs with and without MiT overexpression (nub-Gal4 UAS-MiT versus nub-Gal4), and separately the profiles of HG+MT tissue from WT and MiT mutant (MiTTZ2/Df(4)TZ). Then, we focused on genetic loci that were upregulated by gain of MiT in the wing and downregulated by loss of MiT in the HG+MT.

mTORC1 feedback to AKT modulates MiT/TFE-driven lysosomal biogenesis and EGFR degradation

Project description:MiT/TFE transcriptional activity controls lysosomal biogenesis and is negatively regulated by the nutrient sensor mTORC1. Some tumors bypass this regulatory circuit via genetic alterations that drive MiT/TFE expression and activity; however, the mechanisms by which cells with intact or constitutive mTORC1 signaling maintain lysosomal catabolism remain to be elucidated. Using the murine epidermis as a model system, we find that epidermal Tsc1 deletion results in a wavy hair phenotype due to increased EGFR degradation. Unexpectedly, constitutive mTORC1 activation increases lysosomal content via up-regulated expression and activity of MiT/TFEs, while genetic or prolonged pharmacologic mTORC1 inactivation has the reverse effect. This paradoxical up-regulation of lysosomal biogenesis by mTORC1 is mediated by feedback inhibition of AKT, and a resulting suppression of AKT-induced MiT/TFE proteasomal degradation. These data suggest that oncogenic feedback loops work to restrain or maintain cellular lysosomal content during chronically inhibited or constitutively active mTORC1 signaling respectively, and reveal a mechanism by which mTORC1 regulates upstream receptor tyrosine kinase signaling.
