Project description:Dysregulated autophagy is associated with cardiovascular and metabolic diseases, where impaired flow-mediated endothelial cell responses promote cardiovascular risk. The mechanism by which the autophagy machinery regulates endothelial functions is complex. We applied multi-omics approaches and in vitro and in vivo functional assays to decipher the diverse roles of autophagy in endothelial cells. We demonstrate that autophagy regulates VEGF-dependent VEGFR signaling and VEGFR-mediated and flow-mediated eNOS activation. Endothelial ATG5 deficiency in vivo results in selective loss of flow-induced vasodilation in mesenteric arteries and kidneys, along with increased cerebral and renal vascular resistance. We found a crucial pathophysiological role for autophagy in endothelial cells in flow-mediated outward arterial remodeling, prevention of neointima formation following wire injury, and recovery after myocardial infarction. Together, these findings unravel a fundamental role for autophagy in endothelial function, linking cell proteostasis to mechanosensing.

Project description:Background: Cellular communication among different types of vascular cells is indispensable for maintenance of vascular homeostasis and prevention of atherosclerosis (AS). However, the biological mechanism involved in cellular communication among these cells and whether this biological mechanism can be used to treat AS remain unknown. This study hypothesizes that endothelial autophagy mediates the cellular communication of vascular tissue through exosome-mediated delivery of AS-related genes.Methods: ApoE-/- mice fed with high-fat diet (HFD) were received rapamycin to activate autophagy, or were intravenously injected with adeno-associated virus (AAV) vectors carrying shRNA against Atg7 mRNA under the Tie (tyrosine kinase) promoter to specifically inhibit endothelial autophagy. miRNA microarray, in vivo and in vitro experiments and human vascular tissues/blood vessels were used to explore the molecular mechanisms of atheroprotective effect of endothelial autophagy. Exosomes were isolated and characterized. Immunofluorescence and exosomes coculture experiments were conducted to examine the role of endothelial autophagy in regulating communication between endothelial cells (ECs) and smooth muscle cells (SMCs) via exosome.Results: Endothelial autophagy was inhibited in ECs of thoracic aortas in HFD fed ApoE-/- mice. Furthermore, rapamycin alleviated HFD-triggered atherosclerotic burden and endothelial dysfunction, while endothelial-specific depletion of Atg7 aggravated atherosclerotic burden. miRNA microarray, in vivo and in vitro experiments and human vascular tissues analysis revealed that endothelial autophagy alleviated endothelial dysfunction by downregulating miR-204-5p expression by directly targeting BCL2 to regulate apoptosis in ECs. Moreover, endothelial autophagy decreased miR-204-5p expression by loading this molecule into multivesicular bodies and secreting them out of cells via exosomes, and ECs-derived exosomal miR-204-5p could be transferred to SMCs, antagonized SMCs calcification.Conclusions: Our study first revealed that endothelial autophagy was a critical regulator of atherogenesis by transferring miR-204-5p from ECs to SMCs by exosomes, and endothelial autophagy activation by rapamycin, could alleviate AS in a “one stone hit two birds” manner.

Project description:The cAMP-dependent protein kinase A (PKA) regulates a plethora of cellular functions in health and disease. During angiogenesis, PKA activity in endothelial cells controls the transition from sprouting to vessel maturation and limits tip cell formation independently of Notch signaling. The molecular PKA targets mediating these effects remain unknown. We report a chemical genetics screen identifying endothelial-specific substrates of PKA in human umbilical vein endothelial cells (HUVEC). We identified ATG16L1, a regulator of autophagy, as novel target of PKA. Biochemical validation, mass spectrometry and peptide spot arrays revealed that PKA phosphorylates ATG16L1α at Ser268 and ATG16L1β at Ser269. The phosphorylations drive degradation of ATG16L1 protein. Knocking down PKA or inhibiting its activity increased ATG16L1 protein levels and endothelial autophagy. In vivo genetics and pharmacological experiments demonstrated that autophagy inhibition partially rescues vascular hypersprouting caused by PKA deficiency. We propose that endothelial PKA activity restricts active sprouting by reducing endothelial autophagy through phosphorylation of ATG16L1.

