Project description:9-day-old Sertoli cells have a stronger proliferative activity compared with 21-day-old Sertoli cells. We used microarrays to detail the global programme of gene expression underlying cellularisation and identified distinct classes of up-regulated and down-regulated genes during this process. Rat Sertoli cell were selected at postnatal day 9 and postnatal day 21 for RNA extraction and hybridization on Affymetrix microarrays. We sought to obtain the differentially expressed genes that were involved in cell proliferation, cell cycle, and cell death which were associated with the function of Sertoli cell.
Project description:Neonatal mouse intervertebral discs lose regenerative potential between postnatal day 14 and day 28during a period of Intervertebral disc (IVD) defects heal poorly and can cause back pain and disability. We identified that IVD herniation injury heals regeneratively in neonatal mice until postnatal day 14 (p14) and shifts to fibrotic healing by p28. This age coincides with the shift in expansive IVD growth from cell proliferation to matrix elaboration, implicating collagen crosslinking. -aminopropionitrile treatment reduced IVD crosslinking and caused fibrotic healing without affecting cell proliferation. Bulk sequencing on naïve IVDs were depleted for matrix structural organization from p14 to p28 to validating the importance of crosslinking in regenerative healing. We conclude that matrix changes are key drivers in the shift to fibrotic healing, and a stably crosslinked matrix is needed for IVD regeneration.

Project description:The developing brain is particularly sensitive to ethanol during the brain growth spurt or synaptogenesis (third human trimester equivalent). This has been shown to lead to abnormal brain development and behavioural changes in the adult mouse that are relevant to those seen in humans with fetal alcohol spectrum disorders (FASD). We evaluated the acute (4h post-treatment) gene expression changes that occur in the brain due to ethanol exposure during synaptogenesis (postnatal day 7). We used microarray analyses to evaluate the changes in brain gene expression at postnatal day 7 that occur due to ethanol treatment at postnatal day 7 (synaptogenesis).
