MRNA expression profiling in nine human tissues

ABSTRACT: Profiled mRNA gene expression in nine different commercially purchased total RNA samples which were also profiled for miRNAs. 500ng toal RNA from each tissue was labeled and hybridized in at least duplicate, using Agilent Low Input Linear Amplification Kit.

miRNA Microarray Profiling of Nine Different Normal Human Tissues

Project description:We measured the expression of 470 human miRNAs in nine human tissues using Agilent DNA microarrays. miRNA microarrays were manufactured by Agilent Technologies (Santa Clara, CA), and contain 20-40 features targeting each of 470 human miRNAs (Agilent design ID 016436). Sequences of the 470 miRNAs were obtained from the Sanger miRBase, release 9.1. Labeling and hybridization of total RNA samples were performed according to the manufacturer’s protocol. 100ng total RNA was used as input into the labeling reaction, and the entire reaction was hybridized to the array for 20 hours at 55°C. The labeling and hybridizations of the nine human tissues were done 4-5 times using the same starting RNA sample for each tissue.

Expression profiling of lymph nodes in tuberculosis patients reveal inflammatory milieu at site of infection

Project description:Extrapulmonary manifestations constitute 15-20% of tuberculosis cases, with lymph node as the most common site. Understanding of disease etiology is limited due to the lack of understanding patientsM-bM-^@M-^Y infected tissue milieu. This study was designed to perform global transcriptome analysis of lymph node tissues from healthy individuals and Mycobacterium tuberculosis infected lymph nodes of patients to decipher the local response of infected tissue. This study was designed to elucidate gene expression signatures in the event of Lymph Node Tuberculosis. Total RNA was extracted from lymph node tissue samples of LNTB patients in BSL3 facility and together with healthy lymph node RNA samples (commercially purchased) their global transcriptome profiling was performed using Illumina HumanHT-12 V4 expression beadchip.

Transcriptomic profiling of bovine IVF embryos revealed candidate genes and pathways involved in early embryonic development

Project description:Using microarrays, genome-wide RNA expression was profiled and compared for in vitro fertilization (IVF) - derived blastocysts and embryos undergoing degenerative development up to the same time point. Surprisingly similar transcriptomic profiles were found in degenerative embryos and blastocysts. Nonetheless, we identified 67 transcripts that significantly differed between these two groups of embryos at a 15% false discovery rate, including 33 transcripts showing at least a two-fold difference. Several signaling and metabolic pathways were found to be associated with the developmental status of embryos, among which were previously known important steroid biosynthesis and cell communication pathways in early embryonic development. Three biological replicates for each of blastocysts and degenerative embryos.
