Expression profile in wild type, fkh2delta and fkh2-S2A mutants

ABSTRACT: Mitotic entry is accompanyed by the expression of a cluster of so called mitotic genes, whose activation is critical for mitosis in human and yeast cells. We found a link between the transcription machinery and cell cycle control network at mitosis in fission yeast, involving the Cdk8 kinase dependent phosphorylation of the fork head transcription factor Fkh2. We have generated a non-phosphorylatable fkh2 mutant (fkh2-S2A) also. We have used microarrays to study the gene expression changes in fkh2 deletion and phosphomutants vs Wt in exponentially growing fission yeast cells to detect how the phosphorylation may affect global and mitotic gene expression. RNA samples were taken from exponentially growing fission yeast cells and hybridized to affymetrix microarrays: wild type, fkh2delta and fkh2-S2A mutants

Temporal changes in transcription programs are coupled to control of cell growth and division. We here report that Mediator, a conserved coregulator of eukaryotic transcription, is part of a regulatory pathway that controls mitotic entry in fission yeast. The Mediator subunit cyclin-dependent kinase 8 (Cdk8) phosphorylates the forkhead 2 (Fkh2) protein in a periodic manner that coincides with gene activation during mitosis. Transcriptome analysis of mediator Med20 mutant and dcr1 mutant in S.pombe

Project description:At Schizosaccharomyces pombe centromeres, non-coding RNAs are transcribed by RNA polymerase II and processed by the RNAi machinery to direct heterochromatin formation. Mediator is a regulator of RNA polymerase II transcription and we here demonstrated that it also controls the levels of centromeric transcripts. Loss of the Mediator subunit Med20 increases transcription of non-coding RNAs and disrupts the centromeric heterochromatin structure. We have used tilling arrays to study the gene expression changes in Med20 deletionVs WT and Med20 dcr1 double mutant Vs Dcr1 mutant in exponentially growing fission yeast cells to detect how the Med20 gene affect centromeric expression. RNA samples were taken from exponentially growing fission yeast cells and reverse transcribed cDNA from the RNA are hybridized to affymetrix pombe tilling arrays: wild type,med20 mutant, Dcr1 mutants and med20,dcr1 double mutant.

Project description:Many genes are transcriptionally regulated as a function of the cell cycle; genes needed for mitosis are strongly cell cycle regulated. From yeasts to humans, forkhead transcription factors are involved in mitotic gene expression. However, the mechanisms of regulation are unclear, especially since forkhead transcription factors can be activators or repressors, and often function in concert with other proteins. Here, we study mitotic regulation in the fission yeast S. pombe. We find that the forkhead transcription factor Fkh2 is a repressor of many mitotic genes, while a different forkhead transcription factor, Sep1, is an activator of a subset of these genes. The activator of the majority of mitotic genes appears to be Sak1, a transcription factor of the RFX family, which has not previously been implicated in cell cycle control. 3 test samples were analyzed; 1 control (no-tag) sample was processed (each sample has 2 replicate experiments)

Project description:Mitotic genes are one of the most strongly oscillating groups of genes in the eukaryotic cell cycle. Understanding the regulation of mitotic gene expression is a key issue in cell cycle control but is poorly understood in most organisms. Here, we find a new mitotic transcription factor, Sak1, in the fission yeast S. pombe. Sak1 belongs to the RFX family of transcription factors, which have not previously been connected to cell cycle control. Sak1 binds upstream of mitotic genes in close proximity to Fkh2, a forkhead transcription factor previously implicated in regulation of mitotic genes. We show that Sak1 is the major activator of mitotic gene expression and also confirm the role of Fkh2 as the opposing repressor. Sep1, another forkhead transcription factor, is an activator for a small subset of mitotic genes involved in septation. From yeasts to humans, forkhead transcription factors are involved in mitotic gene expression, and it will be interesting to see whether RFX transcription factors may also be involved in other organisms.

Condensin-mediated remodeling of the mitotic chromatin landscape in fission yeast

Project description:Chromatin fibres dynamically change their organisation during cell cycle. In interphase nucleus, chromatin fibres are evenly distributed whereas their spatial occupancy are reorganised to form condensed chromosomes in mitosis. This process called chromosome condensation is necessary for an accomplishment of faithful chromosome segregation. One of the Structural Maintenance of Chromosomes complexes, Condensin, is indispensable for chromosome condensation. It remains, however, unknown how Condensin plays its role in shaping mitotic chromosome. Here we show that chromatin fibres change their interacting partners; short-range contacts in interphase nucleus are converted into long-range interactions to shape condensed chromosomes. This conversion of interactions among chromatin fibres results in the formation of larger domains within mitotic chromosomes. Condensin is solely in charge of the conversion and large domain formation in fission yeast mitosis. Our results show how fission yeast Condensin is involved in shaping mitotic chromosomes.

Expression analysis of Candida albicans following a block in mitotic exit

Project description:Candida albicans demonstrates three main growth morphologies yeast, pseudohyphal and true hyphal forms. Cell separation is distinct in these morphological forms and the process of separation is closely linked to the completion of mitosis and cytokinesis. In Saccharomyces cerevisiae the small GTPase Tem1 is known to initiate the mitotic exit network, a signalling pathway involved in signalling the end of mitosis and initiating cytokinesis and cell separation. We have characterised the role of Tem1 in C. albicans, and demonstrate that it is essential for mitotic exit and cytokinesis, and that this essential function is signalled through the kinase Cdc15. Consistent with its role in activating the mitotic exit network Tem1 localised to spindle pole bodies in a cell cycle dependent manner. Cells depleted of Tem1 displayed highly polarised growth but ultimately fail to complete cytokinesis and re-enter the cell cycle following nuclear division. At the transcriptional level genes downregulated following the depletion of Tem1 where significantly enriched for genes whose expression peaks early in the cell cycle and for those associated with glycolysis. For expression analysis triplicate RNA samples were generated from exponentially growing C. albicans yeast cells and from cells following 8 h inhibition of TEM1 expression. Gene expression was determined using RNA-seq on the Illumina HiSeq 2000 platform.
