Project description:From the result of the gene expression analyses of human hepatoma cell line, HepG2, a number of genes associated with cell proliferation and DNA repair were distinctively up-regulated by Ag-nanoparticle exposure, suggesting that Ag-nanoparticles might stimulate cell proliferation and DNA damage, which are considered to be mechanisms playing an important role for carcinogenesis and tumor progression. The inductions of these genes involved in cell proliferation were also observed in PS-nanoparticles and Ag2CO3-exposed cells. In addition, the inductions of DNA repair-associated genes were also observed in Ag2CO3-exposure. These results suggest that both “nanoshape” and “silver” can cause the inductions of these gene expression patterns. Furthermore, cysteine, a strong ionic silver ligand partially abolished these gene expressions induced by silver nanoparticles. Ionic silver sourced from Ag-nanoparticles could not fully explain these gene expressions.

Silver nanoparticles affect genes involved in ethylene signaling and stimulate root elongation of white mustard seedlings

Project description:This work was focused on the germination of white mustard in the presence of high concentration of nitrate and silver nanoparticles (Ag NPs). High NO3- reduced primary root length while Ag NPs reversed the growth inhibition. Ag NPs increased root length also under normal conditions. RNA-seq of the seedling roots revealed that the presence of Ag NPs negatively affected the expression of genes involved in the ethylene signaling pathway. NO3- up-regulated auxin-responsive genes from SAUR family and down-regulated several auxin efflux carries, while Ag NPs down-regulated YUC9 associated with auxin biosynthesis and some SAUR genes. Comparison of transcriptional profiles of seedlings germinated on high NO3- with and without Ag NPs indicated that in the presence Ag NPs fewer genes involved in cell wall biogenesis and membrane functions and development were down-regulated and fewer genes involved in salicylic and jasmonic acid signaling were up-regulated. However, both Ag NPs and NO3- increased the expression of genes involved in response to various stresses (e.g. pathogens, hypoxia, oxidative stress) and suberin and cutin biosynthesis. We assume that the effect of Ag NPs was primarily related to the ethylene and auxin signaling pathways.

Chronic Caloric Restriction Preserves Mitochondrial Function in Senescence Without Increasing Mitochondrial Biogenesis

Project description:Caloric restriction (CR) without malnutrition appears to mitigate many detrimental effects of aging, in particular the age-related decline in skeletal muscle mitochondrial function. Although the mechanisms responsible for this protective effect remain unclear, CR is commonly believed to increase mitochondrial biogenesis; a concept that is now demanding closer scrutiny. Here we show that lifelong CR in mice prevents age-related loss of mitochondrial function, measured in isolated mitochondria and permeabilized muscle fibers. We find that these beneficial effects of CR occur without increasing mitochondrial abundance. Furthermore, whole-genome expression profiling and large-scale proteomic surveys revealed expression patterns inconsistent with increased mitochondrial biogenesis. These observations, combined with lower protein synthesis rates support an alternative hypothesis that CR preserves mitochondrial function not by increasing mitochondrial biogenesis, but rather by decreasing mitochondrial oxidant emission, increasing antioxidant scavenging, thereby minimizing oxidative damage to cellular components. Mitochondrial COA7 is a heme-binding protein with disulfide reductase activity, which is involved in the early stages of complex IV assembly

Project description:Cytochrome c oxidase assembly factor 7 (COA7) is a metazoan-specific assembly factor, critical for the biogenesis of mitochondrial complex IV (cytochrome c oxidase). Although mutations in COA7 have been linked to complex IV assembly defects and neurological conditions such as peripheral neuropathy, ataxia and leukoencephalopathy, the precise role COA7 plays in the biogenesis of complex IV is not known. Here we show that the absence of COA7 leads to arrest of the complex IV assembly pathway after the initial step where the COX1 module is built, progression from which requires the incorporation of copper, through the biogenesis of the CuA site. The crystal structure of COA7, determined to 2.4 Å resolution, reveals a ‘banana-shaped’ molecule composed of five helix-turn-helix repeats, tethered by disulfide bonds. We find that in solution, purified COA7 binds heme with micromolar affinity, through axial ligation to the central iron atom by histidine and methionine residues. COA7 interacts transiently with the copper metallochaperones SCO1 and SCO2 that function in copper delivery to COX2 and catalyzes the reduction of disulfide bonds within these proteins, which constitute the protein copper binding sites. We therefore propose that COA7 is a heme-binding disulfide reductase that acts in the biogenesis of the CuA site, that underpins complex IV assembly.
