Project description:OsMADS1 in rice is an important transcription factor in controlling flower development, not only in flower organs development, but also in floral meristem determinacy. Early flower panicle we used is a high expression stage for OsMADS1. We used microarrays to detail the global gene expression underlying functions of OsMADS1 in rice flower development. Rice panicles in length of 2 mm and 5-7 mm were collectecd for RNA extraction and hybridization on Affymetrix microarrays.Rice panicles in length of 2 mm is the stage for the initiation of inner floral organ development and in length of 5-7 mm is the late stages for floral organ development.Though comparing the microarray data of osmads1 and wild-type (control) at these two stages, we can find the functions of OsMADS1 in the early and late flower development stages. Three biological repeats were used. And totally 12 samples were analyzed.

Project description:We cultured the cells of S. pmbe in EMM2, and harvested them in the OD600 of 0.5 and 10 respectively. We used microarrays to detail the the global program of gene expression of SP-Q01 and SP-Q01 carrying vevtor pESP3X to overexpress either ribosomal protein L32 paralog in the OD600 of 0.5 and 10 respectively. We constructed the ribosomal protein L32 paralog overexpression strains to mimic the expression pattern of RPL32 paralogs in wild-type cells of S.pombe. We cultured the cells to different growth phase, and used microarray to identify the genes were regulated distinctively by RPL32 paralogs.

Project description:The steriod-induced necrosis of femoral head(SINFH) is a serious clinical problem and the underlying mechanism of this disease remains unknown. As its etiology and pathogenesis are not clear, there hasn’t been an effective treatment yet. In-depth studies on its pathogenesis will provide important information for the prevention and treatment of this devastated disease. Animal model researches are helpful to a better understanding of its etiology and pathogenesis, to early diagnosis and treatments and provide the basis for clinical treatment It has been proved that the occurrence of SINFH is related to gene itself and its polymorphism. These studies, in which only one or a few genes were investigated, could not provide a comprehensive understanding to the pathogenesis of SINFH. We used microarrays to compare the gene expression profile of SINFH rats with that of normal rats and identified gene expression changes between SINFH rats and normal rats. Adult healthy male Wistar rats(body weight 200±10g) were randomly divided into control and experimental groups. All the rats were given injection of E. coli endotoxin at a dose of 20μg/kg body weight into abdominal cavity for two times with a 24 hour interval. 24 hours after the second administration of E. coli endotoxin, methylprednisolone 40mg/kg was administered into the left gluteus muscle of experimental group rats for three times with a 24 hour interval. Rats were then killed 6 weeks after steroid administration. In experiment, immediately after death the left femoral heads were isolated. Femoral heads were used for RNA extraction and hybridization on Affymetrix microarrays. We sought to obtain the gene expression profile of SINFH rats and that of normal rats in order to identified gene expression changes between SINFH rats and normal rats by comparing the gene expression profile of SINFH rats with that of normal rats. To that end, we have established the gene expression profiles of normal and SINFH disease of rat femoral head and found out differentially expressed genes.

Project description:The h+ and h- haploid cells of S. pombe mixtures flocculated and formed diploid cells when cultured in carbon limited YEPME medium. We first cultured S. pombe h+ YHL6381 and h- SPQ-01 cells respectively in YEPD medium and then mixed them at an 1:1 ratio and further cultured in YEPME medium. Sexual flocculation occured. We used microarrays to detail the the global programme of gene expression of flocculated YHL6381 and SPQ-01 mixtures cultured in YEPME, compared with the non flocculated cells mixtures cultured in YEPD, and identified distinct classes of up-regulated genes during this process. We first cultured S. pombe h+ YHL6381 and h- SPQ-01 cells respectively in YEPD medium and then mixed them at an 1:1 ratio and further cultured in a new medium for 90min. The flocculated cells mixtures cultured in YEPME named YSD, while the non flocculated cells mixtures cultured in YEPD named YSM.

Project description:Deleting ribosomal protein genes caused haploid cells of S. pombe flocculated in full nutrition YEPD medium. We deleted rpl32-1 and rpl32-2 gene respectively and these two mutant cells cultured in YEPD medium flocculated. We used microarrays to detail the global programme of gene expression between non flocculated rpl32-1 deleted and rpl32-2 deleted, compared with the non flocculated cells SPQ-01, and identified distinct classes of up-regulated genes during this process. Yeast cells cultured in YEPD medium in the log phase were selected for RNA extraction and hybridization on Affymetrix microarrays. The rpl32-1 deleted cells were conformed as SD1 and rpl32-2 deleted cells were conformed as SD2. Control cells SPQ-01 were conformed as WT.

Project description:Using microarray, we compared miRNAs expression profiles of MDA-MB-231 cells transfected with myocardin and empty vector (pcDNA3.1) and found that 25 miRNAs were significantly changed in myocardin-transfected groups (17 up-regulated and 9 down-regulated miRNAs). Moreover, we showed that 18 of 25 miRNAs significantly regulated by myocardin were inhibited by ERM-NM-1 in MDA-MB-231 cells. In addition,through a microarray approach, we identify the subset of miRNAs modulated by ERM-NM-1 in MDA-MB-231 cells. Our results determined that ERM-NM-1 may function as tumor-promoter through down-regulating expression of 3 miRNAs (miR-26b, miR-146a and miR-331-3p) in MDA-MB-231 cells. There are 4 samples in this experiment (pcDNA3.1-transfected, myocardin-transfected, ERM-NM-1-transfected, myocardin plus ERM-NM-1 co-transfecteded groups) and each sample was replicated 3 times. The vector-pcDNA3.1 was used as control.
