Project description:The response of kiwifruit vine (Actinidia chinensis 'Hort16A') canes to feeding by the armoured scale insect Hemiberlesia lataniae was tested by screening RNA extracted from bark against a cDNA library of 17,512 unigenes. Substantial transcriptional changes were noted after 7 days for approximately 30% of the ESTs. Transcripts associated with photosynthesis were down-regulated and those associated with secondary metabolism up-regulated. A large number of transcripts orthologous with defence-related genes were differentially expressed including several orthologous with resistance and pathogenesis. This is the first such analysis of changes to transcript expression in a plant challenged by feeding by insects from the family diaspididae.

Project description:KMD is genetically engenered to be highly resistant to lepidopteran pests through expressing a synthetic cry1Ab gene and its parent non-transgenic rice is Xiushui 11.The developmental duration of BPH feeding on KMD2 was significantly delayed. And moreover, the fecundity of BPH was significantly lower when fed on Bt rice than on the non-Bt parental plants.To investigate unintended effects in KMD2 that causes changes in BPH performance, we performed microarray (GeneChip) analysis to compare the gene expression profiles between Bt rice and non-transgenic parental plants in response to BPH infestation. We used microarrays to detect Bt-independent variation, which might render Bt rice more defensive or less nutritious to BPH. For BPH treatment, 10 second-instar nymphs were infested onto each 30-day-old seedling. After 72 h, the BPH nymphs were carefully removed and rice shoots of both BPH-infested and non-infested plants were sampled for microarray analysis. There were four treatments: Xiushui 11-non infested, Xiushui 11-BPH infested, KMD2-non infested, KMD2-BPH infested, three biological replications.

Project description:We have implemented an integrated Systems Biology approach to analyze overall transcriptomic reprogramming and systems level defense responses in the model plant Arabidopsis thaliana during an insect (Brevicoryne brassicae) and a bacterial (Pseudomonas syringae pv. tomato strain DC3000) attack. The main aim of this study was to identify the attacker-specific and general defense response signatures in the model plant Arabidopsis thaliana while attacked by phloem feeding aphids or pathogenic bacteria. Defense responses and networks, unique and specific for aphid or Pseudomonas stresses were identified. Our analysis revealed a probable link between biotic stress and microRNAs in Arabidopsis and thus opened up a new direction to conduct large-scale targeted experiments to explore detailed regulatory links among them. The presented results provide a first comprehensive understanding of Arabidopsis - B. brassicae and Arabidopsis - P. syringae interactions at a systems biology level. Arabidopsis thaliana (ecotype Colombia-0) seeds were grown in 6-cm-diameter pots filled with a sterile soil mix (1.0 part soil and 0.5 part horticultural perlite), 3 plants per pot. Plants were kept in growth chambers VM-CM-6tsch VB 1514 (VM-CM-6tsch Industrietechnik GmbH, Germany) under the following conditions: a 8/16 h (light/dark) photoperiod at 22M-BM-0C/18M-BM-0C, 40%/70% relative humidity, and 70/0 M-NM-<mol m-2s-1 light intensity. After 32 days plants had 8 fully developed leaves. Each plant was infested with 32 wingless aphids [Brevicoryne Brassicae] (4 per leaf), which were transferred to leaves with a fine paintbrush. Infested plants and aphid-free controls were kept in plexiglass cylinders. Plants were harvested 72 h after infestation between the 6th and 8th hour of the light photoperiod. Four biological replicates were prepared from aphid infested and control plants, each sampled from 15 individual plants. Whole rosettes were cut at the hypocotyls and aphids were removed by washing with Milli-Q-filtered water. Differences in transcriptional responses were measured by comparing genes expression of aphid infested plants against non-infested control plants.

Project description:Infestation with white-backed planthopper (WBPH) to rice caused induced resistance to rice pathogens but brown planthopper (BPH) infestation induce weaker resistance to rice pathogens. We compared changes in gene expression in rice plants infested with WBPH and BPH to gain some insight into the WBPH-induced resistance to rice pathogens. An analysis, using microarrays, of gene expression in rice plants infested with these planthoppers revealed that WBPH infestation caused high induction of many defense-related genes including pathogenesis-related (PR) genes than BPH infestation. Furthermore, hydroperoxide lyase 2 (OsHPL2) which is an enzyme to produce C6 volatiles was induced by WBPH infestation, but not by BPH infestation. Keywords: response to herbivory; induced resistance

To decipher the molecular mechanisms involved in cotton boll/fibre cell development, transcriptome analysis has been carried out by comparing G. hirsutum L cv. Bikaneri narma cotton boll samples induced by biotic stress (bollworm infested) and that their respective control cotton bolls collected under field conditions. Cotton bolls were collected at fibre initiation (0, 2 dpa/days post anthesis) and elongation (5, 10 dpa) stages for both control and biotic stress condition and gene expression profiles were analyzed by Affymetrix cotton GeneChip Genome array.

