Project description:Arsenic is a carcinogenic metalloid element found naturally in the environment; however human activities such as mining and pesticide use have led to increased concentrations in some areas. Silurana tropicalis eggs were exposed to sublethal concentrations of sodium arsenate (0, 0.5 and 1 ppm) for 3 days during embryogenesis. Four pooled of 10 larvae were sampled per treatment and analyzed for gene expression changes using a microarray approach. Silurana tropicalis eggs were exposed to sublethal concentrations of sodium arsenate (0, 0.5 and 1 ppm) for 3 days during embryogenesis. There were 5 controls, 5 low dose and 6 high dose microarrays perfomed.

Project description:An ex vivo protocol was optimized to expose testis tissues from juvenile Silurana tropicalis to 5 uM finasteride. Testis tissues were incubated in L-15 media with (n = 8) and without (n = 8) finasteride for 6 h at 26 °C on top of an orbital shaker. Afterwards, the tissues were flash frozen on dry ice and RNA was isolated using Trizol. The isolated RNA of four control tissue samples and four finasteride exposed tissue samples were then analyzed using a custom microarray for S. tropicalis containing 44,000 probes . Testis tissues were incubated in L-15 media with (n = 8) and without (n = 8) finasteride for 6 h at 26 °C

Project description:Dyes used in fabric and leather industry are being released and accumulated into Canadian ecosystems. Recent studies have demonstrated that dyes made of azo compounds significantly increase toxicity in biota, which is explained by their toxic metabolites (e.g., aromatic amines). The metabolites of azo compounds interact with hydrophobic surfaces of cell membranes causing expansion of the membrane which impede normal cellular functions. It has been suggested that this process leads to cell death due to improper ion balance. Currently, it is estimated that between 10 and 15% of azo dyes are released in the environment as effluent. The aim of this study was to evaluate toxicity and gene networks altered by azo compounds in amphibians using ecotoxicogenomic approaches. Larvae of the frog Silurana tropicalis (Western clawed frog) were exposed to sediment contaminated to 887 ppm Disperse Yellow 7 (DY7). Larvae were exposed from Nieuwkoop-Faber developmental stage 12 to 46. Data suggest that the azo dye DY7 induced cellular stress and interfered with androgen biosynthesis in early tadpole development. At exposure completion, RNA was isolated from whole larvae and quality was ascertained using bioanalyzer analysis. A custom Agilent 4 X 44 K microarray for S. tropicalis was used to characterize gene regulatory networks underlying toxicity. This study presents the transcriptional regulatory pathways affected by DY7 in S. tropicalis early development. Embryos were exposed to the DY7 dye during development. There were 4 control groups exposed and 4 treatment groups exposed.

Project description:The objectives of the study were to determine the effects of progesterone in the male testis. Reproductive fathead minnows were used in the study. Testis at stage 2 and 3 were dissected and testis explants were treated progesterone or no progesterone (10^-6M and 10^-8M) for 12 hours. Androgens were measured and there was a significant increase in testosterone, but not 11-tetotetosterone. Project description:The herbicide linuron is an endocrine disruptor with a suspected anti-androgenic mode of action (MOA) but the complete MOA for LIN is not fully characterized. The objectives of this study were to better characterize the MOA of LIN in the fathead minnow (FHMs) ovary by comparing expression profiles of LIN to dihydrotestosterone (DHT) and flutamide (FLUT), both model compounds with well defined androgenic and anti-androgenic MOAs respectively. Ovarian explants from vitellogenic FHMs were exposed to 10-6 M, 10-7 M, and 10-8 M of DHT, FLUT, and LIN in vitro in a12 hour incubation experiment. Ovary explants exposed to DHT showed a significant increase in E2 production compared to controls but FLUT and LIN did not affect E2 production. Microarray analysis and support vector machine classification revealed that expression patterns of FLUT and LIN in the ovary were more similar to each other compared to DHT and other androgens. Gene set enrichment analysis identified the notch signaling cascade was affected by all three chemicals. DHT down-regulated the WNT-Frizzled pathway while LIN down-regulated angiopoietin receptor signaling and increased biosynthesis of cholesterol. LIN shared 27 expression sub-networks (e.g. beta-3 adrenergic receptor, MAP3K1, interleukin, signlaing) in common with FLUT, and only 4 sub-networks with DHT. A reciprocal gene expression network was constructed using DHT and FLUT data, and the network revealed that steroid metabolism, translation, and DNA replication are potentially regulated through AR signaling. Project description:Male Sprague-Dawley (SD) rats (350 - 400 g) were anaesthetized with 3% isoflurane for 3 min in a closed induction chamber. A helium driven shock tube was used at various shock wave pressures (20 - 30 psi). The exposure system has been developed so that the animals are exposed head only to a ?primary blast? that being a single pulse shock wave with only minimal concussive and whiplash effects. After exposure, rats were closely observed for either 1 day or 7 days before being sacrificed. At the end of the observation period, rat whiskers were quickly pulled and placed in a 1.5 ml micro centrifuge tube containing 0.5 ml of RNAlater. Samples were then stored at -80C until further analysis.

