Heterotypic signaling between dermal fibroblasts and melanoma cells induces phenotypic plasticity and proteome rearrangement in malignant cells

Project description:The signaling events triggered by soluble mediators released from both transformed and stromal cells shape the phenotype of tumoral cells and have significant implications in cancer development and progression. In this study we performed an in vitro heterotypic signaling assay by evaluating the proteome diversity of human dermal fibroblasts after stimulation with the conditioned media obtained from malignant melanoma cells. In addition, we also evaluated the changes in the proteome of melanoma cells after stimulation with their own conditioned media as well as with the conditioned medium from melanoma-stimulated fibroblasts. Our results pointed out to a significant rearrangement in the proteome of stromal and malignant cells upon crosstalk of soluble mediators. The main proteome signature of stimulated cells was related to protein synthesis, which may indicate that this process might be an early response of stimulated stromal cells. In addition, the conditioned medium derived from ‘primed’ stromal cells (melanoma-stimulated fibroblasts) was more effective in altering the functional phenotype (cell migration) of malignant cells than the fibroblast conditioned medium alone. Collectively, self- and cross-stimulation may play a key role in shaping the tumor microenvironment and, more importantly, enable tumoral cells to succeed in the process of melanoma progression and metastasis. Although the proteome landscape of cells participating in such a heterotypic signaling represents a snapshot of a highly dynamic state, understanding the diversity of proteins and enriched biological pathways resulting from stimulated cell states may allow for targeting specific cell regulatory motifs involved in melanoma progression and metastasis.

DNA methylation variations are required for reversible EMT induced by cancer-associated fibroblasts in PCa cells

Project description:Epithelial-to-mesenchymal transition (EMT) and cancer stem cells play relevant roles in metastasis and drug resistance in castration-resistant PCa. Conditioned-media from Cancer-Associated Fibroblasts from two patients with aggressive PCa induce EMT, reversible DNA methylation and transcriptional variations in androgen independent PC3, but not in androgen dependent LN-CaP cells. Focal CpG islands hyper-methylation associated to transcriptional repression of epithelial markers occurs together with widespread hypo-methylation, including promoters of EMT and stemness regulating genes resulting in their transcriptional activation. Remarkably, DNA methylation and transcription patterns are entirely reverted upon exposure to serum-free medium (mesenchymal-to-epithelial transition). DNMT3A is required for de novo methylation and silencing of CDH1 and GRHL2, the ZEB1 direct repressor, while its knock-down prevents EMT entry. These unprecedented results highlight that CAF-released factors induce reversible DNA methylation patterns required for transcriptional variations essential for EMT and stemness in androgen independent PCa cells, suggesting that similar plasticity might occur in tumour microenvironment.

Project description:Paracrine signals relayed between heterogenous cell types through small and large extracellular vesicles promote cancer cell growth, invasion and metastasis. Microplasts, also called as cytoplasts or cytokineplasts are large extracellular vesicles (0.5-11µm diameter) originating from migratory cells. Microplasts are known to be associated with invasive phenotypes of cancer cells and promote metastasis. Treatment with macrophage conditioned medium induces significant shedding of microplasts from MCF-7 breast adenocarcinoma cells. To characterise microplasts and to study their potential role in intercellular communication in cancer, we isolated microplasts derived from macrophage conditioned medium treated MCF-7 cells and investigated their protein cargo through LC-MSMS.

Microarray Analysis of Genes Regulated by Macrophage-Conditioned Media Treatment of Human Pre-Adipocytes

Project description:A major development in the study of obesity is the recognition that the condition is characterised by chronic mild inflammation. Within adipose tissue, this involves the infiltration of macrophages, as well as the direct inflammatory response of the adipocytes and pre-adipocytes. This study has used Agilent whole-genome microarrays to examine the effects of macrophage-conditioned medium on the global inflammatory response of human pre-adipocytes. Human pre-adipocytes (SGBS cells) were treated with macrophage (U937 cells) conditioned medium for 24 h. Control pre-adipocytes were treated with unconditioned medium (control) or RPMI-1640 media alone to control for differences in media used to culture the U937 cells. There were 5 biological replicates per group.
