Project description:We investigate the effect of the perturbation of two transcripts involved in target-dependent microRNA degradation (TDMD): NREP is a trigger for miR-29b; SERPINE1 is a trigger for miR-30. We perturbed the genes and monitored the activity of miR-29 or miR-30 analysing the expression changes of their predicted target genes by RNA sequencing.

Project description:We investigate the potential of candidate microRNA responsive elements (MRE, contained in the 3'UTR of candidate transcripts) to trigger target-dependent microRNA degradation (TDMD). To this purpose, we over-expressed the MRE by delivering a lentiviral construct in SUM159PT cells and we profiled microRNAs by small RNA sequencing 7 days after the infection.

Project description:This purpose of this experiment was to investigate the transcriptional differences between C57BL6, TIMP1 and Serpine1 knock-out mice infected with SARS MA15 virus. Overview of Experiment: Groups of 20 week old C57BL6, TIMP1 and Serpine1 (PAI1) knock-out mice were infected with SARS MA15 virus. Infections were done at 10^4 PFU or time-matched mock infected. Time points were 4 and 7 d.p.i. There were 2-4 animals/dose/time point. Lung samples were collected for virus load, transcriptional and proteomics analysis. Weight loss and animal survival were also monitored.

Notch1 Signaling Regulates the Aggressiveness of Differentiated Thyroid Cancer and Inhibits SERPINE1 Expression

Project description:To identify novel down-stream effectors of Notch1 signaling, we have employed cDNA microarray expression profiling as a discovery platform. Human follicualr thyroid carcinoma cell line FTC236 was engineered with tet-on inducible system to establish a stable cell line. The stable cell line FTC236-Notch1 showed robust inducibility with doxycycline, which triggers the expression of Notch1 intracellular domain (NICD). The mRNA samples from FTC236-Notch1 cells with or without doxycline treamtment were analyzed in this assay. Top 50 gene sequences were identified as of the most significance which distinguished between doxycycline treated samples and control samples. Expression of SERPINE1, a down-regulated gene duing Notch1 activation from this signature, was quantified in the same RNA samples by real-time PCR. The protein level was further confirmed by Western blot that restoration of NICD inhibits the expression of SERPINE1.
