Transcription profiling by array of Drosophila embryonic chordotonal neurons expressing the cato GFP reporter gene construct

Project description:Further to a previous transcription profiling experiment using a GFP reporter gene construct expressing the Drosophila proneural gene atonal (Cachero et al. 2011), a later expressing transcription factor and downstream target of ato, cato (cousin of ato) was explored as the GFP reporter in this experiment. cato-GFP expressing cells were isolated from 10h45min and 11h45min old embryos. GFP+ and GFP- cells were compared on Affymetrix Drosophila version 2.0 chips.

Project description:Expression of mutant lamins in human muscle causes muscular dystrophy. We have generated a drosophila model that expresses mutant lamins, modeled after those that cause disease in humans. We used affymetrix microarrays to determine the global changes in gene expression caused by mutant lamins. Dissected 3rd instar larval body wall from transgenic Drosophila expressing either a full length wildtype Lamin C, an N-terminal Lamin C truncation of the first 42 amino acids or a single amino acid substitution in Lamin C at position 489 from a G residue to a V residue under control of the Gal4-UAS. RNA was extracted from 3rd instar larval body wall expressing these mutant Lamin C transgenes and microarray analysis using Affymetrix microarrays.

Transcription profiling of Drosophila expressing four mutant forms of MYOC (R342K, Q368X, D380N and K423E) to model Myocilin-Associated Glaucoma

Project description:We have established Drosophila melanogaster as a model system for ocular hypertension by expressing wild-type human myocilin (MYOC) in the Drosophila eye. Here, we have created transgenic flies that express four clinically relevant mutant forms of MYOC (R342K, Q368X, D380N and K423E) in their eyes using the gmr-Gal4/UAS binary system. We compare and identify human glaucoma candidate genes based on the transcription profiles of flies that express wt-MYOC or mutant-MYOCs.

Project description:To determine the extent of alternative 3' end cleavage during Drosophila spermatogenesis, we performed 3' end sequencing of Drosophila testes from flies at different time points post heat shock (pHS) that were mutant for bam and also had a heat shock inducible transgene expressing Bam. We then performed Polysome profiling of testes at 24, 48 and 72 hours PHS and created 3' end sequencing libraries from the free, 40S, 60s, 80S, 2-3 ribosomes, and 4+ ribosomes fractions for each dataset
