Transcription profiling of mouse muscle and bone marrow SP and MP cells

ABSTRACT: This study identified gene expression of Side Population (SP) and Main Population (MP) cells, isolated from adult murine skeletal muscle and Bone Marrow. Five different preparations of muscle SP, muscle MP, Bone marrw SP and Bone marrow MP cells were used as replicates. Side Population (SP) cells, isolated from murine adult bone marrow (BM) based on the exclusion of the DNA dye Hoechst 33342, exhibit potent hematopoietic stem cell (HSC) activity when compared to Main Population (MP) cells. A novel five-gene signature predicts overall and recurrence-free survival in NSCLC

Project description:Earlier studies had shown that side population cells isolated from established non-small cell lung cancer (NSCLC) cell lines exhibit cancer stem cell properties. Microarray data from side population (SP) and main population (MP) cells isolated from 4 NSCLC lines (A549, H1650, H460, H1975) were used to examine gene expression profiles associated with stemness. Total RNA extracted from SP and MP samples were used to generate cRNA targets, which were hybridized to Human Genome U133 Plus 2.0 probe arrays. Raw data was processed and the mean center expression level for each gene was determined. Four cell lines (A549, H1650, H460, H1975), each having 1 SP and 1 MP sample.

Transcriptome analysis of blast Side and Main populations cells in patients with acute myeloid leukemia

Project description:Acute myeloid leukemia (AML) represent a series of hemopathies characterized by the clonal expansion in bone marrow and blood of immature myeloid cells blocked in differentiation, called blasts. Despite significant supportive care progresses, few medical discoveries radically changed LAM prognosis (5 year survival rate of 30%). An aggressive conventional chemotherapy is used to kill AML cells. However, many patients relapse mainly due to the persistence of rare AML cells able to re-initiate the disease. Quiescence and protection against environmental stresses (chemoresistance) are major characteristics of these cells. Drug efflux leading to chemoresistance is observed by Side Population (SP) detection through ABC transporter activation. So, AML blasts presenting SP functionality could represent a chemoresistant population. This SP phenotype is essentially acquired by circulating AML blasts after contact with medullar mesenchymal stromal cells (MSC). A transcriptome analysis of AML blasts sorted according to their SP functionality or not (Non SP cells or Main Population, ie MP) after co-culture on MSCs was realized.
