Expression profiling of mammary stem cell enriched fractions and other fractions from fetal and adult mouse.

ABSTRACT: Examination of genome-wide, gene expression level differences among adult and fetal mammary sub-populations including fetal CD24high cells and adult CD49fhighCD24medium cells which harbor enriched mammary stem cell activity. A genome wide 12-plex expression array using linear-amplified (Eberwine) mRNA from dissociated and FACS sorted fetal and adult mammary gland populations. Samples represent independent pooled biological replicates.

ORGANISM(S): Mus musculus

Gene expression signatures relating mammary stem cell populations to breast cancers have focused on adult tissue. Here, we identify, isolate, and characterize the fetal mammary stem cell (fMaSC) state since the invasive and proliferative processes of mammogenesis resemble phases of cancer progression. fMaSC frequency peaks late in embryogenesis, enabling more extensive stem cell purification than achieved with adult tissue. fMaSCs are self-renewing, multipotent, and coexpress multiple mammary li  ...[more]
