ABSTRACT: The NCI-60 cancer cell lines have been used for studying cancer pathogenesis and for drug discovery. This dataset adds to the growing number of datasets on various genomic platforms that have been made public on Cellminer (http://discover.nci.nih.gov/cellminer/) 60 cell lines (including PR:DU145 and its drug resistant derivative PR:RC01) were arrayed. LC:A549 was done with 8 replicates. LC:NCI_H226 was done on one array. All others were done with 2 replicates. All cell lines with 2 or more replicates were done with dye flips

Project description:The NCI-60 cancer cell lines have been used for studying cancer pathogenesis and for drug discovery. This dataset adds to the growing number of datasets on various genomic platforms that have been made public on Cellminer (http://discover.nci.nih.gov/cellminer/) 61 cell lines (including PR:DU145 and its drug resistant derivative PR:RC01) were arrayed. Of these, 56 were done one time, four cell lines were done with either two or three replicates, and one cell line (LC:A549) was done with three replicates and two dye flips. Two color arrays were used, with Cy3 mostly used for sample and Cy5 for the reference pool of 6 normal male samples.

Senescence-correlated gene expression programs of human prostate stromal cells in response to multiple proliferation-arresting stress and their potential in driving prostate neoplasia

Project description:As an age-related process, cellular senescence is cell cycle arrest and it safeguards against incidental oncogenic activation by playing antiproliferative roles. Despite the establishment that senescence is a beneficial anticancer mechanism, increasing evidence indicates its deleterious effects that might contribute to age-related pathologies. We applied a group of methods that causes typical senescence to PSC27, a primary normal human prostate stromal cell line, and compared their capacity in influencing the expression of senescence- and proliferation-related genes. Custom Agilent 44K whole human genome expression oligonucleotide microarrays were used to profile PSC27 prostate fibroblast cells treated with agents to induce senescence (oxidative stress, DNA damage, and radiation.) Total RNA was isolated and amplified prior to hybridization against a common reference pool of prostate tumor cell lines.

A Characterization of Osteoblastic and Osteolytic Proteins in Prostate Cancer Bone Metastases

Project description:Our goal was to determine whether previously identified and novel proteins are associated with the osteoblastic or osteolytic response in clinical specimens of PCa bone metastases. Custom Agilent 44K whole human genome expression oligonucleotide microarrays were used to profile 14 PCa metastases from 11 patients with highly osteoblastic and highly osteolytic bone reactions. Total RNA was isolated and amplified prior to hybridization against a common reference pool of prostate tumor cell lines.
