Plant Leaves: Pinellia pedatisecta vs Pinellia ternata

ABSTRACT: miRNA expression profiling of Pinellia pedatisecta leaves comparing Pinellia ternata leaves in the same condition. Then we compared the accumulation of known miRNA families using microarray signals to identify further the differential miRNA expression between Pinellia pedatisecta and Pinellia ternata. Pinellia pedatisecta vs Pinellia ternata, independently grown and harvested. Each sample are mixed. One replicate per array.

Project description:In this study, we utilized the microfluidics chip technology on the gonads of Amur sturgeon to identifiy gender-specific miRNAs. The probes of all miRNAs about 663 published in fish and our novel miRNAs from sturgeon were chosed in the microarray experiment. Above 4Âµg to 8Âµg total RNA from one female and one male separativley were performed the miRNA microarray assays.Each probe was repeated five times on the chip to ensure reproducibility of microarray.

Paired subcloned human large cell lung carcinoma cell lines: low-metastatic 95C vs high-metastatic 95D

Project description:The paired low-metastatic 95C and high-metastatic 95D cell lines were subcloned from a low differentiated human large cell lung carcinomacell line PLA-801. The cells were well authenticated and published by several research groups. The cells were kindly provided by Professor Ying-Lin Lu, Department of Pathobiology, Institute of Basic Medical Sciences, Academy of Military Medical Sciences, Beijing, China on Dec. 5M-oM-<M-^L2009. As described The 95C and 95D cells were cultured in RPMI 1640 (Invitrogen, USA) with 100 units/mL penicillin, 100 M-NM-<g/mL streptomycin and 10% calf bovine serum, and grown at 37M-BM-0 C in atmosphere with 5% CO2. miRNAs differential expression between 95C and 95D was measured using a miR human_01_H10.1_080277 miRNA array (LC Sciences Houston, USA). Paired subcloned human large cell lung carcinomacell line PLA-801 of low-metastatic 95C and high-metastatic 95D cell lines. Biological replicates:1. Five replicate per array.

Microarray analysis of miRNAs involved in Porcine Alveolar Macrophages at the early stage of porcine reproductive and respiratory syndrome virus infection [LCS_miRPig_17]

Project description:By analyzing the changes of miRNAs of piglet PAMs 6h after PRRSV infection,we can find porcine miRNAs associated with viral infection and possible target sites in both PRRSV and pig genome and their roles in protection against viral infections. Then these targets will screened by bioinformatics methods to find the susceptibility genes, resistance genes and immune related genes, and formed molecular biology network of miRNAs at gene and protein level . PAM cells were isolated from 60d old healthy piglets and inoculated with HuN4,a strain of highly pathogenic porcine reproductive and respitory syndrome virus (HP-PRRSV) and were collected at 6h post-inoculation (pi). The uninfected cells served as mock-infected cells.The miRNA chip was according to the porcine miRNA sequences in Sanger miRBase Release 17.0 (http://www.sanger.ac.uk/Software/Rfam/mirna/)added with some miRNAs we found in reference. Microarray hybridization and data analysis helps to find the susceptibility genes, resistance genes and immune related genes, and formed molecular biology network of miRNAs at gene and protein level .

miRNA profiling of gastric epithelium from Pten mutant mouse

Project description:Phosphatase and tensin homologue deleted on chromosome 10 (PTEN) is a tumor suppressor that negatively regulates cell survival and proliferation by antagonizing phosphatidylinositol 3-kinase(PI3K)/protein kinase B(PKB/Akt) signaling. Loss of heterozygosity (LOH) of PTEN, reduced expression of PTEN and overexpression of phosphorylated Akt are frequently found in human gastric cancer, and their changes correlate with tumor progression and prognosis. Previous studies have shown that the deregulated miRNAs in human gastric cancer play important roles in gastric cancer cell proliferation, apoptosis and inflammation. However, miRNAs downstream PTEN/Akt signaling is poorly investigated. To clarify whether PTEN is involved in gastric tumorigenesis, we have generated a gastric epithelium specific PTEN knockout mouse which exhibited gastric tumor formation with enhanced cell proliferation.So the objectives of the microarray experiment were to, a) screen miRNAs which might be regulated by PTEN/Akt signaling by comparing the miRNA expression profiles between PTEN deficient and control gastric epithelia. 2) explore the microRNA mechanism involved in gastric cell proliferation and gastric tumorigenesis. miRNA profiling of mouse gastric epithelium,comparing Pten mutant mouse with controls. 4 samples. Experiments in 2 different time point, 20 days and 60 days after birth, 2 Biological replicates. Mutant tissue vs. controls from mixture of 3-4 mouse.

Distinctive microRNA profiles and target genes of aberrant microRNAs in leukemia chicken infected by subgroup J avian leukosis virus

Project description:Subgroup J avian leukemia is a type of oncology infectious disease caused by Subtype J of avian leukosis virus (ALV-J). It mainly encroaches on bone marrow cells, and metastasizes to liver, kidney, splenic ellipsoids and other organs, leading to myeloid leukosis (ML) and other malignancies, resulting in significant economic losses. microRNA play important roles in oncology infectious diseases. We used miRNA microarray analysis to detail the relationship of aberrant microRNAs and chicken ALV-J leukemia, and to try to find the potential diagnostic and therapeutic target for infections of subtype J of leukemia. ALV-J-infected and non-infected ten-week-old chicken liver tissues were sampled for the array assay. A1, A2, and A3 are the ALV-J infection group samples, and DA1, DA2, and DA3 are the control samples. 623 mature miRNA sequences were assembled and integrated into the LC miRNA microarray design, and different miRNA expressions were measured on the 7000HT Fast Real-Time PCR system.

Recipe for a busy bee: MicroRNAs in honey bee caste determination (microRNA array)

Project description:Social caste determination in the honey bee is assumed to be determined by the dietary status of the young larvae and translated into physiological and epigenetic changes through nutrient-sensing pathways. We have employed microRNA gene-microarray, and observed that both worker jelly and royal jelly showed dynamic changes in miRNA content during the 4th to 6th day of larval development . Adding specific miRNAs to royal jelly elicited significant changes in queen larval mRNA expression and in morphological characters of the emerging adult queen bee. We propose that miRNAs in the nurse bee secretions constitute an additional element in the regulatory control of caste determination in the honey bee. We collected worker and royal jelly of the Italian honeybee (ZND No.1, Apis mellifera ligustica) at 73~90 hours (4th-day larvae), 97~114 hours (5th-day larvae), and 121~138 hours (6th-day larvae) after hatching. After total RNA was extractedM-BM- and quantified , equal amounts of total RNAs from each of the three sampling days were analyzed on the LC Science miRNA-array to observe the expression variation of miRNAs between worker jelly and royal jelly along with the development time points (4th-day, 5th-day and 6th-day).

Microarray identification of conserved microRNAs in Pinellia pedatisecta

Project description:An in situ synthesized custom miRNA microarray was employed to detect miRNAs from P. pedatisecta, A total of 99 miRNAs belonging to 22 miRNA families were identified. in the study present here, the microarray design was based on 2521 well-characterized miRNAs from 43 plant species on Sanger miRBase Release 16, and Eighteen 5S rRNAs that served as internal reference were chosen from A. thaliana, C. reticulata, and O. sativa with an equal amount. Eight quality control probes were synthesized to perfect match (PM) or one nucleotide mismatch (MM) with eight external spiked-in synthetic RNA (20mer).
