Project description:To facilitate tumor growth, cancer cells activate pathways that enhance cell proliferation and suppress pathways responsible for cell differentiation. While the pathways involved in metabolic reprogramming and cell cycle progression in tumors have been extensively studied and understood, knowledge regarding the pathways that influence the differentiation status of cancer cells is limited. Our study characterizes a new mechanism regulating the balance between proliferation and differentiation involving the repression of the neuronal-specific transcription factor ARNT2 by the MYC oncogene. We found that MYC represses the transcription of ARNT2 via the activity of the polycomb complex 2. ARNT2 is reduced in glioblastoma and its levels are inversely correlated with patient survival. Using in vitro and in vivo models, we demonstrate that knocking out ARNT2 has no effect on the growth of glioblastoma cells in vitro, but dramatically increases glioblastoma cell growth in vivo, and overexpression of ARNT2 blunts the growth of fully transformed glioblastoma cells in vivo, with no effects in vitro. Mechanistically, we also confirmed that ARNT2 is required for neuronal differentiation including of human pluripotent stem cells. Our findings provide new insights into the complex mechanisms used by oncogenes to limit differentiation in cancer cells and define ARNT2 as a potential tumor suppressor gene in glioblastoma.

P4HB Maintains Wnt-Dependent Stemness in Glioblastoma Stem Cell as a Precision Therapeutic and Diagnostic Marker

Project description:Glioblastoma stem cells (GSCs) are pivotal in the recurrence and drug resistance of Glioblastoma multiforme (GBM). However, precision therapeutic and diagnostic markers for GSCs have not been fully established. Here, using bioinformatics and experimental analysis, we identified P4HB, a protein disulfide isomerase, as a serum marker that maintains stemness in GSCs through the Wnt/β-catenin signaling pathway. Transcriptional silencing of P4HB induces apoptosis and diminishes stem cell-like characteristics in GSCs. Treatments with the chemical CCF624 or the FDA-approved securinine significantly prolonged the survival in patient-derived xenograft mouse models, underscoring P4HB's potential as a therapeutic target and presenting an expedited path to clinical application through drug repurposing. Additionally, elevated P4HB levels in patient serum were found to correlate with disease progression, underscoring its utility as a biomarker and its promise for precision medicine.

Project description:Kruppel-like factor-9 (KLF9), a member of the large KLF transcription factor family, has emerged as a regulator of oncogenesis, cell differentiation and neural development; however, the molecular basis for KLF9M-bM-^@M-^Ys diverse contextual functions remains unclear. This study focuses on the functions of KLF9 in human glioblastoma stem-like cells. We establish for the first time a genome-wide map of KLF9-regulated targets in human glioblastoma stem-like cells, and show that KLF9 functions as a transcriptional repressor and thereby regulates multiple signaling pathways involved in oncogenesis and stem cell regulation. A detailed analysis of one such pathway, integrin signaling, shows that the capacity of KLF9 to inhibit glioblastoma cell stemness and tumorigenicity requires ITGA6 repression. These findings enhance our understanding of the transcriptional networks underlying cancer cell stemness and differentiation, and identify KLF9-regulated molecular targets applicable to cancer therapeutics. Two cell lines were used as biological replicates. Each cell line has one KLF9 induction sample and one control sample.

Feedback circuitry between miR-218 repression and RTK activation in glioblastoma

Project description:Receptor tyrosine kinase pathway signalings plays a central role in the growth and progression of glioblastoma, a highly aggressive group of brain tumors. We recently reported that miR-218 repression, an essentially uniform feature of human GBM, directly promotes RTK hyperactivation by increasing the expression of key positive signaling effectors, including EGFR, PLCr1, PIK3CA and ARAF. However, enhanced RTK signaling is known to activate compensatory inhibitory feedback mechanisms in both normal and cancer cells. We demonstrate here that miR-218 repression in GBM cells also increases the abundance of additional up stream and downstream signaling mediators, including PDGFRa, RSK2, and S6K1, which collectively funciton to alleviate inhibitory RTK feedback regulation. In turn, RTK signaling suppresses miR-218 expression via STAT3, which binds to the miR-218 locus, along with BCLAF1, to repress its expression. These data identify novel interacting feedback loops by which miR-218 repression promotes increased RTK signaling in high-grade gliomas.
