ABSTRACT: We have examined the role of RNA in the integrity of nuclear and cytoplasmic membrane-less organelles composed of RNA and protein by activating RNase L in the cytoplasm or nucleus. We degraded intracellular RNA by activating endogenous RNase L or RNase L linked to a nuclear localization signal in human A459 cells with the dsRNA mimic poly(I:C) and then examined the impact of RNA loss on several RNP granules by immunofluorescence.
