GATA Transcription Factor (CGA1) Regulates Chloroplast Development in Rice

ABSTRACT: A transcription factor CYTOKININ-RESPONSIVE GATA FACTOR 1 (CGA1) regulates chloroplast development in rice (Oryza sativa) through modifying the expression of important nuclear expressed, chloroplast localized genes. A transcriptome analysis was done in wild type plants and transgenic rice over-expressing this OsCGA1 to identify the set of genes with altered expression. RNA was extracted from leaves of 4-wk old wild type and OsCGA1 overexpressing rice plants and hybridized to Affymetrix Rice Genome Array. Three biological replicates were sampled for wild type and OX plants.

Project description:In this study we analyzed the effects of CRY2 over-expression on chloroplast genome transcription of tomato, by developing and using a tiling array. This array containing about 90,000 overlapping probes (5-nt resolution) is a versatile tool for global functional studies of tomato cp genome. We profiled transcription in leaves of wild-type (WT) and CRY2-overexpressing (CRY2-OX) plants grown in a diurnal cycle, to generate a comprehensive map of plastid transcription and to monitor potential specific modulations of chloroplast transcriptome induced by the overexpression of CRY2.

Project description:Ghd7 regulates a range of functions in growth and development in response to environmental cues to maximize the reproductive success of the rice plant. We used microarrays to detail the regulated genes in OX-Ghd7HJ19 transgenic plants. To gain clues to downstream genes regulated by Ghd7, we performed a microarray analysis using Affimetrix rice gene chips. Young leaves in vegetative stage (35-days old) and developing panicles (0.1cm) from field-grown OX-Ghd7HJ19 transgenic and wild-type plants with two biological replicates were used to isolate RNA for chip analysis.

Analysis of phot2 phosphorylation profiles and interaction partners involved in chloroplast accumulation and avoidance responses in Arabidopsis thaliana

Project description:The project aims to use photocycle mutants to understand the importance of phot2 autophosphorylation for signaling leading to chloroplast accumulation and avoidance. The second part is focused on the identification of new interacting proteins for phot2. Characterization of proteins important for signaling leading to chloroplast avoidance. PHOT2-GFP wild type and PHOT2-V392L-GFP muteins were immunoprecipitated from leaves of transgenic Arabidopsis plants, either dark-adapted or irradiated with low or high blue light. Proteins identified by Mass Spectrometry as interacting with wild-type PHOT2-GFP and PHOT2-V392L-GFP were compared to identify proteins putatively involved in chloroplast avoidance. Phosphorylation profiles of PHOT2-GFP wild type and PHOT2-V392L-GFP were analyzed to distinguish between phosphorylation sites characteristic for chloroplast accumulation and avoidance.

The FtsH-inactive Protein TSL2/FtsHi5 Is Required for Chloroplast Development and Proteostasis at Low Ambient Temperature in Arabidopsis thaliana

Project description:Chloroplasts are indispensable for higher plants. The growth and development of plants are very sensitive to environmental temperature changes, and chloroplast development is also regulated by adverse environmental temperatures. However, the molecular mechanism of how plants coordinate chloroplast development and environmental temperature changes remains largely unknown. Here, a temperature-conditioned chloroplast development defect mutant tsl2 (thermo-sensitive mutant in leaf color 2) of Arabidopsis was obtained through forward genetic screening. The tsl2 mutant showed a weak yellowing phenotype at 22 °C which was more pronounced when the growth temperature was reduced to 16 °C, and was similar to that of the wild type when the temperature was increased to 29 °C. Genomic resequencing analysis revealed that TSL2 gene encodes the FtsHi5 which was shown to function in chloroplast protein import. Genetic complementation analysis showed that TSL2 rescued the chlorophyll content and thylakoid development defects in tls2 mutants at 16 °C. Quantitative mass spectrometry analysis with Tandem Mass Tag (TMT) isobaric labeling revealed broad changes in the proteome composition of tsl2 chloroplasts at low temperature, reflecting impaired chloroplast biogenesis and function. Together, our data suggest that TSL2 has a significant role in chloroplast development and proteostasis especially at low environmental temperatures.

The genes upregulated in WRKY45-ox rice relative to Nipponbare

Project description:WRKY45-overexpressing (WRKY45-ox) rice plants are extremely resistant to rice blast and bacterial leaf-blight diseases. To search for the genes that are potentially responsible for the strong resistance, we performed genome-wide gene expression analysis in WRKY45-ox rice. A microarray analysis using Agilent rice oligo microarray for approximately 42,000 rice genes identified 1,664 genes that are upregulated in WRKY45-ox rice compared with non-transformant rice cv. Nipponbare (NB). Analysis of the genes with altered expression in WRKY45-ox rice by a Gene ontology program (Agri GO; http://bioinfo.cau.edu.cn/agriGO/) revealed that several genes in the biosynthetic pathway for diterpenoid phytoalexins, for which anti-blast fungus activities have been reported, were upregulated in WRKY45-ox rice.
