Project description:As sessile organism, plants evolved a highly complicated signaling system to cope with unfavorable and fluctuating environmental conditions. Rapid and transient Reactive Oxygen Species (ROS) burst is a common response to both biotic and abiotic stresses. Plants exposed with O3 could trigger extracellular similar ROS production through cell wall peroxidases and NPADPH oxidases, resulting in changes in the gene expression and cell death. Whereas ROS induced cell death is not simply due to its toxicity, rather due to interplay with several other signaling pathways, such as salicylic acid (SA), jasmonic acid (JA) and ethylene signaling pathways. Furthermore, the three hormones have both synergistic and antagonistic interactions, where the suppression of JA signaling by SA is the mostly studied. In addition, ethylene promotes cell death while JA has a protective role upon O3 exposure. The role of SA is more complicated; depending on the genetic background it can have either cell death promoting or protecting roles. Hence, a clean system to deliver apoplastic ROS is required to study the role of ROS apart from con-current activation of other signaling pathways. Arabidopsis thaliana offer a convenient system to study apoplastic ROS signaling due to the availability of hormone signaling or biosynthesis mutants including the JA receptor mutant coi1-16 (CORONATINE INSENSITIVE1), the essential ethylene signaling mutant ein2 (ETHYLENE INSENSITIVE2), the SA biosynthesis mutant sid2 (SALICYLIC ACID INDUCTION DEFICIENT2 also known as ISOCHORISMATE SYNTHASE1), and essential regulators in SA/JA/ethylene-induced defense response triple mutant tga2 tga5 tga6 (Clade II TGA transcription factors). Here we used a combination of transcriptome analysis, cell death assays and mutant analysis to systematically quantified the contribution of hormone signaling in relation to apoplastic ROS signaling, identified transcription factors (TFs) involved in ROS regulation and dissected the components involved in defense hormones associated cell death. Abstract:
Recent studies have shown that in addition to the
transcriptional circadian clock, many organisms, including
Arabidopsis, have a circadian redox rhythm driven by the
organism's metabolic activities. It has been hypothesized that
the redox rhythm is linked to the circadian clock, but the
mechanism and the biological significance of this link have
only begun to be investigated. Here we report that the master
immune regulator NPR1 (non-expressor of pathogenesis-related
gene 1) of Arabidopsis is a sensor of the plant's redox state
and regulates transcription of core circadian clock genes even
in the absence of pathogen challenge. Surprisingly, acute
perturbation in the redox status triggered by the immune signal
salicylic acid does not compromise the circadian clock but
rather leads to its reinforcement. Mathematical modelling and
subsequent experiments show that NPR1 reinforces the circadian
clock without changing the period by regulating both the
morning and the evening clock genes. This balanced network
architecture helps plants gate their immune responses towards
the morning and minimize costs on growth at night. Our study
demonstrates how a sensitive redox rhythm interacts with a
robust circadian clock to ensure proper responsiveness to
environmental stimuli without compromising fitness of the
organism.

Project description:Arabidopsis is a host to the fungal powdery mildew pathogen, Erysiphe cichoracearum, and a nonhost to Blumeria graminis f.sp. hordei, the powdery mildew pathogenic on barley. A screen for mutants that allowed increased entry by this inappropriate or nonhost pathogen on Arabidopsis led to the identification of PEN3. While pen3 mutants permitted both increased penetration and increased hyphal growth by B. g. hordei, they were unexpectedly resistant to E. cichoracearum. This resistance was correlated with the appearance of chlorotic patches and was salicylic acid-dependent. Consistent with this observation, microarray analysis revealed that the salicylic acid defense pathway was hyper-induced in pen3 relative to wild type following inoculation with either E. cichoracearum or B. g. hordei. The pen3 phenotypes result from a loss of function of AtPDR8, a ubiquitously and highly expressed ATP binding cassette transporter. PEN3 protein tagged with green fluorescent protein localized to the plasma membrane in uninfected cells. In infected leaves, the protein concentrated to high levels at infection sites and surrounded fungal penetration pegs. We hypothesize that PEN3 may be involved in exporting toxic substrates to sites of infection and that accumulation of these substrates intracellularly in the pen3 mutant may secondarily activate the salicylic acid pathway. Experiment Overall Design: Three week-old wild-type Col and mutant pen3 Arabidopsis thaliana plants were inoculated with Erysiphe cichoracearum, Blumeria graminis hordei, or not inoculated. 1 day post inoculation 16 rosettes were harvested per replicate. 4 replicates were perfomerd per treatment.

Project description:Functions of AtVDAC1 and its partner p26 in Arabidopsis thaliana. We previously showed that knock-out mutants impaired in AtVdac1 and p26 (AtVdac1 protein partner) are hypersensitive to salicylic acid: the root growth is more inhibited in response to 30 uM salicylic acid (SA) compared to the wild type. We want to know now which genes are up- or down-regulated in the mutants in control and SA conditions.<br> We want to compare the RNAm contents in the wild type, vdac1 and p26 mutants and, vdac1-p26 double mutant. One-week old plantlets were transferred for 2 hours on a fresh medium containing either 0 or 30uM salicylic acid.<br> Arabidopsis lines (WT, vdac1.1, p26.1, vdac1.1-p26.1) were grown in vertical position on ABIS medium (Lanquar et al., 2006). One week-old seedlings were transferred on a fresh ABIS medium containing either 0 or 30 uM salicylic acid. After 2 hours, the all seedlings were harvested. Three replicates were done; total RNAs from these 3 experiments were mixed together for the transcriptomic analysis.
