Project description:Exposure to Polychlorobiphenyls (PCBs) is known to cause serious health effects in human but the gene expression profiles leading to development of differnet diseases is not fully understood. The knowledge of global gene expression will help us to devlop early diagnostic biomarkers for PCB induced health effects. We used microarrays to detail the global gene expression profile underlying the effects of PCB153 on human liver cells leading to identification of distinct classes of up-regulated and down-regulated genes and cellular processes. Keywords: time course The HepG2 cells were grown for 48 hours in DMEM supplemented with 10% FBS and 1X Pennicillin-Streptromycin and treated with PCB153 for different times. Trizol extraction of total RNA was performed according to the manufacturer's instructions. Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 6 microgram of total RNA (Expression Analysis Technical Manual, 2001, Affymetrix). Following fragmentation, 10 microgram of cRNA were hybridized for 16 hr at 45C on GeneChip Human Genome Array (HGU133 plus 2.0). GeneChips were washed and stained in the Affymetrix Fluidics Station 450 and scanned using the Affymetrix GeneArray Scanner 3000. The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 150.

Project description:CD4+ T cells from 8-12 week female mice were isolated from wt and AhR-/- mice 24h after injection of 10µg/kg TCDD or solvent control. Since TCDD treatment showed only marginal impact on the gene expression (~10 transcripts effected in wt mice) wt - ko comparisons were performed on pooled profiles from TCDD and solvent mice.

Project description:Dioxin-like chemicals are well-known for their ability to upregulate expression of numerous genes via the AH receptor (AHR). However, recent transcriptomic analyses in several laboratories indicate that dioxin-like chemicals or AHR genotype itself also can downregulate levels of mRNAs encoded by numerous genes. The mechanism responsible for such downregulation is unknown. We hypothesized that microRNAs (miRNAs), which have emerged as powerful negative regulators of mRNA levels in several systems, might be responsible for mRNA downregulation in dioxin/AHR pathways. We used the Exiqon miRNA array platform as well as quantitative RT-PCR to measure miRNA levels in dioxin-sensitive Long-Evans (Turku/AB; L-E) rats vs. dioxin-resistant Han/Wistar(Kuopio; H/W) rats. Treatment with 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) in vivo caused few changes in miRNA levels in rat livers and those changes that were statistically significant were of modest magnitude. AHR genotype had little effect on hepatic miRNA levels, either in constitutive expression or in response to TCDD – only a few miRNAs differed in expression between between L-E rats (that have wildtype AHR) compared to H/W rats (whose AHR has a large deletion in the transactivation domain). It is unlikely that mRNA downregulation by dioxins is mediated by miRNAs, nor are miRNAs likely to play a significant role in dioxin toxicity in adult rodent liver. We conducted a thorough investigation to address the following questions: (1) does AHR genotype itself affect constitutive expression of microRNAs? (2) does TCDD affect microRNA levels and, if so, is this response dependent on the AHR? (3) does TCDD affect microRNA levels differently in animals that are sensitive to dioxin toxicity versus those that are dioxin-resistant? We assessed the in vivo effect of TCDD on microRNA levels in liver at multiple time points after TCDD treatment using microRNA arrays along with quantitative RT-PCR. The cumulative results of our experiments indicate that downregulation of mRNA levels by dioxins in adult rodent livers is very unlikely to involve microRNAs. Experiment Overall Design: There are 6 control chips and representing animals treated with vehicle control. There are 3 biological replicates (3 chips) for each treatment group (eg. Intensities were normalized using the GC-Robust Multiarray (GCRMA) method through Genetraffic software package.

Project description:The One Health initiative connects human and animal health with their shared environment. The development of novel methodology to unbiased identification of mechanisms of action of chemicals and chemicals mixtures would address one of today´s challenges of environmental and human toxicology. Zebrafish embryos offer a unique opportunity to explore the proteome integral solubility alteration (PISA) assay to study the proteins that increase or decrease their solubility as protein targets of the bioactive compounds in an organism at an developmental stage. We presented here 4 different applications of the PISA method in zebrafish embryo with different types of compounds of environmental and health concern: i) endocrine disrupting chemical, 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) to outline the pathways compromised under exposure; ii) complex chemical mixtures composed by TCDD + alpha-endosulfan + bisphenol A (BPA) previously studied in vitro cells lines to evaluate single chemical versus mixture; iii) novel compounds from blue-green circular economy to map the organismal response, and a iv) novel drug directed to anti-cancer activity in lymphoma models to discharge side-effects at non-target organs. Our results demonstrated the unique opportunities of PISA assay in zebrafish for toxicology. We remark some differences to its application for drug target deconvolution. First, we evaluate both targets arising from an increase in solubility and in those decreasing, as both would be involved in modulation the cellular functions. Second, the method could be applicable to bioactive compound before chemical characterization or a chemical mixture of unknown stoichiometry. Third, the results from zebrafish embryo could provide a first glimpse of unspecific interactions with proteins in non-target tissues.The introduction of PISA assay in toxi- and ecotoxicoproteomics offers high-resolution and throughput in support with the 3R, and 3M principles.