Project description:Perfluorooctanesulfonate (PFOS) has been widely used in a variety of industrial and commercial applications as a surfactant and stain repellent. PFOS causes liver damage (including liver tumors) in experimental animals, primarily via interaction with PPARa and CAR/PXR. We investigated the involvement of microRNAs (miRNAs) in PFOS-induced hepatotoxicity, and mechanisms involved in abnormal TH homeostasis, in the livers of adult male rats exposed in feed to 50 mg PFOS/kg diet for 28 days. PFOS-treated rats exhibited expected histopathological and clinical chemistry changes. Global gene expression changes were consistent with the involvement of PPARα and CAR/PXR in PFOS-induced effects. Thirty-eight miRNAs were significantly altered. Three members of the miR-200 family were the most increased, while miR-122 and miR-21 were the most decreased, in PFOS-treated relative to control rats. Expression of the miR-23b/27b/24 cluster also decreased in PFOS-treated animals. Pathway analysis of miRNAs and associated gene expression changes demonstrated enrichment of transcripts involved in epithelial to mesenchymal transition (EMT), which is a primary process involved in tumor cell motility and cancer metastasis. Liver expression analysis revealed transcripts that may mediate PFOS effects on thyroid hormone (TH) homeostasis including: activation of the CAR/PXR pathway, phase II/III enzymes, and deiodinase. These changes are consistent with low serum TH due to enhanced metabolic clearance of TH. However, most TH hepatic target genes were not altered in a manner consistent with reduced TH signalling; suggesting that PFOS exposure did not induce functional hypothyroidism. Collectively, PFOS-induced miRNA perturbations were strongly associated with EMT suggesting an important role for miRNAs in PFOS-induced hepatotoxicity. The work also provides novel insights into the effects of PFOS on TH homeostasis. Four RNA samples from control and four from PFOS treated rats were labelled with Cyanine 5-CTP (Cy5) using Low Input Quick Amp Labelling kits (Agilent Technologies Inc.) following the manufacturer’s instruction. Universal rat reference total RNA (Agilent Technologies Inc.) was labelled with Cyanine 3-CTP (Cy3). Cy5-sample cRNA and Cy3-reference cRNA were hybridized to Agilent G4853A SurePrint G3 Rat GE 8 X 60K microarrays (Agilent Technologies Inc.) at 65°C overnight with Agilent hybridization solution. Slides were washed and scanned on an Agilent G2505B microarray scanner at 5 μm resolution, and the data were acquired with Agilent Feature Extraction software version 10.7.3.1. Microarray data quality was confirmed using Agilent Feature Extraction quality control metrics and in-house metrics (boxplots, cluster analyses, and MA plots to identify poor quality outlier arrays). All samples passed the quality control tests and were used for subsequent analyses.

Project description:Furan is a widely used industrial chemical and a common contaminant in heated foods. Chronic furan exposure has been shown to cause cholangiocarcinoma and hepatocellular tumors at doses of 2 mg/kg bw/day with gender differences in frequency and severity. The hepatic transcriptional alterations induced by low doses of furan (doses below those inducing liver tumors) and the potential mechanisms underlying gender differences are largely unexplored. We used DNA microarrays to examine the global hepatic mRNA and microRNA transcriptional profiles of male and female rats exposed to 0, 0.03, 0.12, 0.5 or 2 mg/kg bw/day furan over 90 days. Marked gender differences in gene expression responses to furan were observed, with many more altered genes in exposed males than females, confirming the increased sensitivity of males even at the low doses. Pathway analysis supported that key events in furan-induced hepatotoxicity in males included gene expression changes related to oxidative stress, apoptosis and inflammatory response, while pathway changes in females were consistent with primarily adaptive responses (regeneration). Pathway benchmark doses (BMDs) were estimated and compared to relevant apical endpoints. Transcriptional BMDs could be examined in males, they ranged from 0.08 M-bM-^@M-^S 1.43 mg/kg bw/day and approximated those derived from traditional histopathology. MiR-34a (a target of P53 signalling) was the only microRNA significantly increased at the 2 mg/kg bw/day, providing evidence in support of the importance of apoptosis and cell proliferation in furan hepatotoxicity. Overall, this study demonstrates the use of transcriptional profiling to discern mode of action and mechanisms involved in gender differences. A total of 50 RNA samples (5 female and 5 male rats per dose group, 5 dose groups total) were labelled with Cyanine 5-CTP using Low Input Quick Amp Labelling kits (Agilent Technologies Inc.) following the manufacturerM-bM-^@M-^Ys instruction.
