Project description:Genome-Scale draft model for Human Peripheral Blood Mononuclear Cells (PBMCs). A GEM for PBMCs was developed by applying the INIT algorithm on Human Metabolic Reconstruction (HMR 2.0) as a template model. GEMs were contextualised/ constrained for different conditions using expression datasets. The gene/transcript expression data obtained from PBMCs of Type 1 Diabetes progressors, non-progressors, and healthy controls were employed to score each reaction of HMR 2.0. Gene expression in circulating mononuclear cells after exposure to ultrafine carbon particles

Project description:Peripheral blood samples were collected before (0 hour) and at 24 hours after exposure from healthy subjects who participated in previous controlled exposures to ultrafine carbon particles (UFP, 50 microg/m3) or filtered air (FA)(n = 3 each). The exposure time was 2 hours. RNA from mononuclear cell fraction (>85% lymphocytes) was extracted, amplified and hybridized to Affymetrix HU133 plus 2 microarrays. We used microarray to explore significantly altered genes after ultrafine carbon particle exposure. Each subject was exposed to filtered air or ultrafine carbon particles. Two peripheral blood samples (pre- and post-exposure) were taken. Mononuclear cells were isolated for gene expression analysis.

Project description:Under certain circumstances, humans can be exposed to ionizing radiation from heavy ions. Examples are cancer patients undergoing particle therapy (e.g. carbon ions) or astronauts in deep Space missions (iron ions which contribute for a large fraction to the possible health effects of cosmic radiation because of their high ionisation power). Understanding the differences in the cellular response to low- and high-LET radiation is important in order to adequately model risk estimates based on extrapolations from low-LET exposures. To address this need, we compared the transcriptional profiles of freshly isolated peripheral blood mononuclear cells after exposure to 1 Gy of X-rays, iron ions or carbon ions.
