Transcription profiling of rat Schwann cells in response to treatment with NRG and/or IGF

ABSTRACT: Comparison of the changes in mitochondrial gene expression of cells in which extracellular growth factors and/or mitogens have been added. Experiment Overall Design: Primary Rat Schwann cells were arrested and synchronized with aphidicolin for 24 hours in 3% FCS containing medium. Cells where then transferred to defined conditions (no factors) with aphidocolin for another 24 hours before appropriate factors were applied for 15 hours prior to cell collection.

Project description:In attempt to understand M.leprae interaction with the human host, Applied Biosystems microarrays containing 30,865 probles were used to identify modulated genes in primary human Schwann Cells (SC) infected with live M. leprae at two early time points, 24 and 48 hours. A total of 4 independent experimental samples were prepared which were hybridized to two replicate microarrays each.The four experimental samples included both uninfected and M. leprae infected Schwann cells at both 24 and 48 hours.

Project description:Comparison of the changes in the gene expression profile of cells in which there is an activation of the Raf oncogene in the absence of any other extracellular signals Experiment Overall Design: Primary rat Schwann cells containing an inducible RafER allele were plated on poly-L-lysine plus fibronectin coated dishes on DMEM + 3% FCS containing medium. Next day cells were transferred to minimal medium (DMEM containing 100 Î¼g ml-1 transferrin, 100 Î¼g ml-1 bovine serum albumin, 16.1 Î¼g ml-1 putrescine, 39 ng ml-1 selenium) for 24 h. Cells were then treated with either the vehicle (EtOH) or with Tamoxifen for 24 h. Three independent experiments were performed.

Project description:In an attempt to understand M. leprae interaction with the human host, Stanford Genomics HEEBO Arrays were use to identify modulated genes in primary human Schwann cells infected with live M. leprae at a MOI of 100:1 for 24 hours. Dual channel competitive hybridizations between M. leprae infected and non infected Schwann cells including 3 independent biological replicates and a technical replicate in the form of dye swap.

Transcription profiling of human Schwann cells after extended exposure to heregulin and forskolin

Project description:Schwann cells, expanded in number by exposure to growth factors in vitro, could be useful in nervous system repair. Our previous results suggest that long term exposure to heregulin and forskolin changes the functional properties of the human Schwann cells, including the ability to myelinate axons after transplantation. Here, we propose to determine the molecular changes in the Schwann cells that occur as a result of extended growth with mitogenic factors. We believe that the information obtained in these studies will provide clues about mechanisms underlying the already observed changes in function. This information will aid in the prediction of the safety and efficacy of neural repair approaches that use cultured, expanded Schwann cells. Finally this data may provide clues into the mechanisms underlying normal human Schwann cell function.,To use gene array analysis to compare gene expression profiles in early and late passage human Schwann cells exposed to the growth factors heregulin and forskolin.,Observed changes in the function of human Schwann cells, including their capacity for growth and differentiation, after prolonged exposure to heregulin and forskolin, are caused by changes in the gene expression profiles in these cells.
