ABSTRACT: Meig1-deficient mice show male germ cell development defect after they are 28 days old. To understand if the phenotype was caused by altered gene expression due to MEIG1 deficiency, total testicular RNA was isolated from 22 day old and 28 day old wild type and Meig1-deficient mice, microarray was conducted and gene expression was compared between wld-type and Meig1-deficient mice at the two time points. Total testicular RNA was extracted from 22 day or 28 day old wild-type and Meig1-deficient mice, and gene expression was compared between wild-type and Meig1-deficient mice at the two time points.

A molecular and phenotypic integrative approach to identify a no effect dose level for anti-androgen induced testicular toxicity

Project description:With the advent of high information content technologies, especially microarrays, it is pertinent to determine the impact of molecular data on the NOAELs. Consequently, we conducted an integrative study to identify a no transcriptomic effect dose using microarray analyses coupled with qPCR and determined how this correlated with the NOAEL. We assessed the testicular effects of the antiandrogen, flutamide (FM), in a rat 28-day toxicity study using doses of 0.2-30 mg/kg/day. Concerning molecular data, we observed differential gene expression starting from 1 mg/kg/day and a deregulation of more than 1500 genes at 30 mg/kg/day. Dose-related changes were identified for the major pathways associated with the testicular lesion (eg fatty acid metabolism), that were confirmed by qPCR. These data, along with standard measurements supported the no effect dose of 0.2 mg/kg/day. Flutamide was administered in suspension to rats (7 weeks old at start of treatment, 10 per group) by oral gavage at a daily dose of 0 (control), 0.2, 1, 6 and 30 mg/kg body weight, for 28 consecutive days. Dose-related changes in gene expression were determined in the testes using whole genome oligonucleotide microarrays.

Transcriptional Metabolic Reprograming Drives Meiotic Fate Decision in Mammalian Germ Cells

Project description:Nitrogen starvation drives meiosis in yeasts1, while retinoic acid (RA) is required for mammalian meiosis through the activation of its germline-specific target, stimulated by retinoic acid gene 8 (Stra8).Here, by using single-cell transcriptome analysis of wild-type and Stra8-deficient testes, our data revealed that the expression of major metabolic genes are downregulated during germ cell entry into meiosis. Recently, we collected mouse testicular cells from wild-type and Stra8-deficient mice at postnatal day 12 (P12) and P21 for single cell RNA-sequencing (scRNA-seq). Upon scRNA-seq, we obtained a total of 23,470 cells from these samples. Using stringent quality control, 22,839 cells were selected for further analysis. All cells, including both wild-type and Stra8-deficient testicular cells, were pooled together to perform clustering analysis, which revealed 6 major cell clusters based on their distinct gene expression patterns. We annotated these six clusters by using known marker genes, including spermatogonia (SPG; Stra8; 3,941 cells), spermatocyte (SPC; Syce1; 4,924 cells), spermatid (STD; Acrv1; 710 cells), Sertoli cell (SC; Sox9; 9,488 cells), Myeloid (MYD; Acta2; 3,531 cells), and Leydig Cells (LY; Star; 245 cells). There is an increase in the percentage of spermatogonia in Stra8-deficient testes at both P12 (28% versus 14% in wild-type) and P21 (17% versus 10% in wild-type). At P21, while spermatocytes are abundant and spermatid are formed in wild-type testes, there are only a few spermatocytes (0.2% versus 73% in wild-type), and spermatids are absent in Stra8-deficient testes. Signature genes are enriched for GO terms for distinct biological function and metabolic pathways in different cell types.

Genome-wide analysis of E2F1 dependent gene expression in mouse testicular tissue

Project description:The effect of E2F1 loss on gene expression in testicular tissue was analyzed in 20-day old E2F1-/-, E2F1+/- mice. The hypothesis ws that there would be clear changes in expression of genes related to cell cycle machinery. The results show that E2F1 has wider implications in gene expression dynamics in testicular cell types than merely controlling cell cycle related genes. Total RNA obtained from testis tissue of 20-day-old E2F1-/- (KO), E2F1+/- (HZ) and E2F1+/+ mice (WT).

Understanding the underlying molecular mechanisms of meiotic arrest in organotypic culture of prepubertal rat testicular tissues

Project description:We report the use of the Bulk RNA-sequencing technology on in vivo and in vitro cultured prepubertal testicular tissues. The goal of this study is to identify the origin of the maturation arrest observed in rat prepubertal testicular tissue cultured with the organ culture system with a transcriptomic approach. We compared the gene expression of 14.5 dpp rat testicular tissues cultured up to 6, 16, 22 and 28 days. The statistical filtration revealed 600 differentially expressed genes showing significant differential abundances in all three comparisons. This study uncovered for the first time several mechanisms that could explain the meiotic arrest observed in cultures of rat prepubertal testicular tissues.
