Expression analysis of circadian light response in Drosophila

ABSTRACT: Light is a strong environmental cue that resets the circadian clock rhythm. We have used Affymetrix microarrays to profile the Drosophila head transcriptome response following a brief light pulse during the early night. Comparison of control (un-pulsed) and experimental flies reveal large number of differentially expressed transcripts underlying the process of light entrainment of the clock. Wild type flies exposed to light pulse at ZT15 compared to control group (no pulse)

Project description:The aim of this study was to identify quantitative trait loci (QTL) associated with variation in circadian photosensitivity in Drosophila, by measuring the locomotor response to early night light pulse (Zt15). We used QTL mapping of recombinant inbred lines (RIL), which was followed by microarrays expression comparison of two RI lines which differed significantly in their light response (RIL 104 shows a strong long phase delay response while RIL 58 shows a weak light response). Transcription profiling of mouse SCN after 30-min light pulse

Project description:The transmission of information about the photic environment to the circadian clock involves a complex array of neurotransmitters, receptors, and second messenger systems. Using laser capture microscopy and microarray analysis, a population of genes rapidly induced by light in the suprachiasmatic nucleus is identified. Experiment Overall Design: Mice were exposed starting at 1 hour after lights off to a 30-minute light pulse. At the end of the light pulse, mice were euthanized and the brains extracted. Cells from the suprachiasmatic nucleus were extracted using laser capture microscopy and gene expression was quantified using Affymetrix microarrays. Genes showing a significant increase in expression following the light pulse as compared to a sham light pulse were identified.

Project description:Light pulses at the end of the day or night be able to control the phase of the circadian clock. Pulses in the middle of the night has not effect on the circadian oscilations. To understand how the circadian clock gate the light signal in the middle of the night we used microarrays to characterize the light effect in the whole expression profile. Plants of Arabidopsis thalina (Columbia ecotype) were grown in 12/12 light/dark cicles for 15 days and then were tranfer to continuous dark . Plants were slplit in two groups. First group recieved 1hr light pulse in the middle of the night (Night treatment), second group recieve 1hr light pulse in the middle of the subjective day (Day Treatment). Samples of both groups were collected without light pulse named as a control.
