Preliminaly gene transcriptional profiling of the oesophageal gland of male S. mansoni

ABSTRACT: This study provides a transcriptomic analysis of the oesophageal gland of S. mansoni male. The results obtained will help direct future research on the functional aspects of this tissue, and will allow the characterisation of currently unannotated genes in S. mansoni Quantification of gene expression of S. mansoni male oesophageal gland was achieved by a combination of laser microdissection microscopy (LMM) and oligonucleotide microarray analysis. We compared the gene expression profile of the adult male oesophageal glands with whole male worm controls.

We compared the gene expression profile of the adult female gastrodermis and male and female reproductive tissues with whole worm controls. This is complemented by a set of data from macrodissected samples from different regions of the worm.

Small RNA content in mouse serum upon S.mansoni infection

Project description:MicroRNAs (miRNAs) are a class of short non-coding RNA that play important roles in disease processes in animals and are present in a highly stable cell-free form in body fluids. Here we examine the capacity of host and parasite miRNAs to serve as tissue or serum biomarkers of Schistosoma mansoni infection. Sequencing of small RNAs from serum confirmed the presence of miRNAs and revealed 11 parasite-derived miRNAs that were detectable by 8 weeks post S.mansoni infection. Small RNA content in serum of naïve and Schistosoma mansoni infected mice were examined in two different librarys. 1- prepared according to the 290 Illumina small RNA Sample Preparation Kit version 1.5 and sequenced on the GAIIX and 2- prepared according to the TruSeq Small RNA protocol (without size-selecting small 295 RNA) and sequenced on the HiSeq2

microRNA expression during ovine adult mammary gland development

Project description:The mammary gland undergoes extensive remodeling between the begin- ning of pregnancy and lactation; this involves cellular processes including cell proliferation, differentiation, and apoptosis, all of which are under the control of numerous regulators. To unravel the role played by miRNA, we describe here 47 new ovine miRNA cloned from mammary gland in early pregnancy displaying strong similari- ties with those already identified in the cow, human, or mouse. A microarray study of miRNA variations in the adult ovine mammary gland during pregnancy and lactation showed that 100 miRNA are regulated according to three principal patterns of expression: a de- crease in early pregnancy, a peak at midpregnancy, or an increase throughout late pregnancy and lactation. One miRNA displaying each pattern (miR-21, miR-205, and miR-200b) was analyzed by qRT- PCR. Variations in expression were confirmed for all three miRNA. Using in situ hybridization, we detected both miR-21 and miR-200 in luminal mammary epithelial cells when expressed, whereas miR-205 was expressed in basal cells during the first half of pregnancy and then in luminal cells during the second half. We therefore conclude that miR-21 is strongly expressed in the luminal cells of the normal mammary gland during early pregnancy when extensive cell prolif- eration occurs. In addition, we show that miR-205 and miR-200 are coexpressed in luminal cells, but only during the second half of pregnancy. These two miRNA may cooperate to maintain epithelial status by repressing an EMT-like program, to achieve and preserve the secretory phenotype of mammary epithelial cells. 5 samples for sheep and 5 samples for mouse

Dnmt2-dependent methylomes lack biologically relevant DNA methylation patterns [Schistosoma mansoni]

Project description:Dnmt2 is a widely conserved protein, which is closely related to eukaryotic DNA methyltransferases. However, Dnmt2 shows a robust tRNA methyltransferase activity and only limited activity towards DNA. Interestingly, a recent study has provided evidence for a biologically important function of Dnmt2-dependent DNA methylation in the blood fluke Schistosoma mansoni, which seemed to contradict the weak activity of the enzyme in other organisms. We now used whole-genome bisulfite sequencing to comprehensively analyze the methylome of adult worms and could not detect any evidence for biologically relevant DNA methylation patterns. We also characterized the methylome of Drosophila melanogaster embryos and did not find any evidence for DNA methylation. Unconverted cytosine residues were detectable only at very low levels and shared many attributes with bisulfite deamination artifacts. Our results thus strongly argue against a DNA methyltransferase activity of Dnmt2 and suggest that Dnmt2-dependent phenotypes are caused by reduced tRNA methylation. FREP3 is a determinant of resistance to schistosome infection in snails (part 2)

Project description:Transcriptional profile of BS-90 snails injected with a cocktail of four FREP3 specific 27-mer DSiRNA oligos and two hours later exposed to S. mansoni miricidia. Compared to BS-90 snails injected with a cocktail of three GFP specific DSiRNA oligos and two hours later exposed to S. mansoni miricidia. Experiments were done over the course of 49 days. Snails were collected (10each) at 2 and 4 dpe to S. mansoni for comparison. Each replicate is comprised of 1 individual snail from the specified treatment group. Ten replicates of each treatment were analyzed on the Snail oligo array.

Towards an understanding of the mechanism of hypoevolution of Schistosoma japonicum schistosomula from Microtus fortis

Project description:More than 40 species of mammal have been reported to be infected naturally with Schistosoma japonicum (Chinese mainland strain) in China. The reed vole, Microtus fortis, is the only known mammalian host in which the schistosomes are unable to mature and cause significant pathogenic changes. Gene expression profiling of the 10 day old schistosomula was performed. Microarray analysis was also used to identify differences in gene expression between Schistosoma japonicum schistosomula from BALB/c mice and from Microtus fortis. 10 day old schistosomula were isolated, total RNA obtained and Agilent one colour labeling used. A custom designed Agilent microarray was used to determine what differential gene expression occurs between parasites maintained in either a permissive (mouse) or non-permissive (vole) hosts.
