SM020 - Infection with SARS MA15 of C57BL/6J mice and KEPI (ppp1r14c) knockouts

ABSTRACT: Comparing the host response of C57BL/6J mice to KEPI (ppp1r14c) knockouts in the same strain of mice. Groups of 10-week-old C57BL/6 mice or KEPI (ppp1r14c) knockout mice were infected with SARS MA15 at a dose of 10^5 PFU or mock infected. Mice were euthanized on days 4 and 7 post-infection to measure virus load and isolate samples for measurement of virus load, lung pathology, transcriptional analysis and proteomics analysis. Mock-infected animals were also harvested at each time point. Mice were weighed every 24 hours to measure general disease progression and any mouse approaching 30% weight loss was euthanized.

Transcriptomic analysis of host response to mouse-adapted SARS virus in wild type, STAT1 -/-, and IFNAR1 -/- mouse genetic backgrounds

Project description:The mechanisms by which pulmonary lesions and fibrosis are generated during SARS-CoV infection are not known. Using high-throughput mRNA profiling, we examined the transcriptional response of wild-type (WT), type I interferon receptor knockout (IFNAR1−/−), and STAT1 knockout (STAT1−/−) mice infected with a recombinant mouse-adapted SARS-CoV (rMA15) to better understand the contribution of specific gene expression changes to disease progression. Ten week old 129S6/SvEv wild-type, STAT1−/− (Taconic Farms, Germantown, NY), and IFNAR1−/− mice bred on a 129SvEv background were anesthetized with a ketamine and infected intranasally with either phosphate-buffered saline (PBS) alone (Invitrogen, Carlsbad, CA) or 1 × 10^5 PFU rMA15-PBS. Mice were euthanized and left lungs were harvested from individual mice (a total of 3 infected mice from each strain) at days 2, 5, and 9 postinfection (dpi) for microarray analyses. Lung samples were taken from mock-infected animals from each of the strains at 5 dpi.

Identifying microRNA-mRNA regulatory networks in colorectal cancer by a combination of expression profile and bioinformatics analysis

Project description:A multi-step approach combining microarray profile and bioinformatics analysis was adopted to identify the CRC specific miRNA-mRNA regulatory network. First, differentially expressed miRNAs and mRNAs were found out in CRC samples compared with normal epithelial tissues by miRNA and mRNA microarray respectively. Secondly the target mRNAs of dysregualted miRNA were identified by a combination of Pearson correlation coefficient between the expression level of miRNAs and mRNAs and online miRNA target predicting databases. Thirdly, the biological pathways which the miRNA-mRNA pairs involved in were identified by DAVID. Finally, some of the dysregualted miRNAs and mRNAs were validated by qRT-PCR RNA isolated from 8 colorectal cancer tissues and their corresponding adjacent normal tissues were analyzed.

The microRNA-205 is correlated to metastatic potential of 21T series: a breast cancer progression model

Project description:To gain insights into the mechanisms involved in breast cancer progression we conducted a microRNA global expression analysis on a 21T series of cell lines obtained from the same patient during different stages of breast cancer progression. These stages are represented by cell lines derived from normal epithelial (16N), atypical ductal hyperplasia (21PT), primary in situ ductal carcinoma (21NT) and pleural effusion of a lung metastasis (21MT-1 and 21MT-2). In a global microRNA expression analysis, miR-205 was the only miRNA to display an important downregulation in the metastatic cell lines (21MT-1; 21MT-2) when compared to the non-invasive cells (21PT and 21NT). The lower amounts of miR-205 found also correlated with high histological grades biopsies and with higher invasion rates in a Boyden chamber assay. This work pinpoints miR-205 as a potential player in breast tumor invasiveness. These experiments are conducted as 2 replicates.

Project description:Purpose of experiment was to perform transcriptomic analysis on C57Bl/6 mice infected with different doses of SARS CoV MA15 at 4 different days post infection. Twenty-week-old C57BL/6 mice were infected by intranasal instillation of 10^2, 10^3, 10^4 or 10^5 PFU of SARS CoV MA15 in 50 µl of PBS or mock-infected with PBS alone. At days 1, 2, 4 and 7 days post-infection, lungs were harvested and total RNA was isolated and subjected to microarray analysis. Maize gene expression during infection with Colletotrichum graminicola

Project description:The hemibiotrophic fungal pathogen Colletotrichum graminicola is the causal agent of anthracnose disease on maize stalks and leaves. After the formation of appressoria the host cell wall is penetrated by the conversion of appressorial turgor pressure into forceful ejection of a penetration peg. Subsequently, C. graminicola establishes biotrophic hyphae in the penetrated epidermis cell at around 36 hours post inoculation (hpi) until a switch of hyphal morphology and lifestyle takes place during the colonization of neighboring host cells at around 72 hpi. During the ensuing necrotrophic growth, dark necrotic lesions are formed that are visible as anthracnose symptoms. We used microarrays to detail the global programme of gene expression during the infection process of Colletotrichum graminicola in its host plant to get insight into the defense response of this compatible interaction and into the metabolic reprogramming needed to supply the fungus with nutrients. In three independent experiments, maize plants were infected with conidia of the Colletotrichum graminicola strain CgM2 by spray inoculation of leaves. Samples from infected leaves were taken at 36 and 96 hours post infection, corresponding to initial biotrophic and necrotrophic phase, respectively. Samples from uninfected control plants were taken at the same time points.
