Expression data from C. elegans

ABSTRACT: We exposed wild-type Vibrio cholerae E7496, multiple Vibrio cholerae virulence factor deleted genes with intact hemolysin A gene [CVD109] and without hemolysin A gene [CVD110] in E7946, and E.coli OP50 to wild-type C.elegans N2 for 18 hours. We used microarrays to detail the global gene expression and identified distinct classes of up-regulated and down-regulated genes during this process. C. elegans were exposed to Vibrio cholerae and E.coli then hybridization on Affymetrix microarray chips.

Whole-genome expression profiling of C. elegans larvae and adults

Project description:Temperature, as a universal enviromental factor, has prolonged effect on physiological and pathological functions of different species. In order to expolore the temperol effect of temperature on C.elegans longevity, we used microarray to check the whole-genome expression profiling of L4 larvae and Day3-old adults of C.elegans maintaining at different temperature Adult worms are collected after maintained at 25 degree and 15 degree for 3days from later L4 stage, while Larva worms are collected after growing at 25 degree and 15 degree from embryo to late L4 stege. Then total RNA are extracted with TRI reagent(Life Technologies) from~120 adults or ~300 larvae. The concentrations and quality of total RNA are checked with Thermo Nanodrop 2000c(Thermo Scientific,WilminGton,DE) and Agilent 2100 BioAnalyzer (Agilent Technologies, Palo Alto, CA). Total RNA samples were hybrydized with Affymetrix C. elegans Gene 1.1 ST Array Strip. Preparation of cDNA, hybridization, quality controls and scanning of arrays were performed according to the manufacturer's protocol (Affymetrix, Santa Clara, CA) at the microarray core facility of University of Michigan .

mRNA microarray analysis on young adults C.elegans exposed to GO and rGO

Project description:In order to evaluate the identification of genes and pathways, the global gene expression profiles were assessed in response to GO and rGO on nematode, Caenorhabditis elegans. We performed whole genome DNA microarray experiments using age synchronized young adult C. elegans population exposed to GO and rGO for 24h. We used whole genome microarrays to screen for global changed in C. elegans transcription profiles and with subsequent quantitative analysis conducted on selected genes. Young adults of C.elegans were selected for RNA extraction and hybridization on Affymetrix microarrays.
