mRNA Sequencing analysis of MED30 deleted mice.

Project description:To study the transcriptome in developing heart controlled by the intact Mediator complex. RNA-seq analysis was performed on RNA extracted from hearts of Med30 cKO and control littermates at E10.5. From this analysis, 10,637 genes were expressed in both mutant and control cardiomyocytes. Of these, 31 genes were significantly upregulated, and 45 genes were significantly downregulated (log2fold > 1, adjusted p value < 0.05) in mutant cardiomyocyte relative to controls. To identify genes responsible for the rapidly developed phenotype in Med30 icKO mice, we performed RNA-seq analysis of RNA extracted from adult cardiomyocytes isolated from Med30 icKO and control mice at 4-weeks post tamoxifen injection. From this analysis, 9517 genes were detected in both mutant and control cardiomyocytes. Of these, 1153 and 1509 were significantly (log2fold > 1, adjust p value < 0.05) up- or downregulated, respectively, in mutant cardiomyocyte relative to controls

Genes regulated in mouse liver by expressing ectopically hURI in hepatocytes

Project description:RNA-seq was performed using the RNA extracted from the bottom half of right lobe of mouse livers. Mice fall into two groups, mutant group which express ectopic hURI and their control littermates which do not express hURI. Two time points were considered in the study, 1-week-old mice, expressing hURI since 1 week (n =3, 4 for control and mutant, respectively) and 8-week-old mice expressing hURI since 8 week (n= 4, 3 for control and mutant, respectively), as hURI is expressed since conception.

CNS and peripheral metabolomics of calorie restriction in a mouse model of Alzheimer’s disease (part II)

Project description:This pilot metabolomic study will evaluate cecal specimens from an established mouse model of AD, the tq2576 mouse model of cerebral amyloid overexpression, in comparison to their non-transgenic (ntg) littermates. These animals were either on a CR or ad libitum (AL) diet, and specimens were collected at two time points (5 and 15 months of age). Tissue from this cohorts of mice have already undergone microbiome analysis, and await coordinated brain and peripheral tissue assessments. Future analysis will include metabolomics, RNA-seq, and microarray data to assess the gut-brain microbiome system in neurodegenerative disorders.

Identification of Sox9-Regulated Pathways During Early Pancreas Organogenesis

Project description:Sox9 target genes were identified by comparing gene expression in Sox9-ablated versus wild-type pancreata using microarray analysis. Sox9 was conditionally ablated in the developing pancreas via recombination of a Sox9-flox allele (Kist et al., 2002) using the Pdx1-Cre transgene (Gu et al., 2002). Total RNA was isolated and pooled from dorsal pancreatic epithelia of e12.5 Sox9fl/fl; Pdx1-Cre (mutant) versus Sox9fl/fl (wild-type) littermates for three biological replicates.
