Project description:We report the transcriptional profiling of Drosophila melanogaster gonadectomized adults following RNAi knockdown of grappa (gpp), lilliputian (lilli), or Suppressor of Triplolethal [Su(Tpl)], as well as induction of Dominant Negative (DN) allele of Cyclin-dependent kinase 9 (Cdk9), or ectopic expression of stand still (stil), with a three-component temperature sensitive system. We also included the transcriptional profiling of testes and ovaries of gppRNAi, lilliRNAi, and Su(Tpl)RNAi flies under permissive temperature. All data includes induced and non-induced sham controls.

Transcriptome changes in Drosophila HB10 cells in response to a temperature shift from 24 to 14°C

Project description:Cells in ectothermic organisms often maintain homeostatic function over a considerable range of ambient temperatures. However, as temperature has pronounced effects on all biological processes, but not necessarily in a uniform manner on each of the myriad of distinct processes, cellular acclimation to distinct temperatures is predicted to involve complex regulation. Initially, to assess the effects of temperature change within the readily tolerated temperature range on the transcriptome, we have performed expression profiling with S2R+ cells, a hemocyte-like cell line derived from the ectothermic organism Drosophila melanogaster. To adress the cell-type specificity of the transcriptome change induced by a temperature downshift from the optimal temperature to a temperature at the lower end of the readily tolerated range, we have performed an anologous experiment with cultured Drosophila HB10 cells. These cells with similarities to adult muscle cell progenitors have recently been derived from Drosophila embryos with the method (Simcox et al. 2008. PLoS Genetics 4, e1000142) involving expression of activated ras using Act5C-GAL4 in combination with UASt-ras85D[V12].

Wind-responsive changes in poplar leaves of different developmental stages

Project description:A developmental series of wind-treated Populus leaf tissue was subjected to array analyses in order to address the issue of age-dependent responsiveness to environmental changes. The following developmental stages were defined for the experiment: Y – “youngest leaf” including the shoot tip = smallest fully enrolled leaf; E – “expanded leaf” = oldest leaf that had not reached full leaf thickness; M – “mature leaf” = 5th leaf below E = has reached full leaf expansion and full leaf thickness; O – “old leaf” = 5th leaf below E. Temperature dependent gene expression levels at 28°C and 37°C for Pseudomonas aeruginosa PAO1

Project description:To further understand the gene expression characteristics of Pseudomonas aeruginosa PAO1, we have applied whole genome microarray expression profiling as a discovery platform to specify the temperature dependent expression of PAO1 genome at soil and human body temperature. We selected 28°C as temperature representative of the soil niche and 37°C for human body. The results from the temperature dependent transcriptome analysis are consistent to our previous published data that the phzM, ptsP and lasI genes expression is upregulated at 37°C [11]. The comparison analysis of the M18 genome expressional profiles at 28°C and 37°C indicated a total of 596 genes expressed in a temperature dependent manner over two fold.

Effect of histone H2Av depletion on temperature dependence of the Drosophila S2R+ cell transcriptome

Project description:The histone H2A variant H2A.Z has a crucial role in the regulation of temperature-dependent gene expression in the plant Arabidopsis thaliana. To evaluate whether the Drosophila homolog histone H2Av has a similar role in the control of the transcriptional response to temperature change, we depleted this histone variant by RNA interference in S2R+ cells and analyzed the effect on the transcriptome after temperature shifts to different temperatures (14, 25 and 30°C). For control, we performed analogous experiments using treatment with lacZ dsRNA instead of His2Av dsRNA.

Project description:To further understand the gene expression characteristics of originating biocontrol strain Pseudomonas aeruginosa M18, we have applied whole genome microarray expression profiling as a discovery platform to to specify the temperature dependent expression of M18 genome at rhizosphere and human body temperature. We selected 28°C as temperature representative of the rhizosphere niches and 37°C for human body. The results from the temperature dependent transcriptome analysis are consistent to our previous published data that the phzM, ptsP and lasI genes expression is upregulated at 37°C. The comparison analysis of the M18 genome expressional profiles at 28°C and 37°C indicated a total of 605 gene expressed in a temperature dependent manner over about two fold at 28°C compared that at 37°C, covering 10.6% genes in M18 whole genome.
