Managing the tradeoff between reproduction and survival requires flexibility in behavior and gene regulation in threespined stickleback

Project description:Predators exert a powerful selective force, however predatoravoidance can conflict with other important activities such as attracting mates. Decisions over whether to court mates versus avoid predators are vital to fitness, yet the mechanistic underpinnings of how animals manage such tradeoffs are poorly understood. Here, we investigate flexibility of behavior and gene regulation in response to a tradeoff between avoiding predators (survival) and courting potential mates (reproduction) in threespined stickleback (Gasterosteus aculeatus). We compared behavioral and transcriptomic responses of male sticklebacks faced with a courtship opportunity and cues of a predator simultaneously to males faced with a courtship opportunity or cues of a predator alone, and found that males behaviorally compromised courtship in favor of predator avoidance when faced with a tradeoff between them. The need to manage this tradeoff elicited dynamic changes in brain gene expression, and sets of functionally-connected genes were organized into discrete modules based on co-expression. Additionally, we found that behavioral flexibility in response to tradeoffs corresponded to flexibility in gene regulatory network structure. Combined, these results uncover the coordinated response by the brain to a fundamental ecological tradeoff, providing insight into the structure and function of genetic networks underpinning how animals make fitness-influencing decisions.

Project description:We examine whether there are differences in gene expression between aggressive versus nonaggressive behavioral types of individuals within a natural population of male threespined sticklebacks (Gasterosteus aculeatus). We compared gene expression profiles of aggressive male sticklebacks to nonaggressive males in four regions of the brain (brainstem, cerebellum, diencephalon and telencephalon). We identified new candidate genes related to aggressiveness, including SLC38a, a solute carrier which has been linked to GABAergic neuron reuptake of glutamine.

Project description:The aim of this study was to describe the excretory–secretory proteins from the helminth Schistocephalus solidus and its intermediate host, the three-spined stickleback Gasterosteus aculeatus L., which are likely to be involved in interactions between them. Combined samples of washes from the G. aculeatus sticklebacks cavity infected with the S. solidus, and washes from the parasite surface were used as experimental samples (samples 5 to 8), while washes from the uninfected fish body cavity were used as control (samples 1 to 4). The obtained samples were analyzed using mass-spectrometry nLC–MS/MS.

Microarray analysis of di-n-butyl phthalate and 17α ethinyl-oestradiol responses in three-spined stickleback testes

Project description:The data comes from a study, where three-spined sticklebacks (Gasterosteus aculeatus) were exposed to di-n-butyl phthalate (DBP) and 17α ethinyl-oestradiol (EE2) at nominal concentrations 35 μg/L and 40 ng/L, respectively, for four days. The aim of the study was to obtain insight into the acute transcriptional responses putatively associated with endocrine disruption. RNA samples from the testes of eight individuals fish per treatment (including solvent controls, exposed only to DMSO) were used in the microarray analysis, covering the expression of approximately 21000 genes.

Neurogenomic insights into paternal care and its relation to territorial aggression

Project description:Motherhood is characterized by dramatic changes in brain and behavior, but less is known about fatherhood. Here we report that male sticklebacks – a small fish in which fathers provide care – experience dramatic changes in neurogenomic state as they become fathers. Some genes are unique to different stages of paternal care, some genes are shared across stages, and some genes are added to the previously acquired neurogenomic state. Comparative genomic analysis suggests that some of these neurogenomic dynamics resemble changes associated with pregnancy and reproduction in mammalian mothers. Moreover, gene regulatory analysis identified transcription factors that are regulated in opposite directions in response to a territorial challenge versus during paternal care. Altogether these results show that some of the molecular mechanisms of parental care might be deeply conserved and might not be sex-specific, and suggest that tradeoffs between opposing social behaviors are managed at the gene regulatory level.

Proteome of Schistocephalus solidus (Cestoda) after short- and long-term heating

Project description:The protein composition of the cestode Schistocephalus solidus was measured in an experiment simulating the transition of the parasite from a cold-blooded to a warm-blooded host. Infective S. solidus plerocercoids obtained from the three-spined stickleback Gasterosteus aculeatus were heated at 40 °C for 1 hour or cultured in vitro at 40 °C and 22 °C for 48 hours. Plerocercoids taken out from the body cavity of sticklebacks immediately after their sacrifice were used as a reference group.

Project description:We investigated the transcriptomic response to courtship opportunity in four regions of the brain (telencephalon, diencephalon, cerebellum and brain stem) of a teleost fish, the threespined stickleback (Gasterosteus aculeatus). We tested the hypothesis that gene regulatory networks modulate the transcriptional response tocourtship opportunity in the different brain regions and thereby mating. Indeed, transcription regulatory network analysis identified cis-regulatory transcription factor motifs enriched in upregulated genes in one brain region and downregulated in the others. We further validated our findings by qPCR.
