Intrinsic differences between oral and skin keratinocytes

Project description:The morphology and the behavior of skin and oral tissue keratinocytes are different. One significant dissimilarity between the two sites is the response to injury. Oral and skin keratinocytes have intrinsic differences in the response to injury and such differences are reflected in gene expression profiles. We used microarrays to investigate differences in global gene expression patterns between baseline skin and oral epithelium sheets without their underlying connective tissue. Paired skin and oral epithelium was separated from the dermis for RNA extraction and hybridization on Affymetrix microarrays. Skin epidermal tissues were obtained from the tail of mice and oral epidermal tissues were obtained from the hard palate. Enzymatically isolated epithelium was used for analysis.

Project description:Keratinocytes are the major constituent of epithelial cells at mucosal surfaces and skin, which cover organs, internal cavities and the body. Traditionally, keratinocytes have been considered as an inert component of the multilayered epithelium to protect the subepithelial compartments from the pathogenic microorganisms, toxic stimuli and physical trauma. However, accumulated researches of the airway, gastrointestinal tract and skin have demonstrated that keratinocytes function in the development of the immune system, promotion of pathologic inflammation and even impose diverse decisions on immune cells. We used microarrays to detail the global gene expression of oral keratinocyte during oral adaptive immune response. Oral keratinocyte were collected at three time points (control, 48h and 96h) in oral adaptive immune response for RNA extraction and hybridization on Affymetrix microarrays

MicroRNA differential expression in skin and oral mucosal epithelium

Project description:While skin and oral mucosa share many morphological similarities, oral mucosal wounds heal more rapidly than skin wounds. Epithelial cells from oral mucosa exhibit increased migratory and proliferative capacities when compared to cells from skin, suggesting that the improved repair of mucosa may involve intrinsic differences in epithelial cells. This is an exploratory experiment to define the differential microRNA expression of baseline unwounded skin and oral mucosa epithelium.

Developmental plasticity of epithelial stem cells in tooth and taste bud renewal

Project description:In Cichlids, replacement teeth (RT) share a continuous band of epithelium with adjacent taste buds (TB) and both organs co-express stem cell factors in subsets of label-retaining cells. In the mouse and other mammals, the tongue inter molar eminence (IE) oral papillae of Follistatin (Fat, BMP antagonist) mutants exhibited dysmorphic invagination. By using NGS-derived transcriptome profiling (RNAseq) analysis, we compared differential gene expression in the mouse tongue tissue in Follistatin (Fst) mutants mice with the wild type controls. Our results demonstrated ectopic expression of dental markers in tongue IE indicating that vertebrate oral epithelium retains inherent plasticity to form tooth and taste-like cell types mediated by BMP, therefore revealed under appreciated epithelial cell populations with promising potential in bioengineering and dental therapeutics.
