Expression data from Porcine alveolar macrophages infection with Haemophilus parasuis (HPS)

ABSTRACT: Porcine alveolar macrophages (PAMs) play impoartant role in innate immunity. Haemophilus parasuis is the etiological agent of Glasser’s disease in pigs. We used microarrays to study the transcriptome of PAMs infection with Haemophilus parasuis. Healthy Pigs were intratracheally challenged with H.parasuis strain 0165. And the PAMs were isolated at 6 dpi. RNA extraction were extracted from PAMs that obtained from infection pigs and control pigs and hybridization on Affymetrix microarrays. We sought to obtain the differently expressed genes that related to H.parasuis infection to understand the mechanism of PAMs against H.parasuis.

Healthy Pigs were inoculated intranasally with 2 ml of 4.5×108CFU/ml colony forming units of HPS4 strain MD0322. And the PAMs were isolated at 28 dpi. RNA extraction were extracted from PAMs that obtained from infection pigs and control pigs and hybridization on Affymetrix microarrays.

Project description:Porcine alveolar macrophages (PAMs) play an important role in innate immunity. Streptococcus suis serotype 2 is a pathogen responsible for several diseases in both pigs and humans. We used microarrays to study the transcriptome of PAMs infected with SS2. Healthy pigs were inoculated intranasally with 2 ml of 4.84×10^6 colony-forming units of SS2 strain SC19. The PAMs were isolated at 7 dpi. RNA was extracted from PAMs obtained from infected pigs and control pigs, and hybridized on Affymetrix microarrays.

Project description:Haemophilus parasuis(HPS) is a prominent swine pathogen that causes Glässer's disease characterized by fibrinous polyserositis, meningitis and arthritis; however, the molecular mechanisms underlying disease pathogenesis remains poorly understood, particularly the host counteraction to HPS invasion by the immune system. Here, we investigated the global expression changes in spleen following HPS infection using the Affymetrix Porcine Genechip.Our findings indicate previously unrecognized gene transcription changes in case of HPS infection in vivo and many presumed cascades in the study are clearly merit further investigation. Our data should provide new clues for immune response in mammals and identification of candidate genes related to HPS resistance.

PRRSV-infection of porcine alveolar macrophages

Project description:The goal of this study was to produce a deep, global analysis of gene expression changes that occured following infection of normal porcine alveolar macrophages (PAMs) with PRRSV. The goal was to examine the gene expression changes to help determine the mechanisms that result in reduced function and immunosuppression observed in PRRSV-infected pigs. Keywords: time course of infection The PAMs were infected in culture at an MOI of 10 with PRRSV strains VR-2332 and incubated at 37C until 6, 12, 16 or 24 hours post infection. Total cellular RNA was collected from each at the appropriate time. SAGE libraries were prepared from each infected time point as well as from noninfected PAMs. The SAGE libraries were sequenced to at least 95,000 tags each.

Comparison of Gene Expression in Lung of Pigs Exhibiting Different Susceptibilities to Glasser's Disease

Project description:Haemophilus parasuis is the causative agent of Glasser’s disease in pigs, but this bacterium is also routinely isolated from healthy animals. Whether colonization of the respiratory tract ultimately results in the onset of disease is determined by a combination of environmental, bacterial and host factors that are incompletely understood. This study used DNA microarrays to identify genes that were differentially expressed in the lung of colostrum deprived animals previously characterized as being either ‘Resistant’ or ‘Susceptible’ to infection by H. parasuis in a bacterial challenge experiment at either 24 or 72 hours post-inoculation. Gene expression profiles were obtained by comparing gene expression between each group and a pool of ‘Uninfected control’ animals. The aims of the experiment were: 1) To identify genes associated with resistance and susceptibility to H. parasuis. 2) To identify genes associated with the molecular pathology of Glasser's disease. 3) To identify genes involved in the host response to infection in the lung. Lung tissue RNA from 6 'Fully Resistant' (2 at 24 hours and 4 at 72 hours post-inoculation) and 6 'Susceptible' pigs (2 at 24 hours and 4 at 72 hours post-inoculation), together with pools of RNA from 9 mock inoculated control animals were used in this experiment. This two-colour microarray experiment utilized a reference design. Labelled target DNA from the pool of mock infected control animals (the reference) was co-hybridized with labelled target prepared from individual 'Fully Resistant' or 'Susceptible' animals to a porcine oligonucleotide microarray. Four technical replicates were carried out for each animal (2 sets of dye-swaps);a total of 48 hybridizations. The design allowed for four comparisons to be made: 1. Fully Resistant vs Control at 24 hours. 2) Fully Resistant vs Susceptible at 24 hours. 3) Fully Resistant vs Control at 72 hours. 4) Fully Resistant vs Susceptible at 72 hours.

