Project description:Tissues throughout the human body exert finely-tuned specific functions even when there is considerable overlap in the cellular constituents. Even when the major cellular component of tissues is the same – for example in the case of smooth muscle-rich tissues - discrete phenotypes are evident. Yet, the molecular expression profiles underpinning such specialised functionality is often unclear. This presents a challenge for introducing new medicinal therapies for pathophysiologies: can one target what makes tissue functionality unique without producing unwanted effects on the features that are similar. Such a scenario can be considered for the human uteroplacental unit. Ideally, one would like to target complications of pregnancy like pre-eclampsia (hypertension and proteinuria) or spontaneous preterm birth (early activation of parturition) by targeting particular types of smooth muscle tissues (e.g. arteries versus myometrium) without adversely affecting others. This situation is further complicated by the need to consider maternal and fetoplacental circumstances. Three smooth muscle-rich tissues of importance here are myometrium, maternal uterine (myometrial) arteries and placental arteries. Indeed, each display subtly distinct phenotypes suggesting that distinct molecular signatures may underlie these differences. Hence, we aimed to compare expression profiles of these three tissues to identify the underpinning molecular signatures using multi-omics approaches.

Project description:We used full genome microarrays to profile the full lifetime of the mouse placenta from embryonic day 8.5 (e8.5), at the time of chorioallantoic fusion, until postnatal day 0 (P0). At each stage, the fetal placenta and maternal decidual tissues were dissected and profiled separately Experiment Overall Design: Mouse placentas were obtained from timed pregnant female mice at each timepoint, and fetal tissues were used to confirm embryo staging. Fetal placenta and maternal decidual tissues were dissected and pooled separately for each litter prior to RNA extraction and hybridization on Affymetrix microarrays.

Placenta-specific DMRs maintain methylation across gestation.

Project description:One possible mechanism leading to the apparent polymorphic placenta-specific DMRs would be the failure to maintain allelic methylation during gestation. For a temporal comparison, we performed methylation profiling on first trimester chorionic villus sampling (CVS) and compared it with corresponding samples at term. This revealed that DNA methylation level at placenta-specific DMRs is highly stable between the two points. In addition, to ensure that the methylation profiles were uniform across the placental plate, we determined the placenta-specific DMR profiles from multiple biopsies from the same term placentas. Biopsies collected from the opposite sides of the cord insertion site also showed high correlations, suggesting that methylation does not vary greatly between sampling sites. Finally, we compared placenta samples from dizygotic twins and triplets. As in the other cases, this revealed that the correlations between samples of the same gestations (sharing the same in utero environment and maternal exposures) were also higher than between unrelated samples.
