Project description:Obesity causes the change of microenvironment of adipose tissue and induces the occurrence of adipose tissue inflammation. The objective of this study was to analyze the internal mechanism of microenvironment change in mouse parepididymal adipose tissue by single-nucleus RNA sequencing. Nutritional obesity mouse model was established and verified by morphology, blood index and liver oil red O staining. Select Ctrl group, Mid_Ob group and Ob group, and take 8 mice at 3 time points. The changes of cell population, gene and signaling pathway expression in the adipose tissue of the parastriatic muscle in mice during obesity were detected by single-nucleus RNA sequencing and immunohistochemistry. According to the results of single-nucleus RNA sequencing, adipose tissue was divided into fibroblasts, macrophages, neutrophils and endothelial cells. It is found that all cell types have changes during the development of obesity and play an important role in regulating the microenvironment of adipose tissue.

Plasticity of epididymal adipose tissue in response diet-induced obesity at single-nucleus resolution.

Project description:Adipose tissues display a remarkable ability to adapt to the dietary status. Here, we have applied single-nucleus RNA-seq to map the plasticity of mouse epididymal white adipose tissue at single cell resolution in response to high-fat diet-induced obesity. The single-nucleus approach allowed us to recover all major cell types and to reveal distinct transcriptional stages along the entire adipogenic trajectory from preadipocyte commitment to mature adipocytes. We demonstrate the existence of different adipocyte subpopulations and show that obesity leads to disappearance of the lipogenic subpopulation and increased abundance of the stressed lipid scavenging subpopulation. Moreover, obesity is associated with major changes in the abundance and gene expression of other cell populations, including a dramatic increase in lipid handling genes in macrophages at the expense of macrophage-specific genes. The data provide a powerful resource for future hypothesis-driven investigations of the mechanisms of adipocyte differentiation and adipose tissue plasticity.

Project description:Visceral white adipose tissue is closed correlated with obesity and metabolic dysfunction. Epididymal adipose tissue (eWAT) is considered as typical visceral white adipose tissue. Induction of browning of white adipose tissue improves metabolic dysfunction such as insulin resistance. In contrast to mice subcutaneous adipose tissue, visceral fat do not show significant browning under 4°C. However,under physiologically tolerable low temperature visceral adipose tissue can turn brown. We used microarrays to detail the global programme of gene expression in C57Bl/6 mice epididymal adipose tissue exposed to thermoneutral 30°C, 4°C and temperatures lower than 4°C.

The Ubiquitin Ligase Siah2 Regulates Obesity-induced Adipose Tissue Inflammation

Project description:Chronic, low-grade adipose tissue inflammation associated with adipocyte hypertrophy is an important link in the relationship between obesity and insulin resistance. Although ubiquitin ligases are essential regulators of inflammatory processes, the role of these enzymes in metabolically driven adipose tissue inflammation is relatively unexplored. In this study, we found that the ubiquitin ligase Siah2 is a central factor in obesity-related adipose tissue inflammation. When challenged with chronic excess energy intake, Siah2-null mice become obese with enlarged adipocytes, but do not develop obesity-induced insulin resistance. Proinflammatory gene expression is substantially reduced in the Siah2-null epididymal adipose tissue of the obese Siah2KO mice.