Pulmonary alveolar macrophages (PAMs) of Tongcheng piglets response to HP-PRRSV infection in vivo

Project description:Porcine reproductive and respiratory syndrome (PRRS), caused by PRRS virus (PRRSV), is the most economically important disease in pig populations. Lung damage is one major pathological condition following PRRSV infection, often leading to animal death. In vivo, PRRSV productive infection occurs predominately in alveolar macrophages of the lung. Here, transcriptome profiling of pulmonary alveolar macrophages (PAMs) from Tongcheng piglets pre- and post- infection of highly pathogenic PRRSV has been performed using porcine Affymetrix GeneChip. All animal procedures were performed according to protocols approved by the Biological Studies Animal Care and Use Committee of Hubei Province, China. Piglets used in this study were free from PRRSV, pseudorabies virus (PRV) and porcine circovirus type 2 (PCV2) determined by ELISA test for serum antibodies. Twelve of 5-week-old boars were obtained from three litters (four piglets per litter), and raised in pathogen-free facilities. In order to perform a paired experiment, every four full-sib individuals were divided equally into two groups: one infected group and one control group with 6 piglets in each group. The infected groups were challenged with PRRSV-Wuh2 (3 ml/15 kg, 10-5 TCID50/ml) by intramuscular inoculation. Slaughters were carried out at 0 days post-infection (dpi) for uninfected (control) groups, and at 5 or 7 dpi for infected groups. The PAMs for microarray analysis were collected by bronchoalveolar lavage from three uninfected pigs and three infected pigs at 5 dpi. Total of 6 microarrays have been hybridized in this experiment.

All animal tissue collection procedures were performed according to protocols approved by The Hubei Province, PR China for Biological Studies Animal Care and Use Committee. Piglets at 30 days old were determined to be HPS-free by serum indirect haemagglutination test before artificial bacterial challenges. Each piglet was intratracheally challenged with 5M-CM-^W108 colony-forming units (CFU) per millilitre of HPS strain 0165 (serotype 5) . A total of 5 piglets were challenged, and 5 pigs were used as controls. These two groups were raised in isolated facilities. Six microarrays were used in the experiment, corresponding to the RNAs from spleen tissues of three most severe piglets with fibrinous polyserositis, meningitis and arthritis after HPS infection and three controls.

Global Gene Expression Profiles of Cecropin B Induces Resistance in Haemophilus parasuis

Project description:To understand the mechanism of the adaptations, global gene expression profiles of the cecropin B--resistant strains of Haemophilus parasuis, The development of cecropin B (CB) resistance in H. parasuis SH0165 by exposing SH0165 using various concentrations of CB was investigated. One colony of bacterial cultures grown on TSA containing 30μg/mL CB was named CBR30, CBR30 cultured in TSA without CB for 50 passages was named CBR30-50. we used microarray technology to analyze the variation of the H. parasuis SH0165 transcriptional profile in CBR30 and CBR30-50. Our analysis to identify several genes whose system could be involved in inorganic ion transport, amino acid transport, and metabolism. Quantitative PCR was used to validate the differential expression of selected genes.