The effect of FUNDC1 was mediated by endothelial cells rather than smooth muscle cells. Further, inducible loss of endothelial Fundc1 in postnatal mice was sufficient to cause PH spontaneously, whereas augmenting endothelial Fundc1 protected against PH before and after the onset of disease. Mechanistically, Fundc1 deficiency impaired basal mitophagy in endothelial cells, leading to accumulation of dysfunctional mitochondria, metabolic reprogramming towards aerobic glycolysis, pseudohypoxia and senescence, likely via a mtROS-HIF2α signaling pathway. Subsequently, Fundc1-deficient endothelial cells increased IGFBP2 secretion that drove pulmonary arterial remodeling to instigate PH. Finally, proof-of-principle in vivo studies showed significant efficacy on PH amelioration by targeting endothelial mitophagy, pseudohypoxia, senescence or IGFBP2.

Project description:Laminar shear stress (LSS) plays a variety of biological roles, one of which is regulation of gene expression in vascular endothelial cells. Prevention and treatment of atherosclerosis by targeting the inflammatory response in vascular endothelial cells has attracted much attention in recent years. In this study, we found that LSS could specifically inhibit the increased expression of intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), cyclooxygenase-2 (COX-2), and matrix metallopeptidase-9 (MMP-9) caused by tumor necrosis factor-α (TNF-α) stimulation in an autophagy-dependent pathway in human aortic endothelial cells (HAECs) and human umbilical vein endothelial cells (HUVECs). Whole-transcriptome sequencing analysis revealed that erythropoietin-producing hepatocyte receptor B2 (EPHB2) was a key gene in response to LSS. Moreover, co-immunoprecipitation (co-IP) assay indicated that LSS could activate autophagy flux by enhancing the EPHB2-mediated nuclear translocation of high mobility group box-1 (HMGB1), which interacts with Beclin-1 (BECN1) and finally leads to autophagy in HAECs. We also found that overexpression of EPHB2 can activate autophagy and exert the same anti-inflammatory effect as LSS. Simultaneously, we identified a novel LSS-sensitive long non-coding RNA (lncRNA), LOC10798635, and constructed an LSS-related LOC107986345/miR-128-3p/EPHB2 regulatory axis. Further research revealed the inhibition of endothelial inflammatory by LSS depends on autophagy activation resulting from the nuclear translocation of HMGB1 via the LOC107986345/miR-128-3p/EPHB2 axis. In conclusion, our study demonstrates for the first time that LSS regulates the expression of EPHB2 in HAECs, and the LOC107986345/miR-128-3p/EPHB2 axis plays a vital role in atherosclerosis development.

Autophagy is a homeostatic process critical for cellular survival, and its malfunction is implicated in myriad human diseases including neurodegeneration. Loss of autophagy contributes to cytotoxicity and tissue degeneration, but the mechanistic understanding of this phenomenon remains elusive. Here we have generated autophagy-deficient human embryonic stem cells (hESCs), from which we have established human neuronal platform to investigate how loss of autophagy affects neuronal survival. ATG5 deficient neurons exhibit basal cytotoxicity accompanied by metabolic defects. Depletion of nicotinamide adenine dinucleotide (NAD) due to hyperactivation of NAD-consuming enzymes is found to trigger cell death via mitochondrial depolarisation in ATG5 deficient neurons. Boosting intracellular NAD levels improve cell viability by restoring mitochondrial bioenergetics and proteostasis in ATG5 deficient neurons. Our findings elucidate a mechanistic link between autophagy deficiency and neuronal cell death that can be targeted for therapeutic interventions in neurodegenerative and lysosomal storage diseases associated with autophagic defect.