Project description:Arabidopsis thaliana plants were infested i) with sucking insect herbivores (the generalist aphid Myzus persicae and the specialist aphid Brevicoryne brassicae), ii) with chewing insect herbivores (generalist caterpillars of Spodoptera exigua and specialist caterpillars of Pieris rapae) or iii) were treated by wounding. For each treatment, rosette leaves were harvested at two time points (6h and 24h) after removal of insects. For chewing herbivores and wounding both local, i.e. immediately damaged leaves, and systemic, i.e. undamaged leaves from the same plant, were collected. Control plants were uninfested, but otherwise equally treated and harvested in parallel. We tested the hypothesis that Arabidopsis can recognize and respond differentially to insect species at the transcriptional level using a genome wide microarray. Transcriptional reprogramming was characterized using co-expression analysis in damaged and undamaged leaves at two times in response to mechanical wounding and four insect species. In all, 2778 (10.6%) of annotated genes on the array were differentially expressed in at least one treatment. Responses differed mainly between aphid and caterpillar and sampling times. Responses to aphids and caterpillars shared only 10% of up-regulated and 8% of down-regulated genes. Responses to two caterpillars shared 21% and 12% of up- and down-regulated genes, whereas responses to the two aphids shared only 7% and 4% of up-regulated and down-regulated genes. Overlap in genes expressed between 6h and 24h was 3-15%, and depended on the insect species. Responses in attacked and unattacked leaves differed at 6h but converged by 24h. Genes responding to the insects are also responsive to many stressors and included primary metabolism. Aphids down-regulated amino acid catabolism; caterpillars stimulated production of amino acids involved in glucosinolate synthesis. Co-expression analysis revealed 17 response networks. Transcription factors were a major portion of differentially expressed genes throughout and responsive genes shared most of the known or postulated binding sites.

Likewise after two and five dayM-bM-^@M-^Ys infestation the bolls were collected and labelled as 2 and 5 dpa, respectively. After five days the insect was removed from bolls and left the bolls for up to 10 days and collected then labeled as 10 dpa. Meanwhile respective control samples also tagged and collected. Total RNA was isolated from control (WT) and biotic stress (bollworm infested) induced samples collected at 0, 2, 5 and 10 dpa boll development stages using SpectrumTM Plant Total RNA kit (Sigma, USA) according to the manufacturerM-bM-^@M-^Ys protocol. Affymetrix cotton GeneChip Genome array (Affymetrix, USA) having 23,977 probe sets representing 21,854 cotton transcripts was used for transcriptome analysis. Three biological replicates were maintained to test the reproducibility and quality of the chip hybridization. cDNA labeling, array hybridization, staining and washing procedures were carried out as described in the Affymetrix protocols. CEL files having estimated probe intensity values were analyzed with GeneSpring GX-11.5 software (Agilent Technologies, USA) to get differentially expressed transcripts. The Robust Multiarray Average (RMA) algorithm was used for the back ground correction, quantile normalization and median polished probe set summarization to generate single expression value for each probe set. Normalized expression values were log2 transformed and differential expression analysis was performed using unpaired t-test. The p-values were corrected by applying the false discovery rate (FDR) correction (Benjamini and Hochberg, 2000).

Project description:Salt stress is a rising threat to agriculture system. The accumulation of salts near the plant roots hampers the normal uptake of water causing osmotic stress and ionic toxicity to the plant. Thompson Seedless is a popular table grape variety of Vitis vinifera L., which is sensitive to salt stress when grown on its own roots; grafting it onto a wild rootstock such as 110 Richtor (110R) makes it tolerant to salt stress. In the present study, shotgun-proteomics approach was used for the investigation of salt stress induced molecular response of own rooted and 110R grafted Thompson Seedless grapevines. A salt stress experiment was conducted on sixteen month old potted grapevines. The grapevines were treated with 150mM NaCl solution for seven days and the control vines were irrigated with tap water. The young leaf samples were collected from control and treated vines at three time-points viz. 6 hours, 48 hours and 7 days of salt stress. The stress responsive proteins identified through statistical analysis revealed a distinct response to salinity in both the vines.
