Identification of transcripts bound by an RNA binding protein (RBP) in early erythroid progenitor using RNA-binding protein immunoprecipitation-microarray (RIP-Chip) experiment

ABSTRACT: Early erythroid progenitors were isolated from mouse E14.5 fetal liver. After cell lysing, control IgG or RBP specific antibody were incubated with cell lysis. Immunoprecipitation followed by microarray experiments were carried out to identify transcripts that are immunoprecipitated by either control IgG or RBP specific antibody. Examination of transcripts that are immunoprecipitated by either control IgG or RBP specific antibody in primary mouse early erythroid progenitor

Stem cells and progenitors in many lineages undergo self-renewing divisions, but the extracellular and intracellular proteins that regulate this process are largely unknown. Glucocorticoids stimulate red blood cell formation by promoting self-renewal of early burst-forming unit-erythroid (BFU-E) progenitors. Here we show that the RNA-binding protein ZFP36L2 is a transcriptional target of the glucocorticoid receptor (GR) in BFU-Es and is required for BFU-E self-renewal. ZFP36L2 is normally downre  ...[more]
