Project description:Peg3 (paternally expressed gene 3) is an imprinted gene encoding a DNA-binding protein. This gene plays important roles in controlling fetal growth rates and nurturing behaviors. In the current study, a new mutant mouse model has been generated to further characterize the functions of this DNA-binding protein. Besides known phenotypes, this new mutant model also revealed potential roles of Peg3 in mammalian reproduction. Female heterozygotes produce a much smaller number of mature oocytes than the wild-type littermates, resulting in reduced litter sizes. According to genome-wide expression analyses, several placenta-specific gene families are de-repressed in the brain of Peg3 heterozygous embryos, including prolactin, cathepsin and carcinoembryonic antigen cell adhesion molecule (Ceacam) families. The observed de-repression is more pronounced in females than in males. The de-repression of several members of these gene families is observed even in the adult brain, suggesting potential defects in epigenetic setting of the placenta-specific gene families in the Peg3 mutants. Overall, these results indicate that Peg3 likely controls the transcription of several placenta-specific gene families, and further suggest that this predicted transcriptional control by Peg3 might be mediated through unknown epigenetic mechanisms. Mouse Breeding Two different types of heterozygotes were prepared for breeding experiment: male and female heterozygotes carrying the paternally (+/-p) and maternally (-m/+) transmitted targeted allele. These 4 heterozygotes were bred with their littermates: for Breeding 1, female wild-type littermates x male heterozygotes (+/-P); Breeding 2, female heterozygotes (+/-P) x male wild-type littermates; Breeding 3, female wild-type littermates x male heterozygotes (-m/+); for Breeding 4, female heterozygotes (-m/+) x male wild-type littermates. Genome-wide expression analysis A litter of 14.5-dpc fetuses were harvested from a timed mating between a male heterozygote (-m/+) and wild-type female littermate. Three tissues were obtained from each fetus: embryo head, placenta and amnionic sac. Total 16 samples were used for this series of genome-wide expression analyses: 8 embryo heads and placenta representing two biological replicates of each of the four following combinations: male wild-type, male heterozygote, female wild-type, and female heterozygote.

Project description:Here, we performed the first iTRAQ-based proteomic analysis of early embryonic longissimus dorsi muscle from Landrance (LR) and miniature pig (Wuzhishan WZS) ranging from 21 to 42 days post coitus (dpc). 4431 proteins were identified. In both WZS and LR, the largest amount of differentially expressed proteins (DEPs) were found between 28 and 35 dpc. 252 breed-DEPs were selected by GO analysis, including 8 myofibrillar proteins. Only MYHCⅠ/IIA mRNA were detected due to early embryonic stages, and significantly higher expression of them were found in WZS during these 4 stages. MYHCⅠ was first found in WZS at 28 dpc and expressed in both breeds at later stages, while MYHCII protein was not detected until 35 dpc in both breeds. Thus, 33 myogenic breed-DEPs selected from last two stages were analyzed by STRING, which showed that some myofibrillar proteins (MYH1, TPM4, MYH10, et.al) and functional proteins (CSRP2, CASQ2, OTC, et.al), together with candidate myogenic proteins (H3F3A, HDGFRP2, et.al), probably participate in the regulatory network of myofiber formation. Thus, our study provides new data on myofiber type development during early embryo stages, which contributes to the improved understanding in the regulation mechanism of early myogenesis in agricultural animals.

Project description:Despite the fact that we are constantly exposed to various environmental compounds, very few studies explore the impact of combined exposure to physical and chemical pollutants on reproductive health. Until now, assessment of pollutants is mostly based on the evaluation of single pollutant or combination of chemicals with common features and modes of action. In this context, numerous studies have demonstrated that steroidogenesis and gametogenesis, the main testicular functions, are well-known to be sensitize by endocrine disruptors (as Bisphenol A) and DNA-damaging agents (as γ-rays) respectively. In this study, we aim to investigate short and long term testicular transcriptionnal alterations of combined fetal exposure to well-known environmental toxicants: γ-rays (RAD) and BPA. To discriminate specific signatures of BPA or RAD exposure after combined exposure and evidence the type of synergisms between this pollutants, we performed transcriptomic analyses on testis exposed to BPA or RAD alone and co-exposed with BPA and RAD and compared gene expression with control condition. For this, we exposed pregnant mice from 10.5 dpc to 18.5 dpc to 10µM of BPA (in drinking water) and/or we irradiated mice at 12.5 dpc to 0.2 Gy. We performed transcriptomic analyses on fetal testis (18.5 dpc) and adult testis (3 months) to evaluate short and long term cell response after in utero exposure.

Of the 23,000 genes that were analyzed on the array, B. ovis infection in rams’ tissues revealed 139, 930 and 744 Differentially Expressed Genes (DEGs) in the acute, chronic 1, and chronic phase 2 of infection, respectively. Among the three phases of infection, 44 DEGs (30 known and 14 unknown genes) were expressed in common. The biological functions of immune cell trafficking, immunological disease, infectious disease, inflammatory disease, inflammatory response and cellular movement were significant at the three phases of infection. The results support the first microarray analysis of ram tissues infected with an R strain of B. ovis. A total of twelve 12-month-old healthy Santa Inês rams, previously diagnosed as negative for brucellosis/visna maedi/toxoplasmosis/neosporosis, were selected for the study. The rams were kept in an isolated pen and experimentally challenged with a rough (R) virulent B. ovis PA strain (ATCC 25840). For the experiment, the rams were classified during the course of infection according to time: 0 dpc (control), 60 dpc (acute phase), 120 dpc (chronic phase 1) and 240 dpc (chronic phase 2). For each time, the tissues epididymus, testicles, ampolae, vesicular glands, bulbourethral glands, and a pool of lymph nodes (inguinal and scrotal) were collected. Three rams were euthanized and necropsied for tissues collection purposes for each time. To define the gene expression changes associated with infected rams, the profile of a control group (0 dpc) of rams was compared using microarray technology.
