Transcription profiling of Populus tremuloides cell suspension cultures to methyl jasmonate feeding

ABSTRACT: Gene expression response of Populus cell cultures subjected to methyl jasmonate feeding was analyzed using the Affymetrix poplar genome microarrays. Experiment Overall Design: Populus tremuloides suspension cells wereÂ treated with methyl jasmonate (MJ) in DMSO orÂ with DMSO alone (C).Â Cells were harvested after 48 hours.Â Two biological replicates were obtained for each condition.

Project description:To better understand the role of phenylpropanoid pathway perturbation in plant metabolism, Populus tremuloides cell cultures previously established from leaf mesophyll tissue were fed with methyl jasmonate, alpha-aminooxy-beta-phenylpropionic acid (AOPP), or both approximately 5 days after transfer to fresh medium. Methyl jasmonate is an elicitor that activates a suite of defense responses including phenylpropanoid metabolism. In contrast, AOPP acts as an inhibitor of phenylalanine ammonia lyase (PAL). Samples were harvested 48 hrs after initiation of the experiment. Total RNA was extracted and gene expression measured using Affymetrix poplar genome microarrays.

Gene expression response of Populus tremuloides cell suspension cultures to salicyl alcohol feeding

Project description:Aspen cell cultures were fed 5 mM salicyl alcohol at the early exponential phase (5 days after subculture). Cells were harvested from fed (SL) or unfed (C) cells after 48 hours. Gene expression changes were analyzed using a custom 7K aspen EST array. RNA was extracted from 3 independent control (C) and 3 salicyl alcohol-fed (SL) samples, and one control and one treated sample were randomly paired for hybridization (3 biological replicates). Dye-swap hybridizations were carried out for each pair of samples.
