Project description:Gene expression on peripheral blood mononuclear cells (PBMC) from SPARKS CHARMS juvenile idiopathic arthritis (JIA) cohort pre and post methotrexate therapy. This is the first study to our knowledge, to evaluate gene expression profiles in children with JIA before and after MTX, and to analyze genetic variation in differentially expressed genes. We have identified a gene, which may contribute to genetic variability in MTX response in JIA. PBMC were isolated from blood samples taken from 11 patients with JIA pre methotrexate therapy and at 6 months into therapy

Restoration of CD56dim NK Cells by Methotrexate Contributes to the Treatment of Juvenile Idiopathic Arthritis

Project description:Juvenile idiopathic arthritis (JIA) is the most common chronic rheumatic disease in childhood. To elucidate the mechanism of methotrexate (MTX) treatment-induced JIA clinical remission, we conducted single-cell RNA sequencing (scRNA-seq) analysis of peripheral blood mononuclear cells (PBMCs) obtained from a female JIA patient before and after MTX treatment. Besides, we integrated our data with public bulk RNA-seq data. We found that the proportion of the CD56dim NK subsets was restored after MTX treatment, suggesting that CD56dim NK subsets are mainly involved in MTX treatment-induced JIA clinical remission. Differential expression analysis showed that the NK cell-mediated cytotoxicity pathway was riched in CD56dim NK subsets. Furthermore, bulk RNA-seq results showed that MTX-treated NK-92 cell lines were also enriched for NK cell-mediated cytotoxicity pathways. Our data highlight the importance of CD56dim NK subsets in MTX treatment-induced JIA clinical remission.

Proteome characterization of synovial fluid polarized healthy monocytes

Project description:This project involves proteome characterization of healthy monocytes polarized (activated) with synovial fluid (SF) versus serum from patients with juvenile idiopathic arthritis (JIA). The objective is to investigate the influence of SF on monocytes at the proteome level, using serum polarized monocytes as a control. Monocytes were isolated from peripheral blood mononuclear cells of healthy donors following negative selection and were subsequently polarized overnight with 20% of a pool of either serum or SF. They were subsequently detached and used for proteome analysis.

The predictive biomarkers of TNFα-blocking agent responsiveness are specific for adalimumab and etanercept

Project description:The objective of this study was to identify specific gene expression profiles able to predict the response of rheumatoid arthritis patients treated with methotrexate (MTX)/adalimumab (ADA) or MTX/etanercept (ETA). Twenty RA patients were received subcutaneously Adalimumab (40 mg each other week) and eleven RA patients were received Etanercept (50 mg per week). The drug efficacy was evaluated with the DAS28 score after 3 months of treatment according to the EULAR response criteria. A blood sample was carried out in patients just before the first injection of treatment in order to isolate peripheral blood mononuclear cells (PBMC) and extract total RNA.

Global profiling of microRNAs in peripheral blood and synovial fluid mononuclear cells of patients with Enthesitis Related Arthritis

Project description:Enthesitis related arthritis (ERA) is the most common category of juvenile idiopathic arthritis in Asia. MicroRNA dysregulation has been shown in chronic arthritides however there is no data in ERA. MicroRNA dysregulation in ERA may help understand the pathogenesis of disease. Thus we determined the difference in global miRNA expression in peripheral blood mononuclear cells (PBMCs) of patients with ERA (n=8) as compared to healthy controls (HC) (n=8) and further to understand the changes at the site of inflammation, the PBMCs profile was compared with the synovial fluid mononuclear cells (SFMCs) (n=8) of patients with ERA.
