Response of the mature nodules of the Medicago truncatula/sinorhizobium medicae md4 symbiotic association to systemic signaling associated to plant water deficit (PEG treatment)

Project description:The experiment aims to identify the response of the mature nodules and denodulated roots of Medicago truncatula /Sinorhizobium medicae md4 to the systemic signaling of plant water deficit. The root systems of nodulated symbiotic plants were divided into two compartments. The N source of N2 fixing plants was exclusively the air. To apply the water stress, we treated half root system with PEG while the other was kept on control conditions. To characterize the systemic responses of the transcriptome to the PEG treatment , nodules and roots of the non-treated half root system were harvested and total RNA isolated for RNAseq analysis. We performed a time-course (6 hours, 1-, 3- and 5-days treatment) .

To reduce the plant nitrogen (N) supply (N-deficient conditions), we flushed one half root system with Ar/O2 (80/20 v/v) while the other half root system was kept on control conditions (Air). To characterize the systemic responses of the transcriptome to plant N deficit , nodules and roots of the non-treated half root system were harvested and total RNA isolated for RNAseq analysis. We performed a time-course (6 hours, 1-, 3- and 5-days treatment).

Transcriptomic changes in Medicago truncatula and Lotus japonicus root nodules during drought stress

Project description:Drought is one of the major environmental factors limiting biomass and seed yield production in agriculture. In this research we focused on plants from Fabaceae family, which have a unique ability for establishment of symbiosis with nitrogen-fixing bacteria, and are relatively susceptible to water limitation. We present the changes in nitrogenase activity and global gene expression occurring in Medicago truncatula and Lotus japonicus root nodules during water deficit. Our results prove a decrease in the efficiency of nitrogen fixation as well as extensive changes in plant and bacterial transcriptomes shortly after watering cessation. We show for the first time that not only symbiotic plant component, but also Sinorhizobium meliloti and Mesorhizobium loti bacteria residing in the root nodules of M. truncatula and L. japonicus, respectively, adjust their gene expression in response to water shortage. Although our results demonstrate that both M. truncatula and L. japonicus root nodules are susceptible to water deprivation, they indicate significant differences in plant and bacterial response to drought between tested species, which may be related to various type of root nodules formed by these species.

Transcriptom analysis of the interaction Medicago truncatula – Ralstonia solanacearum

Project description:affy_ralstonia_medicago - Ralstonia solanacearum is the causal agent of the devastating bacterial wilt disease. Its infection process was studied with an in vitro inoculation procedure on intact roots of Medicago truncatula. The pathosystem involved susceptible A17 and resistant F83005.5 M truncatula lines infected with the pathogenic strain GMI1000. The mutant A17 line, Sickle, which showed a resistant phenotype was also part of the experiment. To identify host signaling pathway triggered by R. solanacearum infection with a focus on the involvment of ethylene, we used the Medicago Affymetrix array to monitore the expression profiles and the molecular process associated with initial symptoms development (12hpi) and colonization (72hpi). In order to maximize chances to observe differential gene expression, RNA samples were extracted from the root infection zone (root tips) -Three Medicago truncatula lines, A17, F83005.5 and sickle were inoculated with GMI1000 Ralstonai solanacearum strain (107 cfu/ml). RNA were extracted from root extremities (1 cm above the root tip) at time 0, 12h and 72h post inoculation. Three biological repeats were conducted

Project description:affy_med_2011_09: In natural ecosystems most vascular plants develop symbiosis with arbuscular mycorrhizal (AM) fungi which help them acquire nutrients such as phosphorus (P) and nitrogen (N). P has long been known to control AM symbiosis which takes place only when P is limiting. For N, however, its role in controlling mycorrhization is less clear. We have chosen the model plant Medicago truncatula to analyze the impact of P limitation and both P and N limitation on Medicago root transcriptome in response to the AM fungus Rhizophagus irregularis (formerly Glomus intraradices (BEG141)). These analyses may help us uncover signaling events involved in the interaction between these symbionts as well as genes encoding transporters potentially important for nutrient exchanges in these conditions. --We will compare the root transcriptome of Medicago truncatula plants inoculated with Rhizophagus irregularis to that of non-inoculated plants grown under P limitation (or both P and N limitation) after 4 weeks of culture

Ethylene signaling is important for isoflavonoid mediated resistance to Rhizoctonia solani in roots of Medicago truncatula

Project description:The root-infecting necrotrophic fungal pathogen Rhizoctonia solani causes significant disease to all the world’s major food crops. As a model for pathogenesis of legumes we have examined the interaction of R. solani AG8 with Medicago truncatula. RNAseq analysis of the moderately resistant M. truncatula accession A17 and highly susceptible sickle (skl) mutant (defective in ethylene sensing) identified major transcriptional reprogramming early in A17. Responses specific to A17 included components of ethylene signalling, numerous class IX ERF transcription factor family members, reactive oxygen species metabolism and consistent up-regulation of the isoflavonoid biosynthesis pathway. Mass-spectrometry revealed accumulation of the isoflavonoid related compounds liquiritigenin, formononetin, medicarpin and biochanin A in A17. Over-expression of an isoflavone synthase (IFS) in M. truncatula roots increased isoflavonoid accumulation and resistance to R. solani. Addition of exogenous medicarpin suggested this phytoalexin may be one of several isoflavonoids required to contribute to resistance to R. solani. Together these results provide evidence for the role of ethylene-mediated accumulation of isoflavonoids during defence against root pathogens in legumes.

Gene expression during early stages of arbuscular mycorrhizal root formation in Medicago truncatula.

Project description:Arbuscular mycorrhiza (AM) interactions between plants and Glomeromycota fungi primarily support phosphate aquisition of most terrestrial plant species. To unravel gene expression during early stages of Medicago truncatula root colonization by AM fungi, we used genome-wide transcriptome profiling based on mycorrhizal root fragments enriched for early fungal infection stages. We used Medicago GeneChips to detail the global programme of gene expression in response to early stages of colonization by arbuscular mycorrhizal fungi and identified genes differentially expressed during these early stages.
