Neuregulin/ErbB2-induced kinase signaling provides essential survival and protection clues for functional integrity of the adult heart and skeletal muscle. Wild type and Neuregulin 4 (Nrg4) knockout mouse livers

Project description:Nrg4 is a brown fat-enriched adipokine that binds to the liver and preserves metabolic homeostasis during chronic energy excess. Adipose tissue Nrg4 expression is reduced in mouse and human obesity. Nrg4 deficient mice develp more severe insulin resistance and hepatic fat accumulation upon high-fat diet feeding. We used microarrays to elucidate the metabolic pathways that are regulated by Nrg4 in the liver. Wild type and Neuregulin 4 (Nrg4) knockout mice were fed high-fat diet (Research Diets D12492) for eight weeks. Tissues were harvested under ad lib condition for RNA isolation and microarray hybridization using Affymetrix Mouse MG-430 PM Strip arrays. Three independent pools of WT and KO mouse liver RNA were included in the study. Neuregulin 1 type III reduces severity in a mouse model of Congenital Hypomyelination Neuropathy

Project description:Purpose: We hypothesised that neuregulin 1 type III would reverse CHN. Using a faithful mouse model of CHN, we rescued the myelination defects by activation of Nrg1 type III. Unexpectedly, rescue did not depend on upregulation of Egr2 or major myelin genes. Therefore we asked whether we could identify any transcriptomic signature of the canonical pathways downstream of Nrg1 type III that could explain the increase in myelination in the sciatic nerve. Methods: Sciatic nerve mRNA profiles of 30-day-old wild-type (WT), P0 mutant (Q215X/+), trangenic HA-NRG1 type III, and HA-NRG1 type III:Q215X mice were generated by deep sequencing, in duplicate, using Illumina HiSeq2500 . The sequence reads that passed quality filters were aligned to the mouse reference genome (mm10) with the transcript annotation (UCSC mm10 annotation download from TopHat website) using TopHat (v2.0.9). Quantification for the gene expression and differential expression analysis were done by the software Cufflinks (v2.2.0). Results: A comprehensive transcriptomic analysis of signals downstream of neuregulin 1 type III in rescued CHN nerves revealed imbalanced activation of MAPK/ERK signals and minor myelin genes, such as Pmp2 and Omg. Conclusions: Our findings demonstrate that although augmented Nrg1 type III activity modifies the composition of myelin, it still promotes thicker myelin and ameliorates nerve conduction velocity in a mouse model of CHN. Controlled increase of Nrg1 type III activity may be a therapeutic strategy in some hypomyelinating neuropathies.

Responses of tobacco BY-2 cells to boron deprivation

Project description:Boron (B) is an essential micronutrient for vascular plants. Boron works to build the cell wall structure by forming borate ester cross-links with pectin, and the deficiency of this element brings about oxidative damage and cell death. However, the process via which B deficiency leads to such serious damage has not been well understood. We investigated the temporal changes in metabolism of B-deprived cultured tobacco (Nicotiana tabacum) BY-2 cells by analyzing their gene expression using cDNA microarray. Three-day-old cells were deprived of B by washing with and transferred to B-free growth media ("-B" cells). Cells similarly treated with standard growth media were employed as a control for the effects of mechanical stress ("+B cells"). Cells were sampled at 1, 3, 6, 12 and 36 h after transfer to the B-free or standard media. Cells without washing treatment were also included in the analysis ("before-treatment cells"). Two independent sets of samples were analyzed (replicate 1 and 2).

The Neuregulin 1-HER axis as a key mediator of hyperglycemic memory effects in breast cancer

Project description:Poor outcomes in diabetic patients are observed across a range of human tumors, suggesting that cancer cells develop unique characteristics under diabetic conditions. Cancer cells exposed to hyperglycemic insults acquire permanent aggressive traits of tumor growth, even after a return to euglycemic conditions. Comparative genome-wide mapping of hyperglycemia-specific open chromatin regions and concomitant mRNA expression profiling revealed that neuregulin-1 gene, an established endogenous ligand for the HER3 receptor, is activated through a putative distal enhancer. Our findings highlight the targeted inhibition of NRG1-HER3 pathways as a potential target for the treatment breast cancer patients with associated diabetes

Project description:To determine the effect of prohibitin overexpression on global gene expression in Caco2-BBE intestinal epithelial cells. 4 individual wells of Caco2-BBE cells, passage 41, were transfected with either empty vector (pcDNA4) or prohibitin/pcDNA4 for 72 hours. Total RNA isolated from 4 wells of cells/per treatment were pooled together for labeling and hybridization purposes.
